Hippocampus and remote spatial memory in rats.

Damage to the hippocampus typically produces temporally graded retrograde amnesia, whereby memories acquired recently are impaired more than memories acquired remotely. This phenomenon has been demonstrated repeatedly in a variety of species and tasks. It has also figured prominently in theoretical treatments of memory and hippocampal function. Yet temporally graded retrograde amnesia has not been demonstrated following hippocampal damage in spatial tasks like the water maze. We have assessed recent and remote spatial memory following hippocampal lesions in three different tests of spatial mem…

Contextual fear, gestalt memories, and the hippocampus.

This review examines the relationship between exploration and contextual fear conditioning. The fear acquired to places or contexts associated with aversive events is a form of Pavlovian conditioning. However, an initial period of exploration is necessary to allow the animal to form an integrated memory of the features of the context before conditioning can take place. The hippocampal formation plays a critical role in this process. Cells within the dorsal hippocampus are involved in the formation, storage and consolidation of this integrated representation of context. Projections from the sub…

Involvement of hippocampus in Creutzfeldt-Jakob disease.

The hippocampus is generally believed to be only rarely affected in Creutzfeldt-Jakob disease. In a systematic study of the hippocampus in 6 cases of Creutzfeldt-Jakob disease, the stratum moleculare-lacunosum showed a definite spongiform change with gliosis in 5 cases. The stratum radiatum et pyramidale showed spongiform change in 3 of the 5 cases. The subiculum, presubiculum, parasubiculum and entorhinal cortex were involved in all the cases. These results suggest that involvement of the hippocampus may not be uncommon in Creutzfeldt-Jakob disease. The stratum moleculare-lacunosum seems to b…

Mental Scene and Memory Construction and the Hippocampus.

A neurocomputational theory of scene and memory construction during memory recall, prospective thinking, and autobiographical memory is desc … ct or person at that location in the scene ("what"), and the reward/affective value conveyed to the hippocampus from the orbitofrontal cortex in part via the ventromedial prefrontal cortex and posterior cingulate cortex. The theory of scene construction then shows how backprojections from hippocampal CA1 via the entorhinal cortex to the medial parahippocampal …

Structural Plasticity of the Hippocampus in Neurodegenerative Diseases.

Neuroplasticity is the capacity of neural networks in the brain to alter through development and rearrangement. It can be classified as structural and functional plasticity. The hippocampus is more susceptible to neuroplasticity as compared to other brain regions. Structural modifications in the hippocampus underpin several neurodegenerative diseases that exhibit cognitive and emotional dysregulation. This article reviews the findings of several preclinical and clinical studies about the role of structural plasticity in the hippocampus in neurodegenerative diseases, including Alzheimer's disea…

A Dedicated Population for Reward Coding in the Hippocampus.

The hippocampus plays a critical role in goal-directed navigation. Across different environments, however, hippocampal maps are randomized, making it unclear how goal locations could be encoded consistently. To address this question, we developed a virtual reality task with shifting reward contingencies to distinguish place versus reward encoding. In mice performing the task, large-scale recordings in CA1 and subiculum revealed a small, specialized cell population that was only active near reward yet whose activity could not be explained by sensory cues or stereotyped reward anticipation behav…

Update on Cysticercosis Epileptogenesis: the Role of the Hippocampus.

Neurocysticercosis (NCC) is the most common helminthic infection of the nervous system and a frequent cause of reactive seizures and epilepsy worldwide. In many cases, multiple episodes of focal seizures related to an identifiable parenchymal brain cyst (and likely attributable to local damage) continue for years after the cyst resolves. However, cases where seizure semiology, interictal EEG abnormalities, and parasites location do not correlate raise concerns about the causal relationship between NCC and either reactive seizures or epilepsy, as well as the epileptogenic potential of parasites

Hippocampus and amygdala volumes in patients with vaginismus.

AIM: To compare hippocampus and amygdala volumes of patients with vaginismus with those of healthy control subjects. METHODS: Magnetic resonance imaging was performed on ten patients with vaginismus and ten control subjects matched for age and gender. Volumes of the hippocampus and amygdala were blindly measured. RESULTS: We found that the mean right amygdala volume of patients with vaginismus were smaller than that of the healthy controls. With regard to hippocampus volumes, the mean left and right hippocampus volumes were smaller than those of the healthy controls. CONCLUSION: Our present…

[Subtle focal diffusion abnormality in the hippocampus].

A 53-year-old female patient presented with sudden onset confusion and disorientation. Further neurological examination was unremarkable and the patient showed a complete recovery after several hours. A magnetic resonance imaging (MRI) examination performed 2 days later revealed a tiny focal lesion in the lateral hippocampus in the diffusion weighted images consistent with transient global amnesia.

Computational principles of learning in the neocortex and hippocampus.

We present an overview of our computational approach towards understanding the different contributions of the neocortex and hippocampus in learning and memory. The approach is based on a set of principles derived from converging biological, psychological, and computational constraints. The most central principles are that the neocortex employs a slow learning rate and overlapping distributed representations to extract the general statistical structure of the environment, while the hippocampus learns rapidly, using separated representations to encode the details of specific events while sufferi…

Is the hippocampus necessary for contextual fear conditioning?

The hippocampus is widely believed to be essential for learning about the context in which conditioning occurs. This view is based primarily on evidence that lesions of the dorsal hippocampus disrupt freezing to contextual cues after fear conditioning. However, lesions that disrupt freezing produce no effect on fear-potentiated startle, a second measure of contextual fear. Moreover, hippocampal lesions also do not disrupt the contextual 'blocking' phenomenon, which provides an indirect measure of contextual fear. In these paradigms, at least, it appears that hippocampal lesions disrupt the exp…

[Diagnosis of seizures originating in the amygdala and the hippocampus].

INTRODUCTION: Lesions in the hippocampus of some epileptic patients were first described one hundred and seventy years ago. Since then our knowledge of the relationship between epileptic seizures and temporal lobe lesions has greatly improved. DEVELOPMENT: The aim of this paper is to systematize the symptomatology of the hippocampus and amygdala seizures. These seizures may have a great number of different clinical features: special 'loss' or 'impairment of consciousness', epileptic automatisms and autonomic changes. Moreover, at the beginning of seizures some patients have reported experienc…

Episodic and declarative memory: role of the hippocampus.

The fact that medial temporal lobe structures, including the hippocampus, are critical for declarative memory is firmly established by now. The understanding of the role that these structures play in declarative memory, however, despite great efforts spent in the quest, has eluded investigators so far. Given the existing scenario, novel ideas that hold the promise of clarifying matters should be eagerly sought. One such idea was recently proposed by Vargha-Khadem and her colleagues (Science 1997; 277:376-380) on the basis of their study of three young people suffering from anterograde amnesia …

Possible mechanisms for atrophy of the human hippocampus.

Recently published work using MRI to image the human brain has revealed that the hippocampal formation undergoes a selective atrophy in diverse conditions such as Cushing's syndrome, post-traumatic stress disorder, recurrent depressive illness, normal aging preceding dementia and in Alzheimer's disease. Hippocampal shrinkage is usually accompanied by deficits in declarative, episodic, spatial and contextual memory performance and the hippocampal changes provide a neural substrate for changes in cognitive function that have been recognized to accompany these various conditions. The hippocampus …

Evidence for dopamine as a transmitter in dorsal hippocampus.

Intrahippocampal distribution of dopamine (DA) and its metabolites, homovanillic acid (HVA) and 3,4-dihydroxyphenylacetic acid (DOPAC), was studied along with those of noradrenaline (NA) and 5-hydroxytryptamine in rats. DA concentration in the dorsal part of the hippocampus was found to be 4--10 times higher than in other parts. A study of metabolite distributions produced similar results, showing a higher concentration in the dorsal part of the hippocampus. A ratio of NA/DA had a value similar to that of the hypothalamus in which apparent dopaminergic innervation has been reported. Haloperido…

Microinjection of kainic acid into the rat hippocampus.

The effects of unilateral injection of kainic acid into the rate hippocampus have been examined in terms of morphologic, neurochemical and behavioral sequelae. Infusion of 10 nmoles if kainate causes a rapid and complete degeneration of neuronal perikarya in the entire hippocampal formation followed by gliosis and atrophy of the region. This unilateral neuronal loss is accompanied by a 50% decrease in the specific activity of the biochemical markers for GABAergic neurons including glutamic acid decarboxylase, endogenous GABA and synaptosomal uptake of [3H]GABA. The extrinsic hippocampal cholin…

p63 Expression in the Gerbil Hippocampus Following Transient Ischemia and Effect of Ischemic Preconditioning on p63 Expression in the Ischemic Hippocampus.

p63 is a transcription factor of p53 gene family, which are involved in development, differentiation and cell response to stress; however, its roles in ischemic preconditioning (IPC) in the brain are not clear. In the present study, we investigated the effect of IPC on p63 immunoreactivity caused by 5 min of transient cerebral ischemia in gerbils. IPC was induced by subjecting the gerbils to 2 min of transie ischemia 1 day prior to 5 min of transient ischemia. The animals were randomly assigned to four groups (sham-operated-group, ischemia-operated-group, IPC plus (+)-sham-operated-group and I
