Ordered Chaos: Photoreceptor Mosaics in the Larval and Adult Turquoise Killifish (Nothobranchius furzeri).

An organism's vision is dependent on the photoreceptor types in the eye and how these cells are organized across the retina. The turquoise killifish (Nothobranchius furzeri) is an annual killifish species that lives in the turbid waters of ephemeral pools. Although turquoise killifish are gaining popularity as a model, we know little about their photoreceptors. Here, the turquoise killifish retina was investigated for photoreceptor subtype identity and mosaic organization at the adult and larval stages to assess specialization for their light environment. Turquoise killifish were determined to

Impacts on Atlantic Killifish from Neurotoxicants: Genes, Behavior, and Population-Relevant Outcomes.

Molecular, cellular, and organismal alterations are important descriptors of toxic effects, but our ability to extrapolate and predict ecological risks is limited by the availability of studies that link measurable end points to adverse population relevant outcomes such as cohort survival and growth. In this study, we used laboratory gene expression and behavior data from two populations of Atlantic killifish Fundulus heteroclitus [one reference site (SCOKF) and one PCB-contaminated site (NBHKF)] to inform individual-based models simulating cohort growth and survival from embryonic exposures t

Yellow grub diseases on two seasonal killifish (Cyprinodontiformes, Rivulidae): a histopathological study.

Yellow grub disease is a parasite infection, caused by Clinostomum spp. Metacercariae and affecting axial muscles and internal organs of freshwater fish. Killifishes live in seasonal ponds and are short-lived; in the Neotropical region they are among the most vulnerable vertebrates. The current study analyzes the possible injury and inflammatory reaction caused by parasitic infection with Clinostomum spp. metacercariae in testes, ovaries, liver, intestine and mesentery in the seasonal killifishes Trigonectes aplocheiloides and Titanolebias monstrosus. Apparently, this parasite could migrate th…

The core circadian component, Bmal1, is maintained in the pineal gland of old killifish brain.

Circadian rhythm is altered during aging, although the underlying molecular mechanisms remain largely unknown. Here, we used the turquoise killifish as a short-lived vertebrate model to examine the effects of aging on the major circadian network comprising the four mammalian clock protein homologs, Bmal1, Clockb, Cry1b, and Per3, which are highly conserved in the killifish with 50%-85% amino acid sequence identity to their human counterparts. The amplitude of circadian rhythm was smaller in old fish (14 weeks) than in young fish (6 weeks). In old fish brain, the Bmal1 protein level was signifi

Influence of salinity and pH on bioconcentration of ionizable pharmaceuticals by the gulf killifish, Fundulus grandis.

Estuaries routinely receive discharges of contaminants of emerging concern from urban regions. Within these dynamic estuarine systems, salinity and pH can vary across spatial and temporal scales. Our previous research identified bioaccumulation of the calcium channel blocker diltiazem and the antihistamine diphenhydramine in several species of fish residing in multiple urban estuaries along the Gulf of Mexico in Texas, where field-measured observations of diltiazem in fish plasma exceeded human therapeutic plasma doses. However, there remains a limited understanding of pharmaceutical bioaccumu

Differential sensitivity to pro-oxidant exposure in two populations of killifish (Fundulus heteroclitus).

New Bedford Harbor (MA, U.S.A.; NBH) is a Superfund site inhabited by Atlantic killifish (Fundulus heteroclitus) with altered aryl hydrocarbon receptor (Ahr) signaling, leading to resistance to effects of polychlorinated biphenyls (PCBs) and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). The Ahr is a transcription factor that regulates gene expression of many Phase I and II detoxifying enzymes and interacts with Nrf2, a transcription factor that regulates the response to oxidative stress. This study tested the hypothesis that PCB-resistant killifish exhibit altered sensitivity to oxidative stress

Trace elements in freshwater killifish, Aphanius stoliczkanus, from Oman: A food safety issue.

Freshwater killifish, Aphanius stoliczkanus, were collected from industrial and non-industrial areas in Oman to assess trace element levels in muscle and liver tissues and estimate potential human health risks using the Estimated Daily Intake (EDI) and Target Hazard Quotient (THQ). Samples were obtained from Al Amarart, Al Khawd, and Surur and analyzed using Atomic Absorption Spectroscopy. Fish from Al Amarart exhibited significantly higher concentrations of toxic elements, particularly arsenic (As), cadmium (Cd), and lead (Pb), suggesting a strong influence of industrial activities. In contra

Effects of cadmium and high temperature on some parameters of calcium metabolism in the killifish (Aphanius fasciatus).

This study aims to investigate the influence of high temperature on cadmium (Cd) toxicity in Aphanius fasciatus (Pisces: Cyprinodontidae). For this reason, Cd, mineral, and organic content in the vertebral column as well as the histological structure of gills and bone were compared in fishes exposed for 30 days to Cd (2 mg/L CdCl2) and/or high temperature (26 °C). Cd exposure caused a negative correlation between Cd and Ca concentrations (r = 0.98, p < 0.05), as well as a significant decrease in inorganic components (p < 0.05) and ash weight/dry weight ratio (p < 0.05) in the vertebral column.

Further consideration of phenobarbital effects on cytochrome P-450 activity in the killifish, Fundulus heteroclitus.

1. Ethoxyresorufin O-deethylase (EROD) activity, aldrin epoxidase (AE) activity, cytochrome P-450 content, and levels of cytochrome P-450E (the major BNF-inducible P-450 form and primary EROD catalyst in scup) or its homologues were measured in hepatic microsomes isolated from Fundulus heteroclitus, scup (Stenotomus chrysops) and brook trout (Salvelinus fontinalis) treated with beta-naphthoflavone (BNF) or phenobarbital (PB). 2. In all three teleost species, BNF treatment caused expected increases in P-450 content, EROD activity and P-450E level; but either no change or a slight decrease in AE

The master male sex determinant Gdf6Y of the turquoise killifish arose through allelic neofunctionalization.

Although sex determination is a fundamental process in vertebrate development, it is very plastic. Diverse genes became major sex determinants in teleost fishes. Deciphering how individual sex-determining genes orchestrate sex determination can reveal new actors in sexual development. Here, we demonstrate that the Y-chromosomal copy of the TGF-β family member gdf6 (gdf6Y) in Nothobranchius furzeri, an emerging model organism in aging research, gained the function of the male sex determinant through allelic diversification while retaining the skeletal developmental function shared with the X-ch

Olfactory and gustatory chemical sensor systems in the African turquoise killifish: Insights from morphology.

Smell and taste are extensively studied in fish species as essential for finding food and selecting mates while avoiding toxic substances and predators. Depending on the evolutionary position and adaptation, a discrete variation in the morphology of these sense organs has been reported in numerous teleost species. Here, for the first time, we approach the phenotypic characterization of the olfactory epithelium and taste buds in the African turquoise killifish (Nothobranchius furzeri), a model organism known for its short lifespan and use in ageing research. Our observations indicate that the o

Age-related alterations in the behavioral response to a novel environment in the African turquoise killifish (Nothobranchius furzeri).

The African turquoise killifish (Nothobranchius furzeri) has emerged as a popular model organism for neuroscience research in the last decade. One of the reasons for its popularity is its short lifespan for a vertebrate organism. However, little research has been carried out using killifish in behavioral tests, especially looking at changes in their behavior upon aging. Therefore, we used the open field and the novel tank diving test to unravel killifish locomotion, exploration-related behavior, and behavioral changes over their adult lifespan. The characterization of this behavioral baseline

Zinc chloride rapidly stimulates efflux transporters in renal proximal tubules of killifish (Fundulus heteroclitus).

Multidrug resistance-related protein 2 (Mrp2) is an ATP-driven efflux pump at the luminal membrane in renal proximal tubules. It acts as detoxification mechanism by transporting xenobiotics and metabolic products into urine. The trace element zinc is essential for cellular growth, differentiation and survival. It modulates immune response and is used as dietary supplement. Here, we found that 0.1-10μM ZnCl2 rapidly stimulated transport of the Mrp2 probe substrate Texas Red (TR) in isolated killifish renal proximal tubules, which provide an established model system to measure efflux transporter

Alimentary Tract Anatomy and Morphology in Early Adult Mediterranean Killifish Aphanius fasciatus (Valenciennes, 1821).

Background/Aims: The Mediterranean killifish, Aphanius fasciatus (Valenciennes, 1821), is a small euryhaline and eurytherm cyprinodont. While its ecology and role as a bioindicator are well known, its anatomy remains poorly understood. This study aimed to provide the first detailed description of the digestive tract of A. fasciatus, from the oropharyngeal cavity to the rectum. Methods: An anatomical and morphological approach supported by light microscopy was applied to examine oral, pharyngeal, and intestinal structures in adult specimens. Results: The jaws bear tricuspids incisiform teeth, w

Loss of killifish cGAS attenuates age-related signatures but does not affect organismal life span.

The cGAS/STING pathway is a central innate immune signaling pathway responsive to cytosolic DNA. Chronic activation of this pathway promotes numerous age-related pathologies, but its impact on lifespan remains unknown. Here we engineer a cGAS knockout (KO) in the turquoise killifish Nothobranchius furzeri to assess effects on physiology and aging. In cultured fibroblasts, cGAS deficiency results in elevated DNA damage but reduces radiation-induced senescence and enhances cellular proliferation. In vivo, cGAS KO attenuates DNA damage-induced transcriptional responses in young fish, and blunts a

Organization of Serotonergic Cell Populations in the Brain and Spinal Cord of the Short-Lived African Turquoise Killifish.

The African turquoise killifish is a short-lived vertebrate that is gaining interest as a model for understanding degenerative processes associated with aging, including neurodegeneration. However, due to its relatively recent adoption in laboratory research, detailed studies on the adult neuroanatomy of the killifish, particularly regarding the organization of neurotransmitter systems and their age-related changes, remain limited. Here, we study the organization of serotonergic cell populations in the brain and spinal cord of juvenile/young adult (1-3-months-old) and geriatric (5-6-months-old

Constant temperature and fluctuating temperature have distinct effects on hypoxia tolerance in killifish (Fundulus heteroclitus).

Climate change is leading to rapid change in aquatic environments, increasing the mean and variability of temperatures, and increasing the incidence of hypoxia. We investigated how acclimation to constant temperatures or to diel temperature fluctuations affects hypoxia tolerance in mummichog killifish (Fundulus heteroclitus). Killifish were acclimated to constant cool (15°C), constant warm (25°C) or a diel temperature cycle (15°C at night, 25°C during day) for 6 weeks. We then measured hypoxia tolerance (time to loss of equilibrium in severe hypoxia, tLOE; critical O2 tension, Pcrit), whole-an

Genetic tools for the study of the mangrove killifish, Kryptolebias marmoratus, an emerging vertebrate model for phenotypic plasticity.

Kryptolebias marmoratus (Kmar), a teleost fish of the order Cyprinodontiformes, has a suite of unique phenotypes and behaviors not observed in other fishes. Many of these phenotypes are discrete and highly plastic-varying over time within an individual, and in some cases reversible. Kmar and its interfertile sister species, K. hermaphroditus, are the only known self-fertile vertebrates. This unusual sexual mode has the potential to provide unique insights into the regulation of vertebrate sexual development, and also lends itself to genetics. Kmar is easily adapted to the lab and requires litt

Quantification of noradrenergic-, dopaminergic-, and tectal-neurons during aging in the short-lived killifish Nothobranchius furzeri.

Parkinson's disease (PD) is characterized by phosphorylation and aggregation of the protein α-Synuclein and ensuing neuronal death progressing from the noradrenergic locus coeruleus to midbrain dopaminergic neurons. In 2019, Matsui and colleagues reported a spontaneous age-dependent degeneration of dopaminergic neurons and an even greater neurodegeneration of the noradrenergic neurons in the short-lived killifish Nothobranchius furzeri. Given the great possible relevance of a spontaneous model for PD, we assessed neurodegeneration of noradrenergic and dopaminergic neurons in two further labora
