Frozen section of lung specimens.

CONTEXT: Frozen section of lung tissue is performed to guide the surgeon in subsequent therapy. DESIGN: Practical experience in frozen section of the lung was reviewed in the medical literature and from the records of several academic hospitals. RESULTS: Most frozen sections of the lung are performed for evaluation of a solitary nodule, a mass, or the surgical margins of a resection. Frozen section may also be used to assess the adequacy of a lung wedge biopsy taken for later diagnosis of a condition. CONCLUSION: The pathologic evaluation of intraoperative pulmonary lesions is indicated for

A Retrospective Study to Assess the Accuracy of Frozen Sections in Laser Laryngeal Surgery.

OBJECTIVE: The basis of good vocal outcomes following Transoral Laser Microlaryngeal Cordectomy (TLMC) is the narrow margin that is oncologically accepted for the glottis. Our objective is to evaluate the reliability of frozen section (FS) compared to paraffin section (PS) during TLMC and during laser laryngeal surgery when an incisonal or excisional biopsy is being performed. METHODS: Retrospectively, records of 159 sequential patients who underwent CO2 laser laryngeal surgery with intraoperative FS were reviewed along with the final PS. Group A patients were TLMC patients where FS was utili

A generative adversarial approach to facilitate archival-quality histopathologic diagnoses from frozen tissue sections.

In clinical diagnostics and research involving histopathology, formalin-fixed paraffin-embedded (FFPE) tissue is almost universally favored for its superb image quality. However, tissue processing time (>24 h) can slow decision-making. In contrast, fresh frozen (FF) processing (<1 h) can yield rapid information but diagnostic accuracy is suboptimal due to lack of clearing, morphologic deformation and more frequent artifacts. Here, we bridge this gap using artificial intelligence. We synthesize FFPE-like images ("virtual FFPE") from FF images using a generative adversarial network (GAN) from 98

Utility of intraoperative frozen sections in surgical decision making for acute invasive fungal rhinosinusitis.

BACKGROUND: Acute invasive fungal rhinosinusitis (AIFRS) represents a fulminant, potentially fatal, disease process in immunocompromised patients. The diagnosis often rests on high index of clinical suspicion, with relative paucity of data on the diagnostic and therapeutic implications of intraoperative frozen sections. METHODS: Retrospective review was performed for 18 cases undergoing endoscopic sinus surgery for AIFRS. Reliability of intraoperative frozen section diagnosis was evaluated for all patients using final pathology as the gold standard. RESULTS: A total of 66 frozen sections wer

The reliability of mediastinoscopic frozen sections in deciding on oncological surgery in bronchogenic carcinoma.

INTRODUCTION: This study was carried out to determine the accuracy of mediastinoscopic frozen section examination, performed prior to major surgery-especially where mediastinal lymph node metastasis (N2 disease) was suspected. We aimed to find out whether or not mediastinoscopic frozen section analysis was (i) a reliable tool when deciding to continue resection in lung cancer patients and (ii) reliable in diagnosing mediastinal masses. METHODS: One-hundred and thirty-six patients undergoing mediastinoscopy were enrolled in this study. Resection was planned for each case, and biopsies were tak

Use of frozen sections for the pharmacological characterization of compounds active on neurotransmitter receptors.

Radioligand binding assay represents an important technique in pharmacological and pharmaceutical research for assessing the receptor profile of new drugs or of compounds under development. In this study, the pharmacological profile and the receptor specificity of compounds active on dopamine and muscarinic cholinergic receptor subtypes were evaluated using as a receptor source, membrane preparations or frozen sections. Dopamine D1-like receptors were assayed in membrane preparations or frozen sections of rat striatum and kidney with [3H]-SCH 23390 as a ligand. Rat striatum, kidney and atrium

Immunoarchitecture of normal human bone marrow: a study of frozen and fixed tissue sections.

To provide baseline information on the immunoarchitecture of normal bone marrow, we studied cryostat-cut, frozen, and paraffin-embedded, fixed tissue sections prepared from 21 core biopsies of normal bone marrow obtained during bone marrow harvests for transplantation. A large panel of antibodies was applied that included, for frozen tissue, Leu-6 (CD1), T11 (CD2), Leu-3a (CD4), Leu-1 (CD5), Leu-2a (CD8), J5 (CD10), My7 (CD13), Leu-11 (CD16), B4 (CD19), B1 (CD20), B2 (CD21), Tac (CD25), My9 (CD33), T200 (CD45), NKH-1 (CD56), kappa and lambda chains, beta F1, Ki-67, HLA-DR, TQ1, and keratin, an

[Diagnosis of neurocysticercosis by immunoenzymatic staining technique (IEST) with frozen sections of Cysticercus cellulosae].

This article deals with the sensitivity and specificity of IEST in neurocysticercosis with frozen sections of Cysticerus cellulosae as antigens. 55 out of 56 cases (98.2%) of neurocysticercosis showed positive reaction. All serum specimens from healthy human controls (50 cases), clonorchiasis (23 cases) and paragonimiasis (26 cases) were negative, but 18% (4/22) of serum specimens from echinococcosis revealed positive reactions. The results indicate that IEST is specific, sensitive, economical and easy to perform for the diagnosis of neurocysticercosis.

Peroperative evaluation of parathyroid glands using fat stain on frozen sections. Advantages and limitations.

The possible benefit of fat stain in the diagnosis of parathyroid disease was elucidated in this prospective study comprising 51 patients operated on for primary hyperparathyroidism. The histo-pathological diagnosis was adenoma in 39 patients and hyperplasia in 9 patients. In three patients only normal glands were examined. The majority of adenomas contained very small amounts of intracellular fat, while the rim of normal parathyroid adjacent to the adenoma and additional normal glands examined had abundant intracellular fat. In two cases was the adenoma diagnosis arrived at due to the examina

Foundation Model-Based Computational Pathology Predicts Breast Cancer Biomarker Status from Intraoperative Frozen Sections.

Prediction of breast cancer biomarkers from routine formalin-fixed, paraffin-embedded (FFPE) hematoxylin and eosin (H&E) images has been explored previously, but whether biomarker-associated morphologic signals can be reliably captured from artifact-prone intraoperative frozen-section (FS) specimens remains insufficiently studied. This retrospective proof-of-concept study developed a foundation model-based multiple-instance learning framework to predict estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2), and Ki-67 status directly from H&E-staine

Are frozen sections useful and cost-effective in the era of intraoperative qPTH assays?

BACKGROUND: Since intraoperative quick parathormone (IOqPTH) assays are available, the role of frozen sections (FS) during parathyroid exploration has become questionable. This study compares the results of FS and IOqPTH in primary hyperparathyroidism (pHPT). METHODS: FS and IOqPTH assays were performed in 102 patients who underwent bilateral neck explorations or targeted parathyroidectomy for pHPT. The operation was considered complete when both an IOqPTH drop >50% and a FS diagnosis of parathyroid adenoma were obtained. RESULTS: Cure was achieved in all patients. Potential pitfalls for suc

Translation of frozen sections into FFPE images for skin cancer resection margins using generative AI.

Frozen section (Frozen) analysis is essential for intraoperative margin assessment in skin cancer surgery, but assessment is limited by freezing artifacts that obscure cellular detail. Formalin-fixed paraffin-embedded (FFPE) histopathology, the gold standard, provides higher quality but requires >24 h for processing. Herein, we developed and validated generative AI models to translate Frozen images into AI-generated FFPE images (GenFFPE) using 2594 slides from 283 cases across five major skin cancer types. Four unpaired image-to-image models (CycleGAN, CUT, AIFFPE, and SANTA) were trained and

Optimization of electron microscopy for human brains with long-term fixation and fixed-frozen sections.

BACKGROUND: Abnormal connectivity across brain regions underlies many neurological disorders including multiple sclerosis, schizophrenia and autism, possibly due to atypical axonal organization within white matter. Attempts at investigating axonal organization on post-mortem human brains have been hindered by the availability of high-quality, morphologically preserved tissue, particularly for neurodevelopmental disorders such as autism. Brains are generally stored in a fixative for long periods of time (often greater than 10 years) and in many cases, already frozen and sectioned on a microtome

Calcium uptake in frozen muscle biopsy sections compared with other predictors of malignant hyperthermia susceptibility.

The frozen-section Ca45 uptake procedure of Mabuchi and Sreter has been applied to 51 frozen human muscle biopsies previously tested for halothane and for caffeine contracture responses. In preliminary tests with cat muscle, uptake varied with skeletal muscle fiber type, was absent from smooth muscle, and was blocked by agents which damage or extract sarcoplasmic reticulum. In the human biopsies, however, the calcium uptake values showed no significant correlation with any of the following parameters: time of frozen-biopsy storage up to 5 yr; fiber type distribution; positive versus negative r

Mohs micrographic surgery for dermatofibrosarcoma protuberans: our experience using frozen sections and a literature review.

INTRODUCTION AND OBJECTIVES: Mohs micrographic surgery (MMS) is considered the gold standard for the treatment of dermatofibrosarcoma protuberans (DFSP). Although paraffin-based MMS is the most widely accepted technique, no study has demonstrated its superiority over frozen-section MMS. We present our experience with frozen-section MMS for DFSP and review the literature. MATERIALS AND METHODS: We conducted a retrospective, observational, single-center study of all patients with DFSP treated with frozen-section MMS between July 2014 and July 2023. Demographic and tumor characteristics, surgica
