Neuronal ketone body utilization couples exercise and time-restricted feeding to cognitive enhancement.

OBJECTIVE: Ketone body metabolism is linked to brain health benefits, including delaying age-related cognitive decline. Exercise, particularly when combined with an overnight fast, stimulates ketone body turnover and improves brain metabolism and cognition. Yet, whether ketone metabolism is obligatory for this response is unknown. Here, we use chronic exercise via voluntary wheel running plus time-restricted feeding (VWR + TRF) to explore whether ketones mediate exercise-induced brain health benefits in middle-aged mice. METHODS: To distinguish the roles of neuronal ketone metabolism vs. hepa

Causal enhancement of cognitive reappraisal through synchronized dorsolateral and ventrolateral prefrontal cortex activity.

The dorsolateral prefrontal cortex (DLPFC) and ventrolateral prefrontal cortex (VLPFC) are central to cognitive reappraisal, yet the mechanisms underlying their collaboration remain unclear. This study uses transcranial alternating current stimulation (tACS) to modulate neural synchrony between the DLPFC and VLPFC in the theta band, aiming to elucidate the effects of synchronous versus asynchronous neural interaction on reappraisal. It also examines whether these effects are unique to reappraisal or extend to other emotion regulation strategies. In Experiment 1, 43 participants underwent in-ph

Neuroethical issues in cognitive enhancement: Modafinil as the example of a workplace drug?

The use of cognitive-enhancing drugs by healthy individuals has been a feature for much of recorded history. Cocaine and amphetamine are mod … ding coercion and increasing disparity in society. As a society, we need to consider which forms of cognitive enhancement (e.g. pharmacological, exercise, lifelong learning) are acceptable and for which groups under what conditions and by what methods we would wish to improve and flourish.

RANDOMIZED TRIAL OF D-CYCLOSERINE ENHANCEMENT OF COGNITIVE-BEHAVIORAL THERAPY FOR PANIC DISORDER.

BACKGROUND: Initial studies have provided a mixed perspective of the efficacy of d-cycloserine (DCS) for augmenting the efficacy of exposure-based cognitive behavioral therapy (CBT) for panic disorder. In this multicenter trial, we examine the magnitude of DCS augmentation effects for an ultra-brief program of CBT. METHODS: We conducted a double-blind, controlled trial at three treatment sites, randomizing 180 adults with a primary diagnosis of panic disorder to five sessions of treatment, with study pill (50 mg DCS or matching placebo) administered 1 hr prior to the final three sessions. Two

Cholinergic Enhancement of Brain Activation in Mild Cognitive Impairment during Episodic Memory Encoding.

OBJECTIVE: To determine the physiological impact of treatment with donepezil (Aricept) on neural circuitry supporting episodic memory encoding in patients with amnestic mild cognitive impairment (MCI) using functional magnetic resonance imaging (fMRI). METHODS: Eighteen patients with MCI and 20 age-matched healthy controls (HC) were scanned twice while performing an event-related verbal episodic encoding task. MCI participants were scanned before treatment and after approximately 3 months on donepezil; HC were untreated but rescanned at the same interval. Voxel-level analyses assessed treatme

Phosphodiesterase as a Target for Cognition Enhancement in Schizophrenia.

Schizophrenia is a severe mental disorder that affects more than 1% of the population worldwide. Dopamine system dysfunction and alterations in glutamatergic neurotransmission are strongly implicated in the aetiology of schizophrenia. To date, antipsychotic drugs are the only available treatment for the symptoms of schizophrenia. These medications, which act as D2-receptor antagonist, adequately address the positive symptoms of the disease, but they fail to improve the negative symptoms and cognitive impairment. In schizophrenia, cognitive impairment is a core feature of the disorder. Therefor

Klotho Is a Neuroprotective and Cognition-Enhancing Protein.

In this chapter, we will describe what has been learned about Klotho and its potential functions in the brain. Klotho is localized in the choroid plexus and, to a lesser extent, in hippocampal neurons. Cognitive decline is a common issue in human aging affecting over 50% of the population. This cognitive decline can also be seen in animal models such as the Rhesus monkey. A long-term study undertaken by our lab demonstrated that normal brain aging in rhesus monkeys and other animal models is associated with a significant downregulation of Klotho expression. This observation substantiates data

Transcranial magnetic stimulation effects on cognitive enhancement in mild cognitive impairment and Alzheimer's disease: a systematic review and meta-analysis.

INTRODUCTION: Transcranial magnetic stimulation (TMS) is a non-invasive intervention that holds promise for improving cognitive function in individuals with Alzheimer's disease (AD). However, the effectiveness of this therapy and the optimal TMS parameters has not reached a consensus. The purpose of the meta-analysis was to systematically discern the effectiveness of different components of TMS protocols on cognitive improvement in patients with mild cognitive impairment (MCI) and AD. METHODS: The meta-analysis was preregistered on Prospero (registration number: CRD42022345482). PubMed, Web o

Aerobic Exercise for Cognitive and Functional Enhancement in Schizophrenia: Evidence, Mechanisms, and Practical Recommendations.

Schizophrenia is a psychiatric condition often characterized by cognitive deficits which precede the onset of positive symptoms and contribute significantly to enduring functional impairments and poor quality of life. Those cognitive deficits can have a profoundly negative impact on daily activities such as social relationships and work functioning. Although pharmacological interventions remain central to the treatment of schizophrenia (SZ), they have a negligible impact on cognitive functioning. Yet multiple reviews and meta-analyses support the hypothesis that Aerobic Exercise (AE) could ser

A novel cervical image recognition framework based on feature cognitive enhancement for improved performance.

Cervical cancer ranks as the fourth most prevalent cancer among women worldwide. Early diagnosis facilitates timely intervention and treatment. Traditional colposcopy is a widely employed technique for evaluating cervical lesions. Nevertheless, the growing number of patients with cervical cancer, and the rising workload of doctors may lead to misdiagnosis and underdiagnosis. Therefore, many researchers use deep learning to make paramedical diagnoses of cervical disease. However, the current neural network models for cervical image recognition lead to poor diagnostic results due to limitations

Sex matters for the enhancement of cognitive training with transcranial direct current stimulation (tDCS).

BACKGROUND: Transcranial direct current stimulation (tDCS) can influence brain network activity and associated cognitive and behavioural functions. In addition to the extensive variety in stimulation parameters, numerous biological factors drive these effects, however these are yet poorly understood. Here, we investigate one of the major biological factors by focusing on sex-dependent effects of tDCS on a challenging cognitive control task (adaptive paced auditory serial addition task [PASAT]) in healthy humans. METHODS: This sex-specific re-analysis was performed on data of 163 subjects who

Binaural Beat Stimulation for Cognitive and Psychological Enhancement in Alzheimer's Disease: A Pilot Study.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that impairs cognitive and psychological performance and adversely impacts daily routines and activities. We hypothesized that Binaural beat stimulation (BBS) enhances psychological performance by increasing connectivity in the $\theta $ -and $\alpha $ -bands over the fronto-parietal region. Twenty-five AD patients received either Binaural beat (BBS) or Standard Auditory beat stimulation (SAS) for 12 consecutive days. Psychometric assessments (Blessed Dementia Scale, Depression Anxiety Stress Scale, and Mini-Mental State Exam

Nanocarrier-Delivered Polyphenols: A Novel Strategy for Gut Microbiota Modulation and Cognitive Health Enhancement.

INTRODUCTION: The gut microbiota is a matrix ecosystem vital for the maintenance of host metabolic, immune, and neurological health. Disruption of this balance, known as gut dysbiosis, is associated with numerous diseases, including diabetes and neurological disorders. Dietary polyphenols, particularly flavonols like quercetin, have shown potential in restoring microbial balance. However, their poor solubility and low bioavailability limit therapeutic outcomes. METHODS: This review evaluates the role of dietary polyphenols in modulating gut microbiota and explores nanocarrier-based approaches

Cognitive enhancement in Korsakoff's psychosis by clonidine: a comparison with L-dopa and ephedrine.

Each of three catecholamine agonist medications (clonidine, L-dopa, and ephedrine) and placebo were administered for 2-week trials to a group of amnesics with Korsakoff's psychosis following a double-blind, counterbalanced design. During the last 3 days of each treatment, patients were given a battery of neuropsychological tests to determine the effects of the drugs on memory and related cognitive processes. Only clonidine had a significant effect on measures of anterograde amnesia, and this response was restricted to the same psychometric measures that improved with clonidine in an earlier st

Prescribing trends in cognition enhancing drugs in Australia.

BACKGROUND: This study sought to examine the trends in the prescribing of subsidized and unsubsidized cognition enhancing drugs (CEDs) in Australia over five years from 2002 to 2007. Subsidized cholinesterase inhibitor medication could be prescribed to people with mild to moderate Alzheimer's disease (AD) once a specialist physician had confirmed this diagnosis. Memantine was available for use in moderately severe AD but not subsidized. METHODS: We analyzed the Medicare Australia and Drug Utilisation Sub-Committee databases for CED prescription data, 2002-2007, by gender, age and prescriber c

Multimodal colour modulation for cognitive enhancement in intelligent rehabilitation: a systematic review and translational guidance.

BACKGROUND: Colour modulation has been investigated as a non-invasive means of supporting cognitive performance, yet translation to rehabilitation-oriented applications has been constrained by fragmented evidence, inconsistent parameterisation, and heterogeneous assessment strategies. MAIN BODY: This systematic review synthesises 75 studies published between 2011 and 2025 through a Colour-Modality-Cognition framework that links stimulus design, measurement strategy, and cognitive goals. Across the evidence base, a graded and context-dependent pattern emerged. Contrast-related variables, parti

6,7-Dihalo-Benzothiadiazines as Potent and Selective AMPA Receptor Modulators for Cognitive Enhancement and Neuroprotection.

α-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are tetrameric ionotropic glutamate receptors that mediate fast excitatory synaptic transmission in the brain and represent important therapeutic targets for neurological disorders. Positive allosteric modulators (PAMs) of AMPA receptors enhance rapid excitatory signaling by increasing receptor's sensitivity to glutamate and have been widely explored as agents to improve cognitive function in central nervous system (CNS) diseases. Structural modification of 3,4-dihydro-2H-1,2,4-benzothiadiazine 1,1-dioxides (BTDs) analogs is

Neurostimulation for cognitive enhancement in Alzheimer's disease (the NICE-AD study): a randomized clinical trial.

New therapies for symptoms in Alzheimer's disease (AD) are urgently needed. Prior studies suggest that transcranial direct current stimulation (tDCS), a noninvasive neuromodulatory method, may be a safe and potentially effective treatment, but conclusions have been limited by small-sample sizes and brief stimulation protocols. This double-blind randomized trial involving 100 older adults with mild-to-moderate AD examines effects of 6 months of at-home active tDCS or sham delivered over the dorsolateral prefrontal cortex. The primary outcome is global cognitive performance. Secondary outcomes i
