Inhibition of human phospholipases A2 by cis-unsaturated fatty acids and oligomers of prostaglandin B1.

Inhibition of human phospholipases A2 by cis-unsaturated fatty acids and their oxidative metabolites and/or polymers was studied using partially purified human phospholipases A2 and [1-14C]oleate labelled, autoclaved E. coli as substrate. As previously reported for other phospholipases A2, oleic and arachidonic acids inhibited human synovial fluid phospholipase A2 with IC50s of 15 and 30 microM respectively. Air oxidation of arachidonic acid or hydroxylation of oleic acid (12-hydroxy-oleate) substantially relieved that inhibition. Similarly, the enzymatically oxidatized metabolite of arachidon

Changes in unsaturated fatty acids in serum lipids of hamsters infected with schistosomes (S. mansoni).

Measurements have been made of the fatty-acid composition of lipids in normal hamster sera compared with sera of hamsters infected with Schistosoma mansoni. The results show a consistent decrease of linoleic and eicosapentaenoic acids, and an increase in arachidonic acid in the phosphatidylcholine and lysophosphatidylcholine of the infected animals. Arachidonic acid was also increased in cholesterol ester, and docosahexaenoic acid was raised in free fatty acid and in phosphatidylcholine in sera from infected animals.

Gut microbiota regulates serum metabolites in mice with nonalcoholic fatty liver disease via gut metabolites: mechanisms involving branched-chain amino acids and unsaturated fatty acids.

INTRODUCTION: In recent years, nonalcoholic fatty liver disease (NAFLD) has become the most common chronic liver disease globally. Studies indicate that the gut-liver axis plays an important role in the occurrence and development of this disease. Our previous studies showed that the gut microbiota and gut metabolites in mice with NAFLD changed significantly. However, it is unclear whether these changes influenced the disease process through serum metabolites. METHODS: We conducted a non-targeted metabolome analysis on serum metabolites and systematically investigated the correlations between

Raffinose esters of long-chain unsaturated fatty acids: Preparation, characterization, and in vitro digestive behavior.

This study synthesized raffinose monoesters with polyunsaturated fatty acids (PUFA) including oleic acid, linolenic acid, and eicosapentaenoic acid through chemical synthesis. The HLB values of monoesters ranged from15.2 to16.7. Emulsions stabilized by monoesters exhibited exceptional stability over a 16-day period (50-90 °C), with the average diameter varying by less than ±30 nm. The PV values of three monoesters changed by less than 0.07. During in vitro simulated digestive model, the FA release rate exceeded 72.2 % in emulsions. However, the monoesters with lower chain length, higher satura

Regulation of HDL dysfunctionality by phosphatidylethanolamine links poly-unsaturated fatty acids with atherosclerotic cardiovascular diseases.

AIMS: Low plasma high-density lipoprotein (HDL)-cholesterol levels are associated with increased risk of atherosclerotic cardiovascular disease (ASCVD), potentially reflecting impaired antiatherogenic HDL functions. These latter are strongly influenced by the HDL phospholipidome, which is frequently altered in ASCVD patients. Several studies reported that plasma levels of phosphatidylethanolamine (PE) species, particularly PE (36:5), were positively associated with ASCVD, but the underlying mechanisms remain unclear. Plasma PE (36:5) exists as eicosapentaenoic (EPA)-PE and arachidonic acid (AR

SLC27A4-mediated selective uptake of mono-unsaturated fatty acids promotes ferroptosis defense in hepatocellular carcinoma.

Aberrant lipid metabolism mediated by the selective transport of fatty acids plays vital roles in cancer initiation, progression, and therapeutic failure. However, the biological function and clinical significance of abnormal fatty acid transporters in human cancer remain unclear. In the present study, we reported that solute carrier family 27 member 4 (SLC27A4) is significantly overexpressed in 21 types of human cancer, especially in the fatty acids-enriched microenvironment surrounding hepatocellular carcinoma (HCC), breast cancer, and ovarian cancer. Upregulated SLC27A4 expression correlate

Structure-function relationships of antifungal monohydroxy unsaturated fatty acids (HUFA) of plant and bacterial origin.

This study investigated the relationships between the structures of hydroxy unsaturated fatty acids (HUFA) and their antifungal activities. Structurally diverse HUFA, including four monohydroxy-18:1 isomers, two monohydroxy 18:2 isomers and two monohydroxy 18:2 isomers were extracted from seeds of plants (Coriaria nepalensis, Thymus vulgaris, Mallotus philippensis and Dimorphotheca sinuata) for which information was available on PlantFAdb database, and from culture supernatants of lactobacilli. They were purified by high-speed counter current chromatography (HSCCC) and identified by LC-MS/MS.

Differential effects of unsaturated fatty acids on phospholipid synthesis in human leukemia monocytic U937 cells.

The biosynthesis of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) in monocyte-like leukemia U937 cells was monitored by adding [3H]choline, [14C]ethanolamine or [14C]glycerol to the culture media; incorporation into phospholipid (PL) increased with time. The effect of unsaturated fatty acids (UFA) on PC and PE synthesis was investigated by pretreating U937 cells for 72h with 10 microM 18:1 (n - 9), 18:2 (n - 6), 18:3 (n - 3), 20:4 (n - 6) and 20:5 (n - 3). The UFA caused no alteration in cell growth, as evidenced by light microscopy and the incorporation of [3H]thymidine and [3H]l

The effects of unsaturated fatty acids on hepatic microsomal drug metabolism and cytochrome P-450.

1. The effects of unsaturated fatty acids on drug-metabolizing enzymes in vitro were measured by using rat and rabbit hepatic 9000g supernatant fractions. 2. Unsaturated fatty acids inhibited the hepatic microsomal metabolism of ;type I' drugs with inhibition increasing with unsaturation: arachidonic acid>linolenic acid>linoleic acid>oleic acid. Inhibition was independent of lipid peroxidation. Linoleic acid competitively inhibited the microsomal O-demethylation of p-nitroanisole and the N-demethylation of (+)-benzphetamine. 3. The hepatic microsomal metabolism of ;type II' substrates, aniline

Co-regulation of hepatic steatosis by ferritinophagy and unsaturated fatty acid supply.

Both iron overload and iron deficiency have been reported in obesity and metabolic syndromes. Due to the presence of multiple intracellular iron pools and the dynamic nature of iron mobilization and use, the actual status and contribution of free and metabolically active iron toward metabolic syndrome remain to be established. The discovery of nuclear receptor coactivator 4 (NCOA4) as a ferritinophagy receptor provides an opening to address the connection between iron and metabolic diseases. This study aims to specifically dissect the role of hepatic ferritinophagy in lipid metabolism and hepa

Exploring unsaturated fatty acid cholesteryl esters as transdermal permeation enhancers.

The intrinsic protective barrier property of skin, one of the major challenges in the design of transdermal drug delivery systems, can be overcome through the use of chemical permeation enhancers (CPEs). Herein, we explore the potential of unsaturated fatty acid (UFA) esters of cholesterol (Chol) viz., oleate, linoleate and linolenate, as transdermal CPEs using tenofovir (TNF) as a model drug. All Chol UFA esters at 1% w/w were found to be more effective enhancers when compared to their respective parent fatty acids (FAs) and saturated FA counterparts. Cholesteryl linolenate (Chol-LLA) showed

Ultrahigh magnetic field selectively promotes starch-fatty acid complexation depending on crystal type and fatty acid unsaturation.

Ultrahigh magnetic field (UMF) processing can modulate the properties of starch and alter starch crystallization. However, the effect of UMF in the formation of starch-fatty acid complexes remains limited, particularly regarding systematic comparisons across different crystalline starch and varied fatty acids. Using A-type wheat starch, B-type potato starch, saturated palmitic acid, and unsaturated palmitoleic acid as model systems, this study examined the effects of UMF treatment at two distinct stages (pretreatment and post-treatment) on starch-fatty acid complexation. UMF treatment at diffe

Unsaturated fatty acids as immunometabolic modulators in high-risk pregnancies: mechanisms, clinical evidence, and translational perspectives.

High-risk pregnancies (HRPs) are characterized by the convergence of metabolic dysfunction, chronic low-grade inflammation, and placental maladaptation, leading to increased maternal and perinatal morbidity and mortality. Emerging evidence indicates that these conditions are driven by a tightly integrated immunometabolic axis, in which metabolic stress activates innate immune pathways, amplifying systemic and placental inflammation. Unsaturated fatty acids (UFAs), including monounsaturated and polyunsaturated fatty acids (PUFAs), have gained attention as key modulators of this axis. Omega-3 PU

Long-chain unsaturated fatty acids are involved in the viability and itraconazole susceptibility of Aspergillus fumigatus.

The prevalence of invasive aspergillosis with azole resistance is increasing, but the mechanisms underlying the development of resistance and treatment strategies are still limited. The present work is focused on finding a relationship between long-chain unsaturated fatty acids (LCUFAs), Aspergillus fumigatus development, and antifungal resistance. The effects of LCUFAs on antifungal agents in vitro were determined, and the stearic acid desaturase gene (sdeA) of A. fumigatus was characterized. In in vitro antifungal tests, LCUFAs antagonized the antifungal activity of itraconazole by extractin

Soybean Oil-Derived Poly-Unsaturated Fatty Acids Enhance Liver Damage in NAFLD Induced by Dietary Cholesterol.

While the impact of dietary cholesterol on the progression of atherosclerosis has probably been overestimated, increasing evidence suggests that dietary cholesterol might favor the transition from blunt steatosis to non-alcoholic steatohepatitis (NASH), especially in combination with high fat diets. It is poorly understood how cholesterol alone or in combination with other dietary lipid components contributes to the development of lipotoxicity. The current study demonstrated that liver damage caused by dietary cholesterol in mice was strongly enhanced by a high fat diet containing soybean oil-

Saturated and Unsaturated Fatty Acids Differently Modulate Colonic Goblet Cells In Vitro and in Rat Pups.

BACKGROUND: High-fat diets induce intestinal barrier alterations and promote intestinal diseases. Little is known about the effects of long-chain fatty acids (LCFAs) on mucin 2 (MUC2) production by goblet cells, which are crucial for intestinal protection. OBJECTIVE: We investigated the effects of LCFAs on the differentiation of colonic goblet cells, MUC2 expression, and colonic barrier function. METHODS: Upon reaching confluence, human colonic mucus-secreting HT29-MTX cells were stimulated (21 d) with a saturated LCFA (palmitic or stearic acid), a monounsaturated LCFA (oleic acid), or a pol
