The coupling of 2,3,6,2',3',4',6-hepta-O-acetyl-alpha-lactosyl bromide with 1,4-di-O-benzyl-D-threitol using mercury(II) cyanide as a promoter, with subsequent deprotection of one or both of the benzyl groups, further glycosylation, and deacetylation afforded the title compounds. Lectins are carbohydrate-binding molecules that mediate a variety of biological processes. In this work, we identify and characterize a lectin from Bothrops insularis venom, with respect to its biochemical properties and theoretical structure. Initially, from a venom gland cDNA library, we cloned and sequenced a cDNA encoding a protein with high identity to snake venom lectins. Using a new approach based on the NMR spectroscopic analysis of the entire, unpurified spider venom, we identified a family of unusual sulfated nucleoside derivatives from the venom of the hobo spider, Tegenaria agrestis. These compounds are ribonucleoside mono- and disulfates derived from guanosine and xanthosine, some of which are glycosylated, bearing one or two D-fucose units. The use of NMR spectroscopy to characterize the unfractionated venom was central to the discovery of this heretofore overlooked class of venom components.

The complete amino acid sequence of the lectin from Bothrops jararacussu snake venom (BJcuL) is reported. The sequence was determined by Edman degradation and amino acid analysis of the S-carboxymethylated BJcuL derivative (RC-BJcuL) and from its peptides originated from enzymatic digestion.
