Pseudomonas pseudomallei, the causative microorganism of melioidosis, was grown in Mueller-Hinton liquid medium, and glycoprotein fractions were separated from the culture filtrate by ammonium sulfate precipitation, gel-filtration with Sephadex G-75, and column chromatography with DEAE-cellulose. The fractions revealed acid phosphatase activity, and reacted to the sera from melioidosis patient in gel-diffusion precipitation assay.

We have defined one type of acid alpha-glucosidase and two types of neutral alpha-glucosidases from quail skeletal muscle on the basis of differences in the elution patterns on a DEAE-cellulose column. The appearance of the two neutral alpha-glucosidase isoenzymes was age-dependent. A decrease in acid alpha-glucosidase activity was demonstrated in Japanese quails with glycogenosis type II. The characteristics of these three alpha-glucosidase isoenzymes are described.

An improved method has been developed for the detection of heterozygotes for feline and human mucopolysaccharidosis VI. Arylsulfatase-A and -B activities were assayed in leukocyte extracts following separation of the enzymes by batch chromatography on DEAE cellulose. Determination of arylsulfatase-B specific activities did not permit accurate heterozygote identification, whereas the arylsulfatase-A to arylsulfatase-B activity ratio discriminated all 16 obligate heterozygotes for the feline and human disorders.

A microchromatographic method for separating hemoglobin A2 from hemoglobin S and their subsequent quantitation on DEAE-cellulose has been improved. The new procedure is more versatile, since the same system can be used when hemoglobin S is present or absent in the sample without changing to a longer column or another buffer system. Also, the technic is less sensitive to small changes in pH of buffers or resin than are other procedures.

Repair synthesis in human cells in tissue culture can be readily separated from semi-conservative DNA synthesis with the aid of a benzoylated naphthoylated DEAE cellulose (BND-cellulose) column. Antigen PDE1 was purified from pigeon dropping extracts by chromatography on DEAE-cellulose and Bio-Gel A-1.5. It was characterized as a glycoprotein, containing 88% protein and 12% carbohydrate, with a molecular weight of approximately 200,000. PDE1 appears to be derived from pigeon intestinal IgA, showing the typical immunoglobulin structure of heavy and light chains. This antigen represents about 24% of the dry weight of the unfractionated pigeon dropping extract.

The functioning of metallothioneins in the liver of rats, poisoned with copper sulfate and cadmium sulfate has been investigated. By sequential chromatography on sephadex G-50 and DEAE-cellulose the authors obtained metallothioneins (MT-1, MT-1A, MT-2, MT-2a), which differ in molecular weight and composition of associated metals. Heavy metal poisoning leads to activation of synthesis and metal-binding function of metallothioneins, as well as to changes in the composition of their isoforms.

Soluble antigen from bovine blood with a Babesia bigemina parasitemia of 5-6% formed 4 precipitin lines in gel diffusion tests with antiserum from infected cattle. Antigen was obtained from plasma and hemolysate by elution from DEAE cellulose columns and (NH4)2SO4 precipitation as well as from washed parasite-erythrocyte stroma by sonication or freeze-thawing. About 5 ml of antigen suitable for diagnostic tests could be extracted from 200 ml of infected blood.

Following removal of sialic acid by neuraminidase treatment the activity of the beta-glucuronidase inhibitor was remarkably decreased, but the antigenic determinant was not affected. A partial common antigen to the inhibitor was isolated from porcine small intestine, by successive fractionation of trypsin extraction of the latter on Sephadex G-150, DEAE-cellulose and Sepharose 4B column chromatography. Trypanosoma vivax is the principal etiological agent of bovine trypanosomosis, a widely disseminated disease in tropical and subtropical regions. Here, we present a simple and reproducible method for the purification of T. vivax from experimentally infected and immunosuppressed sheep, using an isopycnic Percoll gradient, followed by DEAE-cellulose chromatography, with an estimated yield of 11-15%. This method could be used for the purification of T. vivax geographical isolates from various locations and from different natural hosts.

Mice were found to be protected against Taenia taeniaforMis infection by passive transfer of serum collected from donors 28 days after infection. The protective activity resided exclusively in the first fraction of 7S immunoglobulins eluting from DEAE-cellulose at pH 5.8 with 0.05 M phosphate buffer. This fraction contained 7Sgamma1 and 7Sgamma2 immunoglobulins but no detectable gammaA, gammaM or skin sensitizing activity. Fractions containing 7Sgamma 2 alone were ineffective in passive transfer.

Cellulose acetate electrophoresis, chondroitinase digestions, and reaction product and infrared analyses were used to characterize the component GAGs.
