The physico-chemical properties of a series of hydantoin derivatives and their intestinal absorption from solution were studied. The introduction of the benzenesulfonyl group at the 1-position of the hydantoin ring greatly affects the physico-chemical properties: the acid dissociation constants were increased 1000-fold and the partition coefficients were increased 100- to 1000-fold in the chloroform/water system and 10- to 100-fold in the n-octanol/water system. The solubilities of 1-benzenesulfonylhydantoin derivatives increased with increasing pH of the solution at pH more than 5, but the so

We report an efficient synthetic strategy to diverse hydantoins and thiohydantoins involving a three-component reaction with the aid of deep eutectic solvent. Here, N,N'-dimethyl urea and N,N'-dimethyl thiourea play a dual role as reactant and reaction medium along with l-(+)-tartaric acid. The three-component reaction provides an easy access to 5-amino-1,3-dialkyl-substituted hydantoins and thiohydantoins in good yields.

Epilepsy is not a common cause of morbidity in pregnancy. It has widespread effects on maternal and fetal health necessitating adequate control of seizures. Many anti-seizure medications (ASM) have teratogenic effects on the fetus. We report a case of severe fetal hydantoin syndrome resulting in life-threatening major congenital anomalies. The mother was on phenytoin for the last three years and the pregnancy was not registered. We discuss various features of fetal hydantoin syndrome and the ideal management of epilepsy in pregnancy in brief.

Ethanol and hydantoin are both teratogenic drugs of common usage and are associated with specific syndromes. This is a report on two children born to different heavy drinking, mentally abnormal epileptic women, that were also under treatment with 300 mg/day of hydantoin (mother of case 1 on a regular basis and that of case 2 sporadically) during pregnancy. Both infants displayed the syndrome due to both of these drugs. Particularly relevant were failure to thrive, severe mental retardation, microcephaly, blepharophimosis, hypertelorism, and long philtrum. Additional findings in case 1 included

Hydantoin is an anticonvulsant drug with several side effects. A teratogenic potential has been suggested. The fetal hydantoin syndrome is an entity that consists of a broad range of morphologic and developmental disorders in children born of epileptic mothers exposed to hydantoin during pregnancy. We treated a girl in whom onychopathy was a monosymptomatic or mild form of this syndrome.

The fetal hydantoin syndrome is a variable pattern of altered growth (pre and postnatal), mental deficiency, unusual facies, distal phalangeal hypoplasia, and other defects occurring in some infants exposed in utero to hydantoins. This altered pattern of morphogenesis is distinct from other recognized disorders. It has been reported only in the offspring of women using hydantoins. One case is presented and several aspects of the diagnosis are discussed.

Two siblings have various congenital malformations due to the teratogenic effect of anticonvulsant drugs. Their mother has a 14-year history of continuous anticonvulsant medication. The malformations noted in the older girl are less and in the 8 years younger sister more extensively developed as well as in the fetal hydantoin syndrome. The causes of different severity of the fetal hydantoin syndrome in two sisters are discussed.
