Hepatic flavin-containing monooxygenase 3 (FMO3) is arguably the most important FMO in humans from the standpoint of drug metabolism. Recently, adult hepatic FMO3 has been linked to several conditions including cardiometabolic diseases, aging, obesity, and atherosclerosis in small animals. Despite the importance of FMO3 in drug and chemical metabolism, relative to cytochrome P-450 (CYP), fewer studies have been published describing drug and chemical metabolism. This may be due to the properties of human hepatic FMO3. For example, FMO3 is thermally labile, and often methods reported in the stud

Catalytic desaturations are important strategies for the functionalization of organic molecules. In nature, flavoenzymes mediate the formation of α,β-unsaturated carbonyl compounds by concomitant cofactor reduction. Contrary to many laboratory methods for these reactions, such as the Saegusa-Ito oxidation, no transition metal reagents or catalysts are required. However, a molecular flavin-mediated variant has not been reported so far. We disclose a photochemical approach for silyl enol ether oxidation, which leads to α,β-unsaturated ketones (13 examples) in very good yields. The flavin catalys

Rifamycins have been clinically utilized against mycobacterial infections for more than 50 years; however, their biosynthesis has not been fully elucidated. Here, on the basis of in vivo gene deletions, in vitro enzyme assays, isotope labeling, and site-directed mutations, we found that a flavin-dependent monooxygenase encoded by a rifamycin biosynthetic gene cluster, Rif-Orf17, not only converted the naphthoquinone chromophore of rifamycin S into benzo-γ-pyrone but also linearized rifamycin SV through phenolic hydroxylation. Both oxidation routes lead to inactivation of rifamycins.

The incorporation of radiolabeled riboflavin into flavin mononucleotide, flavin adenine dinucleotide, and flavin covalently bound to protein was determined in Novikoff hepatoma grown in both riboflavin-deficient and normal chow-fed rats. In Novikoff hepatoma, the incorporation of [14C]riboflavin into covalently bound flavins relative to that into FAD was substantially greater than that in host liver, and the turnover rate of riboflavin was also accelerated in tumor compared with the liver. The magnitude of incorporation of [14C]riboflavin into each of the various flavin fractions was substanti…

A novel flavin-catalyzed chemoselective aerobic oxygenation of arylheteroarylmethanes to ketones has been developed under metal-free and mild conditions. This catalytic system employs a cationic flavin to activate sp3 C-H bonds through the formation of pyridinium-flavin adducts, enabling precise proton and electron transfer for oxygen atom insertion. As a result, this method exhibits high chemoselectivity, effectively distinguishing between arylheteroarylmethanes and diarylmethanes.

Flavin-dependent halogenases allow halogenation of electron-rich aromatic compounds under mild reaction conditions even at electronically unfavored positions with high regioselectivity. In order to expand the application of halogenases, the enzymes need to be improved in terms of stability and efficiency. A previous study with the tryptophan 6-halogenase Thal demonstrated that thermostable Thal variants tend to form dimers in solution while the wild type is present as a monomer. Based on this a dimeric Thal variant was generated that is covalently linked by disulfide bonds. Introducing two cys

Chlorine gas or electropositive chlorine reagents are used to prepare chlorinated aromatic compounds, which are found in pharmaceuticals, agrochemicals, and polymers, and serve as synthetic precursors for metal-catalyzed cross-couplings. Nature chlorinates with chloride anions, FAD-dependent halogenases, and O2 as the oxidant. A photocatalytic oxidative chlorination is described based on the organic dye riboflavin tetraacetate mimicking the enzymatic process. The chemical process allows within the suitable arene redox potential window a broader substrate scope compared to the specific activati

The primary role of the water-soluble vitamin B2, i.e. riboflavin, in cell biology is connected with its conversion into FMN and FAD, the cofactors of a large number of dehydrogenases, reductases and oxidases involved in energetic metabolism, redox homeostasis and protein folding as well as in diverse regulatory events. Deficiency of riboflavin in men and experimental animal models has been linked to several diseases, including neuromuscular and neurological disorders and cancer. Riboflavin at pharmacological doses has been shown to play unexpected and incompletely understood regulatory roles.

The success of solar fuel technology relies on the development of efficient catalysts that can oxidize or reduce water. All molecular water-oxidation catalysts reported thus far are transition-metal complexes, however, here we report catalytic water oxidation to give oxygen by a fully organic compound, the N(5)-ethylflavinium ion, Et-Fl(+). Evolution of oxygen was detected during bulk electrolysis of aqueous Et-Fl(+) solutions at several potentials above +1.9 V versus normal hydrogen electrode. The catalysis was found to occur on glassy carbon and platinum working electrodes, but no catalysis

BACKGROUND: Friedreich ataxia is a neurodegenerative disease caused by the lack of frataxin, a mitochondrial protein. We previously demonstrated that frataxin interacts with complex II subunits of the electronic transport chain (ETC) and putative electronic transfer flavoproteins, suggesting that frataxin could participate in the oxidative phosphorylation.
METHODS AND FINDINGS: Here we have investigated the effect of riboflavin and its cofactors flavin adenine dinucleotide (FAD) and flavin mononucleotide (FMN) in Saccharomyces cerevisiae and Caenorhabditis elegans models of frataxin deficienc

Reduction rate constants of the chlorinated ethylenes cis-1,2-dichloroethylene (cis-DCE), trichloroethylene (TCE), and tetrachloroethylene (PCE) reacted with flavin mononucleotide hydroquinone (FMNH2) under anoxic conditions were investigated. FMNH2 was produced in methanol solvent by the photoreduction of FMN. In aqueous solution, FMN was not fully reduced to FMNH2 but instead yielded the semiquinone radical FMNH*. However, when FMN was anchored to nanocrystalline TiO2, band gap irradiation resulted in electron transfer from the TiO2 conduction band to FMN, thus yielding FMNH2. The FMNH2 gene

The in vitro effects of flavin nucleotides on human erythrocyte glutathione reductase were studied. Flavin mononucleotide was found to be a potent inhibitor of glutathione reductase in haemolysates, having an effect at a concentration of 1 muM. Flavin adenine dinucleotide inhibited at a concentration at least ten fold this, lower levels producing the expected stimulation of enzyme activity. Different assay conditions also altered the activity of the enzyme, although the flavin mononucleotide inhibition remained. The implications of these findings in our understanding of glutatione reductase an

When cells are exposed to external H(2)O(2), the H(2)O(2) rapidly diffuses inside and oxidizes ferrous iron, thereby forming hydroxyl radica … tration, which explains why its flux increased when NADH levels rose during respiratory inhibition. Flavins that were reduced by purified flavin reductase rapidly transferred electrons to free iron and drove a DNA-damaging Fenton system in vitro. Thus the rate of oxidative DNA damage can be limited by the rate at which electron donors reduce free iron, an …

The search for more sustainable catalytic methods, such as biocatalysis and photocatalysis, has greatly contributed to a sustainable future of synthetic chemistry. Within photocatalysis, flavin-type heterocycles are powerful tools for oxidative transformations but often suffer from poor water solubility, photostability, and limited reusability. Here, we provide a proof-of-concept strategy to bridge the gap between photocatalysis and biocatalysis through the development of artificial flavin biomolecules. A 7-/8-carboxyflavin derivative was synthesized and covalently attached to protein surfaces

Studies from the 1980s and 1990s conducted in Italy, where malaria was once endemic, hypothesized that individuals with erythrocytes deficient in flavin mononucleotide and flavin adenine dinucleotide (FAD)-collectively known as flavins-are partially protected against malaria. The condition was reported to be familial, consistent with a genetic element. This hypothesis, however, has never been tested. Using an erythrocyte FAD-dependent glutathione reductase activity assay, we identified individuals with flavin-deficient erythrocytes (FDE) in Ferrara, Italy (23% of 150 individuals screened), and…
