In Situ Isolation and Culturing of Recalcitrant Soil Bacteria using an Isolation Chip (iChip).

Ecosystems harbor millions of environmental microbes, some of them capable of producing biotechnologically relevant products such as enzymes, vitamins, amino acids, organic acids, anti-parasitic agents, and antimicrobial agents. Most of these environmental bacteria are deemed "unculturable" as the laboratory culturing methods fail to meet their nutritional/environmental requirement. Moreover, as they coexist in nature, they may also be dependent on a nutrient/metabolite produced by another member of the microbial community. Bringing the "uncultured" microbial diversity into the culture will pr

Comparison of three techniques to detect Leptospira interrogans serovar hardjo type hardjo-bovis in bovine urine.

Nucleic acid hybridization, bacteriologic culture, and a fluorescent antibody test were compared for detection of Leptospira interrogans serovar hardjo type hardjo-bovis in bovine urine. Seventy-five urine samples were collected from pregnant cows challenge exposed with type hardjo-bovis. Twenty samples were collected from steers not exposed to hardjo-bovis. Sediments from each sample were examined, using fluorescent antibodies and a repetitive sequence element nucleic acid probe, to detect the presence of leptospires. Urine samples were processed for bacteriologic culture, using standard tech

Collection and evaluation of tracheobronchial washes in the horse.

A flexible endoscope was used to obtain 223 tracheal washes from 191 horses in three clinical categories. Total cell counts, cytological and bacteriological examinations are reported and the features of the main cell types encountered described. The presence and degree of inflammatory airway disease was determined by a semiquantitative assessment of the neutrophil response and was an important consideration in the interpretation of the bacteriological results. Potential pathogens were isolated from approximately 30 per cent of samples. Cytological changes suggestive of lungworm infestation, vi

Evidence of Brucella strain ST27 in bottlenose dolphin (Tursiops truncatus) in Europe.

Marine mammal brucellosis has been known for more than 20 years, but recent work suggests it is more widespread than originally thought. Brucella (B.) pinnipedialis has been isolated from pinnipeds, while B. ceti strains have been associated with cetaceans. Here we report a Brucella strain isolated from multiple lymph nodes of one bottlenose dolphin (Tursiops truncatus) during routine examination of dolphin carcasses found in the Croatian part of the northern Adriatic Sea during the summer of 2015. Classical bacteriological biotyping, PCR-based techniques (single, multiplex, PCR-RFLP) and 16S

Investigating the effect of microtube capping/decapping methods on Escherichia coli collection contamination: Introducing a novel automatic microtube capper/decapper.

This study aimed to evaluate the effect of varying sample volumes (200 μL, 500 μL, and 1000 μL) on the leakage rates of samples between spin-down and non-spin-down conditions for the target genes Tet and uid-A, comparing machine-operated and manual protocols. The study was designed to assess the differences between mechanical versus manual operators handling different-volume specimens. JM109 Escherichia coli suspensions, carrying the plasmid pBR322 and in the logarithmic growth phase, were aliquoted into 1.5-mL microcentrifuge tubes and divided into six groups based on contamination risk (mini

Performance Evaluation and Validation of Air Samplers To Detect Aerosolized Coxiella burnetii.

Coxiella burnetii, the etiological agent of Q fever, is an intracellular zoonotic pathogen transmitted via the respiratory route. Once released from infected animals, C. burnetii can travel long distances through air before infecting another host. As such, the ability to detect the presence of C. burnetii in air is important. In this study, three air samplers, AirPort MD8, BioSampler, and the Coriolis Micro, were assessed against a set of predetermined criteria in the presence of three different aerosolized C. burnetii concentrations. Two liquid collection media, phosphate-buffered saline (PBS

Listeria monocytogenes: cultivation and laboratory maintenance.

This unit describes general procedures for the lab cultivation and storage of the Gram-positive facultative intracellular bacterium Listeria monocytogenes. The basic protocols are relevant for a wide scope of applications including microbial genetics and both in vitro and in vivo infection studies. Commonly used L. monocytogenes strains, serotypes, and growth parameters are also discussed.

Characterization and comparison of biofilm development by pathogenic and commensal isolates of Histophilus somni.

Histophilus somni (Haemophilus somnus) is an obligate inhabitant of the mucosal surfaces of bovines and sheep and an opportunistic pathogen responsible for respiratory disease, meningoencephalitis, myocarditis, arthritis, and other systemic infections. The identification of an exopolysaccharide produced by H. somni prompted us to evaluate whether the bacterium was capable of forming a biofilm. After growth in polyvinyl chloride wells a biofilm was formed by all strains examined, although most isolates from systemic sites produced more biofilm than commensal isolates from the prepuce. Biofilms

Detection of gas leaks in an anaerobic glove box.

An inert gas, Freon, can be added to the atmosphere of an anaerobic glove box without deleterious effect to cultures of anaerobic microorganisms. The sensitive probe of a Halogen Leak Detector passing over the outside surface of the box will pinpoint any escaping Freon and therefore locate the leak.
