BACKGROUND: With the progress of technology in automated hematology analyzers, in the vast majority of cases, nucleated red corpuscles (NRC) can be automatically identified by most types of automated hematology analyzers, thus correcting the leukocyte count and avoiding pseudoleukocytosis by the analyzers themselves. The objective of the study was to explore pseudoleukocytosis due to immature erythroid precursors and/or erythrocyte dysplasia in the peripheral blood resulting from different rare situations.

Pitted erythrocytes counts are used in the quantitative assessment of functional hyposplenism and in the prediction of recurrence of splenic function in splenectomized subjects. Pitted erythrocyte counts were investigated in 16 patients splenectomized for hereditary spherocytosis, in 21 patients splenectomized for other reasons and in 23 healthy controls. In both groups of splenectomized patients the pitted erythrocyte counts were equally increased compared with the healthy controls.

To account for the conflict between the excessive destruction of erythrocytes and the number of parasitized erythrocytes in dogs infected with Babesia gibsoni, we examined the correlation between anti-erythrocyte membrane antibody level (AEMAL) and the number of erythrocytes (RBC count) in dogs with experimentally induced babesiosis using hematological examination and an enzyme linked immunosorbent assay (ELISA). In the infected dogs without splenectomy, more prominent reduction in RBC count accompanied with the elevated AEMAL was presented than anticipated from parasitemia until the 21st day.

Splenic function was assessed using 'pitted' erythrocyte counts in 61 first degree relatives of patients with coeliac disease. 'Pitted' erythrocyte counts were normal in 12 parents, but were raised in 20% of 49 siblings and/or children of coeliac patients. Erythrocyte filterability, reticulocyte count, total serum bilirubin, and CO hemoglobin percent saturation (COHb) were measured throughout one menstrual cycle in 17 women. The filterability was significantly depressed during the progesterone phase compared with the estrogen phase. BACKGROUND: Platelet count is essential for the diagnosis and management of hemostasis abnormalities. Although existing platelet count methods installed in common hematology analyzers can correctly count platelets in normal blood samples, they tend to miscount platelets in some abnormal samples. The newly developed PLT-F channel in the XN-Series hematology analyzer (Sysmex) has been reported to be a reliable platelet count system, even in abnormal samples. To determine the effects of blood transfusions on splenic function in older patients with sickle cell anemia, we investigated splenic function in 12 patients who had had cerebrovascular accidents and who were being treated at two collaborating centers using different transfusion protocols. Haematological changes were monitored in 20 experimentally infected and 70 clinical cases of feline babesiosis. There was a rapid drop in the haematocrit, haemoglobin and erythrocyte count. Experimental animals were most anaemic about 3 weeks after infection. Variations were encountered. There were no definite trends in the erythrocyte indices although the erythrocytes were often macrocytic and hypochromic. No significant changes were found in total leukocyte counts.
