Gluconeogenesis from alanine in patients with progressive malignant disease.

We have studied by tracer technique the conversion of the carbon skeleton of alanine to glucose in patients with progressive malignant disea … rong dependence for carbohydrate and protein metabolism is suggested. We conclude that uncontrolled gluconeogenesis from alanine is probably not significant in terms of energy expenditure in the patient with uncontrolled cancer.

Intestinal gluconeogenesis is downregulated in pediatric patients with celiac disease.

BACKGROUND: Untreated celiac disease (CD) patients have increased levels of blood glutamine and a lower duodenal expression of glutaminase (GLS). Intestinal gluconeogenesis (IGN) is a process through which glutamine is turned into glucose in the small intestine, for which GLS is crucial. Animal studies suggest impaired IGN may have long-term effects on metabolic control and be associated with the development of type 2 diabetes and non-alcoholic fatty liver disease (NAFLD). The aim of this study was to thoroughly investigate IGN at the gene expression level in children with untreated celiac dis…

Gluconeogenesis is essential for trypanosome development in the tsetse fly vector.

In the glucose-free environment that is the midgut of the tsetse fly vector, the procyclic form of Trypanosoma brucei primarily uses proline to feed its central carbon and energy metabolism. In these conditions, the parasite needs to produce glucose 6-phosphate (G6P) through gluconeogenesis from metabolism of non-glycolytic carbon source(s). We showed here that two phosphoenolpyruvate-producing enzymes, PEP carboxykinase (PEPCK) and pyruvate phosphate dikinase (PPDK) have a redundant function for the essential gluconeogenesis from proline. Indeed, incorporation of 13C-enriched proline into G6P…

Metabolic regulation of renal gluconeogenesis in response to sepsis in the rat.

1. The regulation of renal gluconeogenesis was studied in rats made septic by a caecal ligation and puncture technique. 2. Blood glucose concentrations were not markedly different in septic rats, but lactate, pyruvate and alanine concentrations were markedly increased, compared with sham-operated rats. Conversely, blood ketone body concentrations were significantly decreased in septic rats. Both plasma insulin and glucagon concentrations were markedly elevated in response to sepsis. 3. The maximal activities of glucose-6-phosphatase (EC 3.1.3.9), fructose-1,6-bisphosphatase (EC 3.1.3.11), pyru…

Sip4p Regulates the Transcription of Enzymes for Gluconeogenesis in Nakaseomyces glabratus.

To survive in glucose poor niches within the host, the opportunistic pathogen Nakaseomyces glabratus must assimilate alternative carbon sour … cur upon treatment with lactate or ethanol using RNA-seq. In the wild type strain, genes related to gluconeogenesis, the TCA cycle, and the glyoxylate cycle were upregulated in the presence of alternative carbon sources, and transcription of PCK1, encoding the gluconeogenesis enzyme phosphoenolpyruvate carboxykinase, was induced over 2000-fold. Deletion of SIP4 r …

Knockdown of ACOT4 alleviates gluconeogenesis and lipid accumulation in hepatocytes.

Acyl-CoA thioesterase 4 (ACOT4) has been reported to be related to acetyl-CoA carboxylase activity regulation; However, its exact functions in liver lipid and glucose metabolism are still unclear. Here, we discovered explored the regulatory roles of ACOT4 in hepatic lipid and glucose metabolism in vitro. We found that the expression level of ACOT4 was significantly increased in the hepatic of db/db and ob/ob mice as well as obese mice fed a high fat diet. Adenovirus-mediated overexpression of ACOT4 promoted gluconeogenesis and high-glucose/high-insulin-induced lipid accumulation and impaired i…

Gluconeogenesis: Adding a new piece to the puzzle of malaria parasitism.

Disorders of carbohydrate metabolism, including hypoglycemia and lactic acidosis, are common features of malaria. In this issue of Cell Metabolism, Ramos et al. report that regulation of gluconeogenesis and glycemia by the host glucose-6-phosphatase catalytic subunit 1 (G6Pc1) is a key metabolic step that affects both Plasmodium replication and clinical outcome of disease.

Insulin and glucagon regulation of gluconeogenesis in preruminating and ruminating bovine.

Hepatocyte monolayers were established from young preruminating (7 to 14 d of age) or older ruminating (11 to 12 wk of age) calves and used to evaluate the effects of insulin and glucagon on incorporation of carbon from 2.5 mM [2-14C]propionate and 2.0 mM [U-14C]lactate into glucose and glycogen. Developmental state (young preruminating vs older ruminating) of the donor calf did not affect the rate of gluconeogenesis from propionate in the absence of hormones. Insulin decreased (P < .05) gluconeogenesis and increased (P < .05) glycogenesis from propionate and lactate in hepatocytes from prerum…

[Regulation of gluconeogenesis in the liver of vitamin B1-deficient rats].

Radiometric assays revealed that thiamine deficiency in rats to whom hydroxythiamine was administered in variable doses, is concomitant with activation of gluconeogenesis from pyruvate in liver tissue. The most probable mechanism of this effect is the cAMP-dependent activation of key enzymes of intracellular glucose synthesis. This process is facilitated by the diminution of the ratio of free forms of NAD+ and NADPH in the cytoplasm.

The influence of branched chain aminoacids and their ketoderivatives on renal gluconeogenesis.

Isolated kidney tubules served as a model to investigate the direct effect of branched chain aminoacids and their ketoderivatives on gluconeogenesis. The data presented in this paper demonstrate that the ketoderivatives rather than the branched chain aminoacids themselves inhibit renal glucosesynthesis from various precursors entering the glucogenic pathway at different levels. Though the point of the inhibitory attack of ketoacids could not be localized, an impairement of the kidney cortex to respond to metabolic acidosis with an increase of gluconeogenesis is evident from the present data. T…

Gluconeogenesis related gene signatures as biomarkers for nonspecific orbital inflammation.

Nonspecific orbital inflammation (NSOI) is a chronic, idiopathic inflammatory disorder marked by persistent tissue proliferation and immune activation. Gluconeogenesis, a core metabolic pathway enabling endogenous glucose production in the absence of dietary input, plays a vital role in cellular energy homeostasis and immune regulation. Metabolic rewiring of immune cells is not merely a consequence of inflammation but a critical determinant of disease trajectory in orbital disorders. Within this conceptual landscape, the exploration of gluconeogenesis represents a logical and timely extension,…

Metformin inhibits gluconeogenesis via a redox-dependent mechanism in vivo.

Metformin, the universal first-line treatment for type 2 diabetes, exerts its therapeutic glucose-lowering effects by inhibiting hepatic gluconeogenesis. However, the primary molecular mechanism of this biguanide remains unclear, though it has been suggested to act, at least partially, by mitochondrial complex I inhibition. Here we show that clinically relevant concentrations of plasma metformin achieved by acute intravenous, acute intraportal or chronic oral administration in awake normal and diabetic rats inhibit gluconeogenesis from lactate and glycerol but not from pyruvate and alanine, im…

Evidence that thyroid hormones regulate gluconeogenesis from glycerol in man.

We have previously reported that glucose production assessed using radioisotopic methods is 50% increased in hyperthyroidism but 30% decreased in hypothyroidism. These studies, however, do not distinguish between glycogenolysis and gluconeogenesis. In fasting man more than 80% of circulating glycerol is cleared by the liver and enters the gluconeogenic pathway. We have therefore measured glycerol clearance following bolus intravenous glycerol administration as an indirect assessment of gluconeogenic capacity. Hyperthyroid and hypothyroid subjects were compared with separate matched controls af…

Renal gluconeogenesis: a key metabolic hub in health and kidney disease.

Gluconeogenesis, responsible for maintaining systemic glucose availability during fasting, has long been thought to be a process mainly performed by the liver. Over the past decades, physiological and molecular studies have established the kidney as a major site of endogenous glucose production. Renal gluconeogenesis occurs mainly in proximal tubular epithelial cells, where it integrates substrate utilization, hormonal signaling, and acid-base regulation to support systemic homeostasis. The kidney relies on distinct regulatory inputs and dynamically adapts gluconeogenic flux to nutritional sta…

Leptin Decreases Gluconeogenesis and Gluconeogenic Substrate Availability in Patients With Lipodystrophy.

CONTEXT: The effects of leptin, an adipocyte-derived hormone that signals overall energy sufficiency, can only be studied in leptin-deficient conditions. In patients with lipodystrophy, a rare disease and unique model of leptin deficiency, treatment with recombinant leptin (metreleptin) improves glycemia and decreases energy expenditure. We hypothesized that these improvements might be mediated by reduced gluconeogenesis (GNG), an energy-requiring process. OBJECTIVE: To determine the effects of metreleptin on GNG and GNG substrates. METHODS: This was a single-arm prospective study of metrele…

Renal gluconeogenesis: an underestimated role of the kidney in systemic glucose metabolism.

Glucose levels are tightly regulated at all times. Gluconeogenesis is the metabolic pathway dedicated to glucose synthesis from non-hexose precursors. Gluconeogenesis is critical for glucose homoeostasis, particularly during fasting or stress conditions. The renal contribution to systemic gluconeogenesis is increasingly recognized. During the post-absorptive phase, the kidney accounts for ∼40% of endogenous gluconeogenesis, occurring mainly in the kidney proximal tubule. The main substrate for renal gluconeogenesis is lactate and the process is regulated by insulin and cellular glucose levels,…

Energy expenditure due to gluconeogenesis in pathological conditions of insulin resistance.

Gluconeogenesis (GNG), the formation of glucose from noncarbohydrate precursors, requires adenosine triphosphate (ATP). Previous studies have estimated the energetic cost of GNG in humans based on theoretical calculations of rates of GNG, moles of oxygen consumption by GNG, and average oxygen consumption. Few human studies have measured the energy expenditure (EE) due to GNG. We estimated EE attributable to GNG in patients with three insulin resistance conditions and high GNG rates (insulin receptor pathogenic variants, lipodystrophy, and type 2 diabetes) and obesity without diabetes. Fraction…
