Reproductive toxicity of a nano-insecticide (chlorpyrifos) on male albino mice.

Chlorpyrifos (CPF) is a commonly used organophosphorus insecticide in agriculture, and it is linked to reproductive toxicity in males by disrupting the endocrine system. Most previous studies focus on its traditional form, neglecting its nanoform, which may pose a greater risk due to enhanced bioavailability. Our study evaluated the reproductive toxicity of a nano-emulsion of CPF (CPF-NE) in male albino mice. Mice were divided into four groups (n = 10 per group): a control and three treated groups receiving CPF-NE via oral gavage at doses of 3.79, 7.58, and 15.16 mg/kg body weight daily for 35…

Involvement of gut microbiota in chlorpyrifos-induced subchronic toxicity in mice.

Chlorpyrifos (CPF) is one of the most widely used organophosphorus pesticides all over the world. Unfortunately, long-term exposure to CPF may cause considerable toxicity to organisms. Some evidence suggests that the intestinal microbial community may be involved in regulating the toxicity of CPF. In this study, we explored if the intestinal microbial community is involved in regulating the toxicity of CPF. Adult mice were continuously exposed to CPF (4 mg/kg body weight /day) for 10 weeks with or without a 2-week pretreatment of antibiotics to change the ecological structure of intestinal mic…

Plasmid-Mediated Biodegradation of Chlorpyrifos and Analysis of Its Metabolic By-Products.

Organophosphate pesticide persistence is an emerging menace to the environment and despite this fact, its use has been enhanced due to its high efficiency. Bioremediation using microorganisms would be the only means by which these hazardous compounds could be wiped out without disturbing the environmental harmony. The current work studied the molecular mechanism of degradation of Chlorpyrifos (CP) by a bacterial consortium C5 comprising of three soil isolates Staphylococcus warneri (CPI 2), Pseudomonas putida (CPI 9) and Stenotrophomonas maltophilia (CPI 15), which unveiled that the property i…

Potential role of N-acetylcysteine on chlorpyrifos-induced neurotoxicity in rats.

Chlorpyrifos (CPF) is a widely used organophosphate insecticide with several harmful effects. N-acetylcysteine (NAC) represents an ideal antixenobiotic; it can directly enter endogenous biochemical processes and is used as adjunctive treatment for psychiatric disorders. We aimed to evaluate the neuroprotective effect of NAC as an antioxidant drug against CPF-induced neurotoxicity in adult male albino rat brains. Twenty-eight male Wister rats were allocated into four groups (n = 7) and were administered the following for 28 days: group I (control group), physiological saline (0.9% NaCl); group …

Anoxic degradation of chlorpyrifos by zerovalent monometallic and bimetallic particles in solution.

The degradation of highly toxic and persistent chlorinated organic compounds by zerovalent metals (ZVMs) has received considerable attention for in situ groundwater remediation. Due to its abundance and low toxicity, iron has been mostly applied for such purposes, despite several limitations, such as rapid surface passivation and little efficacy towards certain contaminants. Given that, we evaluated monometallic zerovalent iron (ZVI), copper (ZVC) and zinc (ZVZ), and bimetallic copper-coated ZVI (ZVI/Cu) and ZVZ (ZVZ/Cu) for anoxic reductive degradation of chlorpyrifos (CP). Two approaches to …

Toxicological interactions of chlorpyrifos and methyl mercury in the amphipod, Hyalella azteca.

The mechanism of interaction between chlorpyrifos, an organophosphate insecticide, and methyl mercury, an organometal, was assessed utilizing the amphipod, Hyalella azteca. Previous studies have demonstrated that chlorpyrifos and methyl mercury interact additively, with survival as the endpoint. In addition, exposure to chlorpyrifos and methyl mercury increased the accumulation and decreased the elimination of methyl mercury. To elucidate the mechanism responsible for these interactions, biochemical mechanisms indicative of chlorpyrifos and methyl mercury toxicity were assessed in H. azteca. B…

Hypothermia and delayed fever in the male and female rat exposed to chlorpyrifos.

Chlorpyrifos (CHL) is a commonly used organophosphate (OP) pesticide which irreversibly inhibits acetylcholinesterase activity in the CNS. Little is known regarding the thermoregulatory effects of CHL when administered orally and whether the sensitivity to CHL is affected by sex. To address these issues, male and female rats of the Long-Evans strain were administered 0, 10, 50, or 80 mg/kg CHL by gavage while core temperature (T(c)) and motor activity (MA) were monitored continuously by telemetry. Females were generally more sensitive than males to CHL. Significant hypothermic responses to CHL…

Enhancing chlorpyrifos degradation and plant growth through biochar-assisted microbial remediation.

The extensive use of chlorpyrifos (CPF) in agriculture has led to persistent soil contamination, posing serious risks to environmental and human health. Developing sustainable, in situ remediation strategies is therefore a priority. This study evaluated an integrated plant-microbe-biochar system for the remediation of CPF-contaminated soils. Indigenous CPF-degrading bacteria were isolated from agricultural soils and screened for degradation efficiency in mineral salts medium (MSM) and soil microcosms. The most efficient isolate, designated C78, was identified by 16S rRNA gene sequencing as Kle…

Electrospun nanofibers containing CdTe@ZnNi-MOF for electrochemiluminescent determination of chlorpyrifos.

Electrospun nanofibers containing CdTe@ZnNi-metal-organic frameworks (CdTe@ZnNi-MOF NFs) were used for the first time in luminol-O2 electrochemiluminescence (ECL) system to construct ECL sensor for chlorpyrifos (CPF) detection. Firstly, CdTe@ZnNi-MOF NFs obtained by blending method was used to modify the surface of a glassy carbon electrode, and then, the proposed solid-state ECL sensor was constructed by dropping luminol onto the surface of nanofiber benefiting from chitosan (CTS) viscosity. CdTe@ZnNi-MOF NFs with its large surface area and porosity can be used not only as luminol carrier but…

The protective role of Artemisia annua against chlorpyrifos toxicity in Nile tilapia.

The adverse effect of chlorpyrifos (CPF) in fish farms is well known. However, dietary supplementation of Artemisia annua (A. annua) may decrease the negative effect of chlorpyrifos because of the ameliorating effect of A. annua. The aim of this study was to see how A. annua ameliorated the toxic effects of CPF on fish growth, health, growth-related gene expression, and gills, liver, and intestinal histology for 30 days. 120 Nile tilapia fish were divided into four groups according to A. annua with/without CPF. The Cont group was fed a formulated basal diet without CPF water contamination. The…

Intermittent exposure to chlorpyrifos results in cardiac hypertrophy and oxidative stress in rats.

Forbidden in some countries due to its proven toxicity to humans, chlorpyrifos (CPF) still stands as an organophosphate pesticide (OP) highly used worldwide. Cardiotoxicity assessment is an unmet need in pesticide regulation and should be deeply studied through different approaches to better inform and generate an appropriate regulatory response to OP use. In the present study, we used our 4-week intermittent OP exposure model in rats to address the CPF effects on cardiac morphology allied with cardiovascular functional and biomolecular evaluation. Rats were intermittently treated with CPF at …

The Differential Effects of Salinity Level on Chlorpyrifos and Imidacloprid Toxicity to an Estuarine Amphipod.

Agricultural activity within coastal watersheds results in estuaries becoming the receiving environment for pesticide inputs. In estuaries, salinity can alter insecticide responses of exposed crustaceans. The acute toxicity of environmentally relevant doses of chlorpyrifos and imidacloprid were examined using the euryhaline amphipod Gammarus lawrencianus at 20 and 30 Practical Salinity Units (PSU). Responses were recorded every 24 h until an incipient (threshold) L(E)C50 was reached. For chlorpyrifos, LC50 ranged from 0.1 to 0.5 µg/L and was two-fold higher at 30 vs. 20 PSU at all time-points …

Chlorpyrifos-induced toxicity has no gender selectivity in the early fetal brain.

Organophosphorus pesticides induce gender-specific developmental neurotoxicity after birth, especially in adolescents and adults. However, whether and when the selectivity occurs in fetus remains unclear. In this study, we analyzed chlorpyrifos (CPF)-induced neurotoxicity in the early fetal brains of male and female mice. The gestational dams were administered 0, 1, 3, and 5 mg/(kg.d) CPF during gestational days (GD)7-11, and brains from the fetuses were isolated and analyzed on GD12. Fetal gender was identified by PCR technique based on male-specific Sry gene and Myog control gene. The body w…

Effects of occupational exposure to chlorpyrifos on neuropsychological function: a prospective longitudinal study.

BACKGROUND: Exposure to chlorpyrifos (CPF), an organophosphorus (OP) anticholinesterase insecticide, occurs typically in settings where multiple agents are present (e.g., agriculture) and quantitative dose measures may be absent (e.g., pesticide application). Such exposures allow few opportunities to study potential neurobehavioral effects of CPF alone. We studied the relationship between CPF exposure and behavioral function among CPF manufacturing workers, which allowed identification, measurement, and estimation of exposure and important non-exposure variables that potentially could affect s…
