A transcriptional profile of the H2O2-adapted Corynebacterium glutamicum HA strain reveals a list of upregulated regulatory genes. Among them, we selected ORF NCgl2298, designated osrR and analyzed its role in H2O2 adaptation. The osrR-deleted (ΔosrR) mutant had defective growth in minimal medium, which was even more pronounced in an osrR deletion mutant of an HA strain. The ΔosrR strain displayed increased sensitivity to H2O2. In addition to H2O2 sensitivity, the ΔosrR strain was found to be temperature-sensitive at 37 °C. 2D-PAGE analysis of the ΔosrR mutant found that MetE and several other

During extracellular glutamate overproduction in Corynebacterium glutamicum, an increase in metabolic flux toward glutamate synthesis occurs without an increase in the transcriptional or translational levels of relevant enzymes, suggesting the involvement of unprecedented post-translational regulatory mechanisms. Here, we investigated the relationship of enzyme condensation with the metabolic shifts that lead to glutamate overproduction. We systematically analyzed the subcellular localization of enzymes involved in glutamate overproduction using fluorescent protein tagging to identify enzyme c

D-chiro-inositol (DCI), a key signaling molecule in glucose and lipid metabolism and steroid synthesis, plays important roles in ameliorating human metabolic disorders. To achieve de novo DCI biosynthesis in Corynebacterium glutamicum, the inositol catabolism-blocked chassis strain DCI‑1 was engineered by dynamically downregulating pgi, pfkA, and zwf genes. The most effective strain DPa‑1 produced 2.89 g/L myo-inositol (MI), the direct precursor of DCI, and 0.83 g/L DCI, with the total inositol over four times the control. Subsequently, to promote the conversion of MI to DCI and the supply of

UNLABELLED: Bacteria in the Mycobacteriales order like mycobacteria and corynebacteria surround themselves with a multilayered cell envelope. Their cytoplasmic membrane is fortified by a peptidoglycan cell wall that is decorated with branched arabinogalactan (AG) polymers. The AG glycans are further modified with mycolic acids to form an outer membrane. Biogenesis of this mycomembrane requires the transport of mycolates from their site of synthesis in the cytoplasmic membrane to the cell surface. How mycolate transport is controlled and coordinated with AG synthesis has remained unclear. Mycol

BACKGROUND: The nitrogen containing aromatic compound indole is known for its floral odor typical of jasmine blossoms. Due to its characteristic scent, it is frequently used in dairy products, tea drinks and fine fragrances. The demand for natural indole by the flavor and fragrance industry is high, yet, its abundance in essential oils isolated from plants such as jasmine and narcissus is low. Thus, there is a strong demand for a sustainable method to produce food-grade indole.
RESULTS: Here, we established the biotechnological production of indole upon L-tryptophan supplementation in the bac

In this study, we analysed the whcD gene from Corynebacteriumglutamicum, which encodes a homologue of whiB, a Streptomycescoelicolor gene required for the sporulation of aerial hyphae. Deletion of the gene (ΔwhcD) severely affected cell growth in C. glutamicum. The ΔwhcD strain exhibited a large filamentous, branched and bud-shaped morphology with multiple septa. The transcription levels of the cell division genes involved in Z-ring assembly and septal peptidoglycan synthesis, including ftsZ, sepF, ftsQ and ftsI, were markedly decreased in the ΔwhcD strain. The divIVA gene, which is responsibl

BACKGROUND: Antimicrobial peptides (AMPs) such as pediocin PA-1 are attractive for food biopreservation and infection control, but their broader use is limited by low recombinant yields and high production costs. Corynebacterium glutamicum has emerged as a robust GRAS chassis for heterologous peptide and protein production, yet commonly used shuttle vectors provide only moderate plasmid copy numbers and expression capacities. In particular, existing pediocin PA-1 processes in C. glutamicum rely on standard pBL1- or pCG1-family vectors that do not yet leverage replication-origin engineering. RE

Peptidoglycan (PGN) is a unique component of prokaryotic cell walls with immune-enhancing capacities. Here, we extracted PGN from Corynebacterium glutamicum, a by-product of amino acid fermentation, using the trichloroacetic acid (TCA) method. SDS-PAGE analysis confirmed the presence of PGN, with a band of approximately 28 kDa. Further analysis was conducted through amino acid analysis, FTIR, and MALDI-TOF/TOF MS, and the results showed that the chemical structural monomer of PGN is NAG-(β-1,4-)-NAM-l-Ala-d-Glu-l-Lis-d-Ala. The immune activation effects of PGN were evaluated in a RAW264.7 cell

The development of CRISPR interference (CRISPRi) technology has dramatically increased the pace and the precision of target identification during platform strain development. In order to develop a simple, reliable, and dual-inducible CRISPRi system for the industrially relevant Corynebacterium glutamicum, we combined two different inducible repressor systems in a single plasmid to separately regulate the expression of dCas9 (anhydro-tetracycline-inducible) and a given single guide RNA (IPTG-inducible). The functionality of the resulting vector was demonstrated by targeting the l-arginine biosy

We describe the development of a new secretory production system for the enhanced production of a single-chain variable fragment (scFv) against the anthrax toxin in Corynebacterium glutamicum. For efficient secretory production of the antibody fragment, the following components were examined: (1) signal peptides, (2) codon usage of antibody fragment, (3) promoters, (4) 5' untranslated region (5' UTR) sequence, and (5) transcriptional terminator. Among all the systems examined, the use of a codon-optimized gene sequence, a Sec-dependent PorB signal peptide, and a fully synthetic H36 promoter, a

Isopropanol is a major chemical used in the chemical industry as a chemical intermediate, organic solvent, and hand sanitizer. In this study, engineered Corynebacterium glutamicum strains capable of high-cell-density fermentation were developed to efficiently produce isopropanol. Transcriptome analysis identified genes that showed significant expression changes under high-cell-density fermentation condition. Based on this analysis, metabolic pathways involved in fermentation byproducts were inhibited, leading to improved isopropanol production under high-cell-density fermentation. To address t

Inositols (cyclohexanehexols) comprise nine isomeric cyclic sugar alcohols, several of which occur in all domains of life with various functions. Many bacteria can utilize inositols as carbon and energy sources via a specific pathway involving inositol dehydrogenases (IDHs) as the first step of catabolism. The microbial cell factory Corynebacterium glutamicum can grow with myo-inositol as a sole carbon source. Interestingly, this species encodes seven potential IDHs, raising the question of the reason for this multiplicity. We therefore investigated the seven IDHs to determine their function,

We previously demonstrated efficient L-valine production by metabolically engineered Corynebacterium glutamicum under oxygen deprivation. To achieve the high productivity, a NADH/NADPH cofactor imbalance during the synthesis of l-valine was overcome by engineering NAD-preferring mutant acetohydroxy acid isomeroreductase (AHAIR) and using NAD-specific leucine dehydrogenase from Lysinibacillus sphaericus. Lactate as a by-product was largely eliminated by disrupting the lactate dehydrogenase gene ldhA. Nonetheless, a few other by-products, particularly succinate, were still produced and acted to

2,3-Butanediol (2,3-BD) is a versatile bulk chemical, serving as a high-boiling green solvent, a key monomer for polyesters and polyurethanes, and a crucial intermediate in the production of fine chemicals and cosmetics. Additionally, it is a key intermediate for pharmaceuticals and agrochemicals, supporting green chemistry. Microbial production of 2,3-BD offers a sustainable bioprocess but faces the challenge of high-concentration product inhibition. In an effort to improve 2,3-BD tolerance and production in Corynebacterium glutamicum, the engineered strain CGK4 was subjected to atmospheric a

The putative two-component system (TCS) CgtRS5, encoded by the ncgl2572-ncgl2573 gene cluster, plays a critical role in environmental adaptation in Corynebacterium glutamicum. Comparative transcriptomic analysis between the wild-type strain and a ΔcgtRS5 mutant revealed three upregulated and 107 downregulated genes in the mutant. Among these, genes involved in iron metabolism (ncgl1959), aromatic compound degradation (ncgl2320), and drug resistance (cssR) were significantly downregulated. Phenotypic assays demonstrated that the ΔcgtRS5 mutant exhibited impaired growth in media supplemented wit

Strain development is critical for microbial production of bio-based chemicals. The stereo-complex form of polylactic acid, a complex of poly-L- and poly-D-lactic acid, is a promising polymer candidate due to its high thermotolerance. Here, we developed Corynebacterium glutamicum strains producing high amounts of L- and D-lactic acid through intensive metabolic engineering. Chromosomal overexpression of genes encoding the glycolytic enzymes, glucokinase, glyceraldehyde-3-phosphate dehydrogenase, phosphofructokinase, triosephosphate isomerase, and enolase, increased L- and D-lactic acid concent

Single-cell technologies are emerging as a complementary approach to conventional laboratory-scale cultivation setups. Particularly in microfluidic cultivation and screening, precisely controlled environmental conditions and defined minimal media become important for quantitative studies. Nevertheless, conventional bulk media optimized for other scales, are often employed despite differences in nutrient requirements across varying scales and cultivation modes. This study demonstrates the limitations of using bulk media for single-cell analysis of Corynebacterium glutamicum and highlights the n

Protocatechuic acid (PCA) is a valuable phenolic compound, and its rising demand has facilitated sustainable PCA bioproduction. Compared to glucose, low-cost methanol and pentose sugars like xylose do not have food-competing uses. Thus, we decided to engineer Corynebacterium glutamicum to coutilize them for PCA production. First, we introduced the PCA biosynthesis pathway into a methanol-essential strain and deleted its degradation pathway to achieve PCA production from methanol. Subsequently, we repaired the rpe deletion and introduced evolved xylAB (gX), achieving the highest PCA yield of 0.

This study focused on improving the flavor quality of seasonings, and enzymatic hydrolysis of soybean protein isolate (SPI) seasoning via traditional technology may lead to undesirable flavors. Herein, we aimed to develop a new type of SPI seasoning through microbial fermentation to improve its flavor quality. The effect of Corynebacterium glutamicum fermentation on the flavoring compounds of seasonings in SPI enzymatic hydrolysate was examined. Sensory evaluation showed that the SPI seasoning had mainly aromatic and roasted flavor, and the response signals of S18 (aromatic compounds), S24 (al
