Nucleic Acids as Emerging Regulators of Calcium Phosphate Biomineralization.

Calcium phosphate biomineralization is a highly regulated process that underlies the formation of vertebrate mineralized tissues. While proteins have long been recognized as the principal regulators of mineral formation, accumulating experimental evidence suggests that nucleic acids may also contribute to this process through mechanisms that remain largely unexplored. Their negatively charged phosphate backbone, molecular structural versatility, and ability to interact with calcium phosphate phases make nucleic acids attractive candidates for regulating mineral nucleation, growth, and phase tr…

In Situ Mitochondrial Biomineralization for Drug-Free Cancer Therapy.

The common clinical chemotherapy often brings serious side effects to patients, mainly due to the off-target and leakage of toxic drugs. However, this is fatal for some specific clinical tumors, such as brain tumors and neuroma. This study performs a drug-free approach by encapsulating black phosphorus (BP) and calcium peroxide (CaO2) in liposomes with surface-modified triphenylphosphonium (BCLT) to develop mitochondria targeting calcification for cancer therapy without damaging normal cells. BCLT preferentially accumulates inside tumor mitochondria and then is activated by near-infrared (NIR)

Biomineralization of Dental Tissues Treated with Silver Diamine Fluoride.

Silver diamine fluoride (SDF) is a dental biomaterial used to arrest dental caries. To better understand SDF's mechanism of action, we exami … h. These results suggest that SDF treatment on primary teeth affected by caries promotes pathologic biomineralization by altering their physicochemical properties, occluding dentin tubules, and increasing tertiary dentin volume. These seemingly serendipitous effects collectively contribute to the cariostatic activity of SDF.

Ureolytic Biomineralization Reduces Proteus mirabilis Biofilm Susceptibility to Ciprofloxacin.

Ureolytic biomineralization induced by urease-producing bacteria, particularly Proteus mirabilis, is responsible for the formation of urinary tract calculi and the encrustation of indwelling urinary catheters. Such microbial biofilms are challenging to eradicate and contribute to the persistence of catheter-associated urinary tract infections, but the mechanisms responsible for this recalcitrance remain obscure. In this study, we characterized the susceptibility of wild-type (ure+) and urease-negative (ure-) P. mirabilis biofilms to killing by ciprofloxacin. Ure+ biofilms produced fine biomine…

Nacre initiates biomineralization by human osteoblasts maintained in vitro.

When nacreous shell produced by the marine oyster Pinctada maxima, used as a biomaterial in oral surgery, is implanted in human bone, new bone formation occurs, resulting in a tight welding of the bone to the nacre [16]. These findings are consistent with the possibility that nacre adjacent to bone can locally stimulate osteogenic activity. To test this hypothesis, we have evaluated the effect of the simultaneous presence of bone and nacre on human osteoblasts in vitro. Nacre chips (1 mm3) were placed at approximately 1 mm distance from a similarly sized bone chip on a layer of first passage h

Cationization of Silk Nanofibrils for Antibacterial Applications and Enhanced Biomineralization.

Silk nanofibrils (SNF) demonstrate substantial potential as biomedical materials and biomineralization templates as a result of their biocompatibility, biodegradability, and resemblance to ECM structural proteins. However, a typical drawback is that the domesticated silk nanofibrils lack sufficient biological functional sites such as cationic groups that are beneficial for increasing cell adhesion, inhibiting bacterial growth, and promoting mineralized deposition. In this study, we developed a cationic SNF with dual functionalities of antibacterial properties and enhanced biomineralization usi…

Biomineralization of semiconductor quantum dots using DNA-functionalized protein nanoreactors.

Proteins can template the heterogeneous nucleation and growth of size-confined nanocrystals. However, protein-templated mineralization often leads to particles that exhibit low colloidal stability, poor crystal quality, and/or diminished photoluminescence. Here, we report protein cage-spherical nucleic acids (SNAs) that can be used as nanoreactors for quantum dot (QD) synthesis and subsequent intracellular delivery. The resulting QD-SNA structures are monodisperse, colloidally stable, and photoluminescent in aqueous solution. The nanoreactors were prepared using two different proteins (~10 and

An acidic protein, Hf15, from Haliotis fulgens involved in biomineralization.

Biomineralization leads to the hardening of mineralized materials, such as the shell of Mollusk, to fulfill a wide range of functions, such as (but not limited to) skeletal support, protection of the soft tissues, navigation, etc. The study of the proteins responsible for this process, shell matrix proteins (SMPs), allows addressing questions related to structure-function relationship and to the mechanism of mineral formation, which is limited in gastropod species. In this study, a low molecular weight protein was isolated from the insoluble fraction after decalcification with acetic acid of t…

Interactions between acidic proteins and crystals: stereochemical requirements in biomineralization.

Acidic matrix macromolecules are intimately involved in biological crystal growth. In vitro experiments, in which crystals of calcium dicarboxylate salts were grown in the presence of aspartic acid-rich proteins, revealed a stereochemical property common to all the interacting faces. Calcite crystals are nucleated on stereochemically analogous faces when proteins are adsorbed onto a rigid substrate. The importance of this property in biomineralization is discussed.

Advances in Marine Biomineralized Materials in Bone Repair and Regeneration.

Marine biomineralized materials (MBMs), derived from organisms such as corals, mollusks, cuttlefish, sponges, and fish, represent a promising class of natural resources for bone tissue engineering due to their structural similarity to human bone, excellent biocompatibility, and bioactive properties. The composition, hierarchical architecture, biomineralization mechanisms, and species-specific characteristics of MBMs collectively determine their physicochemical properties and regenerative potential. The porous framework of coral bone, the "brick-and-mortar" structure of nacre, the multi-chamber…

The role of biomineralization in disorders of skeletal development and tooth formation.

The major mineralized tissues are bone and teeth, which share several mechanisms governing their development and mineralization. This crossover includes the hormones that regulate circulating calcium and phosphate concentrations, and the genes that regulate the differentiation and transdifferentiation of cells. In developing endochondral bone and in developing teeth, parathyroid hormone-related protein (PTHrP) acts in chondrocytes to delay terminal differentiation, thereby increasing the pool of precursor cells. Chondrocytes and (in specific circumstances) pre-odontoblasts can also transdiffer

Biomineralized vaccine nanohybrid for needle-free intranasal immunization.

Frequent outbreaks and the rapid global spread of infectious diseases have increased the urgent need for massive vaccination especially in countries with limited resources. Intranasal vaccination facilitates the mass vaccination via needle-free delivery of vaccine through nasal mucosal surfaces. Inspired by the strong capability of calcium phosphate (CaP) materials to adhere to cells and tissues, we propose to improve nasal vaccination by using a biomineralization-based strategy. The vaccine nanohybrid was obtained by covering the viral surface with CaP nanoshell, which changed the physiochemi…

Characterization of the calcium biomineral in the radular teeth of Chiton pelliserpentis.

The radula in a group of molluscan invertebrates, the chitons (Polyplacophora), is a ribbon-like apparatus used for feeding and which bears a series of distinctive mineralized teeth called the major lateral teeth. While some chiton species deposit only iron biominerals in these teeth, many others deposit both iron and calcium. In this study, the calcium biomineral in the teeth of one of the latter types of species, the Australian east-coast chiton, Chiton pelliserpentis, has been isolated and examined for the first time. Spectroscopic and crystallographic techniques have identified the biomine

Mimicking natural biomineralization enabling biodegradable and highly lipophobic alginate hydrogels.

Microorganisms can induce biomineralization of inorganic ions to form a lipophobic layer on the surface of cave rocks. Mimicking this, we developed and propose a protocol that produces a highly lipophobic hybrid layer of CaCO3 nanoparticles on hydrogel surfaces. This lipophobic layer endows hydrogels with an oil contact angle of 162°, causing oil droplets placed on the surface to bead up and roll off immediately. The lipophobic surface effectively resists staining from lipophilic dyes, and does not adhere to double-sided tapes. A lipophobic layer on hydrogel tubes effectively prevents the adhe…

Endogenous iron biomineralization in the mouse spleen of metabolic diseases.

As the most abundant essential trace element in our human bodies, iron plays essential roles in multiple physiological and pathological conditions, including oxygen transport, cancer, diabetes, Alzheimer's and Parkinson's diseases. Although it has been shown that iron can form particles in some bacteria, migrating birds and fish, whether iron biomineralization can also occur in mammals is still debated. Here we demonstrate that iron metabolism abnormalities in diseased mice can directly lead to endogenous iron biomineralization and particle formation in the spleen. Excessive iron accumulation …

PbS biomineralization using cysteine: Bacillus cereus and the sulfur rush.

Bacillus sp. Abq, belonging to Bacillus cereus sensu lato, was isolated from an aquifer in New Mexico, USA and phylogenetically classified. The isolate possesses the unusual property of precipitating Pb(II) by using cysteine, which is degraded intracellularly to hydrogen sulfide (H2S). H2S is then exported to the extracellular environment to react with Pb(II), yielding PbS (galena). Biochemical and growth tests showed that other sulfur sources tested (sulfate, thiosulfate, and methionine) were not reduced to hydrogen sulfide. Using equimolar concentration of cysteine, 1 mM of soluble Pb(II) wa

Cementum proteins: role in cementogenesis, biomineralization, periodontium formation and regeneration.

Destruction of the periodontium is normally associated with periodontal disease, although many other factors, such as trauma, aging, infections, orthodontic tooth movement and systemic and genetic diseases, can contribute to this process. Strategies (such as guided tissue regeneration) have been developed to guide and control regeneration using bioresorbable membranes and bone grafts. Although effective to a certain point, these strategies have the problem that they are not predictable and do not completely restore the architecture of the original periodontium. To achieve complete repair and r
