[A case of occupational contact urticaria and oral allergy syndrome due to seafood].

A 20-year-old woman was referred for evaluation after about 2 years of recurrent episodes of localized urticaria during handling of several kinds of raw fish in a sushi shop, where she had worked part-time for 2 years. She had also experienced allergic symptoms such as itching and swelling of her lips, generalized urticaria, laryngeal tightness, stridor and dyspnea immediately after ingestion of raw and cooked seafood, including sole, horse mackerel, sea eel and shellfish, over the previous 1 year before referral. Skin prick tests and blood test for specific IgE antibodies were positive for ma

Molecular evolution of the ankyrin gene family.

Ankyrins are membrane adaptor molecules that play important roles in coupling integral membrane proteins to the spectrin-based cytoskeleton network. Human mutations of ankyrin genes lead to severe genetic diseases such as fatal cardiac arrhythmias and hereditary spherocytosis. To elucidate the evolutionary history of ankyrins, we have identified novel ankyrin sequences in insect, fish, frog, chicken, dog, and chimpanzee genomes and explored the phylogenetic relationships of the ankyrin gene family. Our data demonstrate that duplication of ankyrin genes occurred at two different stages. The fir

Acquisition of neural crest promoted thyroid evolution from chordate endostyle.

The endostyle is an endodermal organ unique to nonvertebrate chordates except for lamprey larvae, where it serves as forerunner to the adult thyroid. Here, we examine whether the acquisition of neural crest in the vertebrate lineage played a role in the elaboration of the endostyle. CM-DiI lineage tracing reveals a neural crest contribution to the endostyle, and CRISPR-Cas9 mutagenesis of key neural crest genes causes endostyle defects including formation of a single rather than bilobed structure. RNA sequencing reveals gene profiles characteristic of embryonic neural crest cells and Schwann c

Evolution of a new function by degenerative mutation in cephalochordate steroid receptors.

Gene duplication is the predominant mechanism for the evolution of new genes. Major existing models of this process assume that duplicate genes are redundant; degenerative mutations in one copy can therefore accumulate close to neutrally, usually leading to loss from the genome. When gene products dimerize or interact with other molecules for their functions, however, degenerative mutations in one copy may produce repressor alleles that inhibit the function of the other and are therefore exposed to selection. Here, we describe the evolution of a duplicate repressor by simple degenerative mutat

Identification, expression, function and localization of a DUF985 domain-containing hypothetical gene from amphioxus Branchiostoma belcheri.

The progress in genome sequencing has led to an increasing submission of uncharacterized hypothetical genes with the domain of unknown function, DUF985, in GenBank, and none of these genes is related to a known protein. We therefore underwent an experimental study to identify the function of a DUF985 domain-containing hypothetical gene BbDUF985 (GenBank Accession No. AY273818) isolated from amphioxus Branchiostoma belcheri (B. belcheri). BbDUF985 was successfully expressed in both prokaryotic and eukaryotic systems, and its recombinant proteins expressed in both systems definitely exhibited an

Folic acid deficiency induces premature hearing loss through mechanisms involving cochlear oxidative stress and impairment of homocysteine metabolism.

Nutritional imbalance is emerging as a causative factor of hearing loss. Epidemiologic studies have linked hearing loss to elevated plasma total homocysteine (tHcy) and folate deficiency, and have shown that folate supplementation lowers tHcy levels potentially ameliorating age-related hearing loss. The purpose of this study was to address the impact of folate deficiency on hearing loss and to examine the underlying mechanisms. For this purpose, 2-mo-old C57BL/6J mice (Animalia Chordata Mus musculus) were randomly divided into 2 groups (n = 65 each) that were fed folate-deficient (FD) or stand

The central role of angiotensin I-converting enzyme in vertebrate pathophysiology.

Genomic epidemiologic data, increasingly supported by clinical outcomes results, strongly suggest that overactivity of angiotensin I-converting enzyme (ACE) may underlie most age-related diseases. Angiotensin II, the main product of ACE, is a pleiotropic hormone, capable of serving as a neurotransmitter, growth factor, angiogenesis factor, vasoconstrictor, pro-thrombotic agent, and cytokine. So it is perhaps not surprising that the ACE D/D genotype is associated with several major psychiatric diseases, most cancers except prostate cancer (where the D/D genotype is actually protective), most ca

Gut-specific expression of cathepsin L and B in amphioxus Branchiostoma belcheri tsingtauense larvae.

Cathepsin L and B are involved in yolk processing during oocyte maturation and embryogenesis in most invertebrates and vertebrates. Here we examine the developmental expression of cephalochordate amphioxus cathepsin L (AmphiCL) and cathepsin B (AmphiCB) in the adult and developing embryos and larvae. In situ hybridization of adult amphioxus demonstrated that both AmphiCL and AmphiCB transcripts were strongly expressed in the hepatic caecum and hind-gut besides expression at various levels in other tissues examined. Whole mount in situ hybridization and RT-PCR techniques revealed that AmphiCL a

Cranial and spinal nerve organization in amphioxus and lampreys: evidence for an ancestral craniate pattern.

The spinal nerves in amphioxus are compared with the spinal and cranial nerves in lampreys. The dorsal spinal roots in amphioxus are similar to the mixed sensory and motor dorsal roots of many cranial nerves in lampreys but not to the purely sensory dorsal spinal roots in lampreys and gnathostomes. Likewise, cranial nerves V, VII, IX and X in lampreys, and all spinal nerves in amphioxus, lack a separate ventral motor root which is a constant feature of all spinal motor roots in lampreys and other vertebrates. Based on these similarities and differences, it is proposed that cranial and spinal n

Occurrence of neurotensin-immunoreactive cells in the digestive tract of lower vertebrates and deuterostomian invertebrates. A correlated immunohistochemical and radioimmunochemical study.

In the mucosal epithelium of the digestive tract of two marine teleost bony fish, one cartilaginous fish, one cyclostome, and in that of two of three representatives of deuterostomian invertebrates studied, endocrine cells of open type were found, exhibiting immunoreactivity with antisera against C-terminal sequences of mammalian neurotensin and of the structurally closely related amphibian neurohormonal peptide xenopsin. From these observations, and from those of previous studies, it is suggested that neurotensin cells do not occur in the digestive tract mucosa until at the evolutionary level

Phylogeny drives large scale patterns in Australian marine bioactivity and provides a new chemical ecology rationale for future biodiscovery.

Twenty-five years of Australian marine bioresources collecting and research by the Australian Institute of Marine Science (AIMS) has explored the breadth of latitudinally and longitudinally diverse marine habitats that comprise Australia's ocean territory. The resulting AIMS Bioresources Library and associated relational database integrate biodiversity with bioactivity data, and these resources were mined to retrospectively assess biogeographic, taxonomic and phylogenetic patterns in cytotoxic, antimicrobial, and central nervous system (CNS)-protective bioactivity. While the bioassays used wer

[Hepatic caecum of amphioxus and origin of vertebrate liver].

Previous studies on morphology and embryology revealed that the Hatschek's pit, endostyle and hepatic caecum in amphioxus were homologous to the precursors of pituitary, thyroid and liver in vertebrates, respectively. Here, we summarize the recent advances on the relationship between amphioxus hepatic caecum and the vertebrate liver as well as the origin of growth hormone/insulin-like growth factor (GH/IGF) and thyroid hormone/thyroid hormone receptor (TH/THR) signal pathways, which will provide molecular support to the homology of amphioxus hepatic caecum to vertebrate liver.

Evolution of the expression and regulation of the nuclear hormone receptor ERR gene family in the chordate lineage.

The Estrogen Related Receptor (ERR) nuclear hormone receptor genes have a wide diversity of roles in vertebrate development. In embryos, ERR genes are expressed in several tissues, including the central and peripheral nervous systems. Here we seek to establish the evolutionary history of chordate ERR genes, their expression and their regulation. We examine ERR expression in mollusc, amphioxus and sea squirt embryos, finding the single ERR orthologue is expressed in the nervous system in all three, with muscle expression also found in the two chordates. We show that most jawed vertebrates and l

Contemporary climate change hinders hybrid performance of ecologically dominant marine invertebrates.

Human activities alter patterns of biodiversity, particularly through species extinctions and range shifts. Two of these activities are human mediated transfer of species and contemporary climate change, and both allow previously isolated genotypes to come into contact and hybridize, potentially altering speciation rates. Hybrids have been shown to survive environmental conditions not tolerated by either parent, suggesting that, under some circumstances, hybrids may be able to expand their ranges and perform well under rapidly changing conditions. However, studies assessing how hybridization i

Tail regression induced by elevated retinoic acid signaling in amphioxus larvae occurs by tissue remodeling, not cell death.

The vitamin A derived morphogen retinoic acid (RA) is known to function in the regulation of tissue proliferation and differentiation. Here, we show that exogenous RA applied to late larvae of the invertebrate chordate amphioxus can reverse some differentiated states. Although treatment with the RA antagonist BMS009 has no obvious effect on late larvae of amphioxus, administration of excess RA alters the morphology of the posterior end of the body. The anus closes over, and gut contents accumulate in the hindgut. In addition, the larval tail fin regresses, although little apoptosis takes place

Hypothesis--origin of the parietal cell: microorganism?

Parietal cells, which exist in the stomach and in Meckel's diverticulum of the ileum, secrete hydrochloric acid by means of H+,K(+)-ATPase (alpha- and beta-subunits) in the human body. Although parietal cells are generated from progenitor cells (stem cells), their lifespan (approximately 150-200 days) is distinctly longer than that of surface epithelial cells (4 days), which are also derived from stem cells. Microorganisms, including bacteria (both aerobic and anaerobic), are able to pump H+ out by means of H(+)-ATPase. Of interest is that 19% of the human H+,K(+)-ATPase (alpha-subunit) compri

Ancient homeobox gene loss and the evolution of chordate brain and pharynx development: deductions from amphioxus gene expression.

Homeobox genes encode a large superclass of transcription factors with widespread roles in animal development. Within chordates there are over 100 homeobox genes in the invertebrate cephalochordate amphioxus and over 200 in humans. Set against this general trend of increasing gene number in vertebrate evolution, some ancient homeobox genes that were present in the last common ancestor of chordates have been lost from vertebrates. Here, we describe the embryonic expression of four amphioxus descendants of these genes--AmphiNedxa, AmphiNedxb, AmphiMsxlx and AmphiNKx7. All four genes are expresse

Molecular phylogeny implies exclusive, repetitive, convergent establishment of cyanobacterial symbiosis in didemnid ascidians (Tunicata: Ascidiacea: Didemnidae).

Despite the great diversity of the phylum Chordata, lifelong obligate symbiosis with cyanobacteria is only known in certain species belonging to four genera of didemnid ascidians: Didemnum, Diplosoma, Lissoclinum, and Trididemnum. This photosymbiosis in didemnid ascidians exhibits diversity in photosymbiont taxa, symbiont location in the host ascidian, and modes of vertical transmission of symbionts. While the family Didemnidae comprises over 600 colonial species, approximately 30 symbiotic species are known from tropical waters, alongside many non-symbiotic congeners. This study aimed to eluc

Antioxidant and Anti-inflammatory Extracts From Sea Cucumbers and Tunicates Induce a Pro-osteogenic Effect in Zebrafish Larvae.

Bone metabolic disorders such as osteoporosis are characterized by the loss of mineral from the bone tissue leading to its structural weakening and increased susceptibility to fractures. A growing body of evidence suggests that inflammation and oxidative stress play an important role in the pathophysiological processes involved in the rise of these conditions. As the currently available therapeutic strategies are often characterized by toxic effects associated with their long-term use, natural antioxidants and anti-inflammatory compounds such as polyphenols promise to be a valuable alternative
