Regulation of the NRF2 transcription factor by andrographolide and organic extracts from plant endophytes.

The transcription factor NF-E2 Related Factor-2 (NRF2) is an important drug target. Activation of NRF2 has chemopreventive effects in cancer and exerts beneficial effects in a number of diseases, including neurodegenerative diseases, inflammatory diseases, hepatosteatosis, obesity and insulin resistance. Hence, there have been great efforts to discover and characterize novel NRF2 activators. One reported NRF2 activator is the labdane diterpenoid andrographolide. In this study, we identified the mechanism through which andrographolide activates NRF2. We showed that andrographolide inhibits the

Diversity of cultivated and uncultivated actinobacterial endophytes in the stems and roots of rice.

A dual approach consisting of cultivation and molecular retrieval of actinobacterial 16S rRNA genes was used to characterize the diversity of actinobacterial community inhabiting interior of rice stems and roots. Streptomyces is the most frequently isolated genus from rice stems and roots. Forty-five clones chosen randomly among 250 clones in the 16S rRNA gene clone library from roots were affiliated with nine genera of actinobacteria and uncultured actinobacteria (Mycobacterium, Streptomyces, Micromonospora, Actinoplanes, Frankia, Dactylosporangium, Amycolatopsis, Corynebacterium, Rhodococcus

Functional characterization of fungal endophytes with antagonistic and plant growth-promoting activities in maize.

Fungal endophytes play a crucial role in plant-microbe interactions by asymptomatically colonizing host tissues, enhancing plant growth, and providing defense. In the present study, three distinct endophytic fungi, namely Fusarium oxysporum CJR-1 (GenBank Accession No. PZ477045), Penicillium sp. CJR-2 (GenBank Accession No. PZ477429) and Aspergillus cf. terreus CJR-4 (GenBank Accession No. PZ477430) were isolated from Crotalaria juncea L. plants and evaluated for antagonistic activity against phytopathogenic fungi and growth promotion in maize. Among the isolates, CJR-4 showed potent antagonis

Dendrobium officinale Endophytes May Colonize the Intestinal Tract and Regulate Gut Microbiota in Mice.

Dendrobium officinale is a traditional Chinese medicine for treating gastrointestinal diseases by nourishing "Yin" and thickening the stomach lining. To study whether D. officinale endophytes can colonize the intestinal tract and regulate gut microbiota in mice, we used autoclave steam sterilizing and 60Co-γ radiation to eliminate D. officinale endophytes from its juice. Then, high-throughput ITS1-ITS2 rDNA and 16S rRNA gene amplicons were sequenced to analyze the microbial community of D. officinale endophytes and fecal samples of mice after administration of fresh D. officinale juice. Steril

Biopolymer-based seed biopriming with fungal endophytes suppresses Fusarium wilt and modulates growth in tomato.

Biopolymer-based seed biopriming is an emerging strategy for delivering endophytic fungal biocontrol agents while maintaining seed viability and fungal persistence, yet the compatibility of carrier matrices with dark septate endophytes (DSE), and the potential of this fungal group against Fusarium oxysporum f. sp. lycopersici (FOL) in tomato, remain unexplored. Three biopolymer matrices - sodium alginate (SA), gum arabic (GA) and guar gum (GG) - were evaluated as carriers for ten endophytic fungal isolates: Torula sp. (BC01, BC11), Sordaria fimicola (BC06), Clarireedia calopus (BC17, BC18), Tr

Inclusion of endophytes in hydroponic cultivation of basils enhances hydroxycinnamic acid levels and antioxidant capacity.

The lack of microorganisms in soil-less hydroponic farming can limit the accumulation of nutrients and secondary metabolites. This work was then aimed for the first time, to identify and quantify polyphenols, determine antioxidant properties of hydroponically grown basils, and assess the effects of endophytic bacteria. Data showed that of the eleven endophytes investigated, five increased (p < 0.05) total phenolic contents to 4.5 ± 0.3-5.8 ± 1.2 mg GAE/g F.W. relative to 3.1 ± 0.5 mg GAE/g for the control basils. The increase was due to the enhanced biosynthesis of hydroxycinnamic acid derivat

Isolation, taxonomic analysis, and phenotypic characterization of bacterial endophytes present in alfalfa (Medicago sativa) seeds.

A growing body of evidence has reinforced the central role of microbiomes in the life of sound multicellular eukaryotes, thus more properly described as true holobionts. Though soil was considered a main source of plant microbiomes, seeds have been shown to be endophytically colonized by microorganisms thus representing natural carriers of a selected microbial inoculum to the young seedlings. In this work we have investigated the type of culturable endophytic bacteria that are carried within surface-sterilized alfalfa seeds. MALDI-TOF analysis revealed the presence of bacteria that belonged to

Effect of UV-A on endophyte colonisation of Arabidopsis thaliana.

UV-A, an important part of sunlight radiation, is typically absent in experiments on plant-endophyte interactions. We examined the impact of UV-A in the 350-400 nm range (UV-A1 waveband) on the plant interactions with fungal endophytes belonging to different taxonomic groups: Paraphoma chrysanthemicola, Phomopsis columnaris, Diaporthe eres, Mucor sp., and yeast Sporobolomyces ruberrimus. Physiologically relevant levels of UV-A did not substantially affect the colonisation of shoots and roots by endophytes. UV-A upregulated the expression of genes involved in the establishment of symbiosis. Spe

Endophytic Fungi: A Source of Potential Antifungal Compounds.

The emerging and reemerging forms of fungal infections encountered in the course of allogeneic bone marrow transplantations, cancer therapy, and organ transplants have necessitated the discovery of antifungal compounds with enhanced efficacy and better compatibility. A very limited number of antifungal compounds are in practice against the various forms of topical and systemic fungal infections. The trends of new antifungals being introduced into the market have remained insignificant while resistance towards the introduced drug has apparently increased, specifically in patients undergoing lon

Endophytic fungi as models for the stereoselective biotransformation of thioridazine.

The stereoselective kinetic biotransformation of thioridazine, a phenothiazine neuroleptic drug, by endophytic fungi was investigated. In general, the sulfur of lateral chain (position 2) or the sulfur of phenothiazinic ring (position 5) were oxidated yielding the major human metabolites thioridazine-2-sulfoxide and thioridazine-5-sulfoxide. The quantity of metabolites biosynthesized varied among the 12 endophytic fungi evaluated. However, mono-2-sulfoxidation occurred in higher ratio and frequency. Among the 12 fungi evaluated, 4 of them deserve prominence for presenting an evidenced stereose

Extending the Metabolite Diversity of the Endophyte Dimorphosporicola tragani.

Fungi are one of the most prolific sources of microbial secondary metabolites. The production of new metabolites can be achieved using multiple fermentation conditions and by adding small-molecule effectors, including epigenetic modifiers. In the framework of our Natural Product screening programme targeting the discovery of new antimicrobial compounds, we applied multiple fermentation conditions and adsorptive polymeric resins on a large collection of fungal endophytes, to increase and stimulate their fungal secondary metabolite production. During this work the endophytic fungus Dimorphospori

From Traditional to Omics-Driven: Emerging Strategies for Isolation, Cultivation, and Identification of Plant Endophytes.

Plant endophytes can regulate host plant growth, improve stress resistance, and facilitate the biosynthesis of secondary metabolites, with great research value and application potential. However, traditional approaches for the isolation, cultivation and identification of plant endophytes are constrained by low culturability, limited species diversity, and loss of their original ecological functions inside host tissues. In recent years, integrated multi-omics strategies combining metagenomics, metatranscriptomics, and metaproteomics have exhibited the greatest potential to mitigate culturabilit

Metagenomic profiling of tobacco root endophytes reveals a disease-suppressive Enterobacter strain against Fusarium solani.

BACKGROUND: Tobacco root rot, caused by Fusarium species, is a persistent soil-borne disease that threatens tobacco production. To identify endophytic contributors to disease suppression, this study compared the root endophytic microbiomes of healthy and diseased tobacco plants using metagenomic sequencing and isolated functional bacteria from healthy roots. RESULTS: Metagenomic analysis of 30 root samples (223 Gb) generated 2.9 million non-redundant genes and identified 1,953 core genera. Healthy plants contained distinct endophytic microbial communities enriched in bacterial taxa and pathwa
