Disulfidptosis links the pathophysiology of ulcerative colitis and immune infiltration in colon adenocarcinoma.

Ulcerative colitis (UC), a chronic inflammatory bowel disease, significantly increases the risk of colon adenocarcinoma (COAD). Disulfidptosis, a novel form of programmed cell death, has been implicated in various diseases, including UC. This study investigates the expression of disulfidptosis-related genes, particularly CD2AP and MYH10, in UC and COAD. Through analysis of public datasets, we found MYH10 significantly upregulated and CD2AP downregulated in UC compared to healthy controls, with consistent patterns in COAD. Immune infiltration analysis revealed correlations between these genes a…

The molecular mechanisms of disulfidptosis and its role in Diabetes mellitus and its complications.

Diabetes mellitus (DM) is a metabolic disorder characterized by chronic hyperglycemia, with pancreatic β-cell dysfunction and oxidative stress playing central roles in its pathogenesis. Recent studies have identified disulfidptosis as a novel form of regulated cell death driven by disulfide stress,a condition marked by abnormal intracellular disulfide accumulation and NADPH/NADP+ imbalance. This process is particularly prominent in glucose-deprived cells with high expression of the cystine transporter SLC7A11, where impaired disulfide reduction leads to cytoskeletal collapse and cell death. Di…

Disulfidptosis-Related Genes as Novel Biomarkers and Therapeutic Targets in Dilated Cardiomyopathy.

BACKGROUND: Dilated cardiomyopathy (DCM) is a severe cardiac condition characterized by ventricular dilation and impaired systolic function, leading to heart failure and sudden death. Current treatments have limited efficacy in reversing disease progression. Recent research suggests that disulfidptosis, a novel cell death mechanism, may play a role in DCM pathogenesis, though its specific involvement remains unclear. METHODS: This study utilized multiple GEO datasets to analyze the expression of 24 disulfidptosis-related genes (DiGs) in DCM patients and healthy controls. Differential expressi…

Unlocking the Potential of Disulfidptosis: Nanotechnology-Driven Strategies for Advanced Cancer Therapy.

Tumor tissues exhibit elevated oxidative stress, with the cystine-glutamate transporter xCT solute carrier family 7 member 11 (xCT/SLC7A11) protecting cancer cells from oxidative damage by facilitating cystine uptake for glutathione synthesis. Disulfidptosis, a newly identified form of programmed cell death (PCD), occurs in cells with high xCT/SLC7A11 expression under glucose-deprived conditions. Distinct from other PCD pathways, disulfidptosis is characterized by aberrant disulfide bond formation and cellular dysfunction, ultimately resulting in cancer cell death. This novel mechanism offers …

Immunometabolic regulation of disulfidptosis in orthopedic diseases: mechanistic heterogeneity and therapeutic targets.

Disulfidptosis is a novel form of programmed cell death triggered by cystine metabolic disorders and disulfide stress, initially studied primarily in the context of tumors. In recent years, its role in the occurrence and development of orthopedic diseases has gained increasing attention. This review systematically explores the dual regulatory mechanisms of disulfidptosis in degenerative orthopedic diseases, such as intervertebral disc degeneration, osteoporosis, and osteoarthritis, as well as in malignant bone tumors like osteosarcoma, along with their immunometabolic basis. The research findi…

Harnessing disulfidptosis-ferroptosis synergy to potentiate radiotherapy in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) displays pronounced molecular heterogeneity and metastatic propensity, which correlates with its poor c … duction and inducing cystine accumulation alongside cysteine depletion, which concurrently triggers disulfidptosis and relieves ferroptosis resistance. Meanwhile, the released ferroptosis inducer FIN56 further amplifies ferroptosis by degrading GPX4. In addition to this, BWFP enhances RT efficacy through radiosensitization effects, consequently intensifying DNA …

Beyond biochemical cascades: the biophysical execution of disulfidptosis via actin network collapse.

The discovery of disulfidptosis has identified a distinct form of regulated cell death in which metabolic redox failure is translated into biophysical disruption of the actin cytoskeleton. This review synthesizes current evidence on the molecular machinery of actin remodeling under disulfide stress, with particular emphasis on the Rac1-WRC-Arp2/3 signaling axis and other actin-regulatory nodes. Mechanistically, disulfidptosis can be conceptualized as a redox-to-mechanics transition. In SLC7A11-high cells, glucose deprivation limits pentose phosphate pathway-derived NADPH production and weakens…

Inhibition of thioredoxin reductase 1 sensitizes glucose-starved glioblastoma cells to disulfidptosis.

Disulfidptosis is a recently identified form of cell death characterized by the aberrant accumulation of cellular disulfides. This process primarily occurs in glucose-starved cells expressing higher levels of SLC7A11 and has been proposed as a therapeutic strategy for cancers with hyperactive SCL7A11. However, the potential for inducing disulfidptosis through other mechanisms in cancers remains unclear. Here, we found that inhibiting thioredoxin reductase 1 (TrxR1), a key enzyme in the thioredoxin system, induces disulfidptosis in glioblastoma (GBM) cells. TrxR1 expression is elevated in GBM w…

The mitochondria enzyme OGDH defends against disulfidptosis by licensing METTL3-regulated NRF2 translation.

Disulfidptosis is a form of regulated cell death triggered by disulfide stress resulting from glucose starvation. The capacity to evade disulfidptosis is crucial for tumour cells to withstand glucose-limited environments. Here we demonstrate that OGDH, a rate-limiting enzyme in citric acid cycle, is critical for conferring resistance to disulfidptosis. High expression of HSPA9 in melanoma protects OGDH from glucose deprivation-induced oxidative inactivation, thereby ensuring OGDH-generated succinyl-CoA for METTL3 succinylation. Succinylated METTL3 recognizes m6A modification on NRF2 mRNA to li…

Regulation mechanisms of disulfidptosis-related genes in ankylosing spondylitis and inflammatory bowel disease.

INTRODUCTION: Disulfidptosis is a recently identified form of cell death that contributes to maintaining the internal environment balance of an organism. However, the molecular basis of disulfidptosis in ulcerative colitis (UC), ankylosing spondylitis (AS), and Crohn's disease (CD) has not been thoroughly explored. METHODS: Firstly, the differentially expressed genes (DEGs) and disulfidptosis-associated genes (DAGs) were obtained through differential analysis between diseases (AS, CD, and UC) and control groups. After the disulfidptosis score was acquired using the single-sample gene set enri…

Exploration of the Role of M2 Macrophages in Hepatocellular Carcinoma: Insights into Disulfidptosis and Cellular Interactions.

BACKGROUND: The interplay between regulated cell death processes, especially disulfidptosis, and the immune microenvironment in hepatocellular carcinoma (HCC) remains poorly understood. Therefore, this study aimed to elucidate these relationships systematically at single-cell resolution. METHODS: Multi-omics data from TCGA (n = 315) were integrated with single-cell RNA sequencing (scRNA-seq) data from 21 HCC samples. Disulfidptosis-related subtypes were identified using non-negative matrix factorization (NMF) clustering of co-expressed long non-coding RNAs (lncRNAs). At the single-cell level,…

Leveraging a disulfidptosis-based signature to characterize heterogeneity and optimize treatment in multiple myeloma.

BACKGROUND: Disulfidptosis is an emerging type of programmed cell death related to ROS accumulation and aberrant disulfide bond formation. Multiple myeloma (MM) is the second most prevalent hematologic malignancy characterized by a high synthesis rate of disulfide bond-rich proteins and chronic oxidative stress. However, the relationship between disulfidptosis and MM is still unclear. METHODS: Using the non-negative matrix factorization and lasso algorithm, we constructed the disulfidptosis-associated subtypes and the prognostic model on the GEO dataset. We further explored genetic mutation m…
