A UFMylation-COPII axis orchestrates lipid transport in intestinal enterocytes and regulates systemic lipid balance.

BACKGROUND: Intestinal lipid absorption and chylomicron secretion are essential for systemic lipid homeostasis, yet the regulatory mechanisms coordinating lipoprotein assembly and ER export remain poorly understood. UFMylation is a newly identified ubiquitin-like modification pathway that plays critical roles in endoplasmic reticulum (ER)-related cellular activities such as protein quality control, ER-associated degradation (ERAD) and ER-phagy. However, its role in intestinal lipid transport and systemic lipid homeostasis is completely unclear. METHODS: To elucidate the role of UFMylation in …

A Call for Use of Lipid Fractionation Studies in Patients With Abnormal Standard Lipid Profiles.

A 54-year-old female individual with a history of hyperlipidemia, hypothyroidism, latent autoimmune diabetes of the adult (LADA) with hypoglycemia unawareness, vitiligo, and asthma presented with exercise-limiting pleuritic chest pain and dyspnea. She underwent a full pulmonary and cardiac workup locally and at Mayo Clinic, which were normal and negative for pulmonary embolism and showed no evidence of cardiac ischemia. Her family history and abnormal lipid panels suggested possible familial hypercholesterolemia by Dutch criteria. Lipid fractionation studies were done to further assess abnorma

Atomistic study of lipid membranes containing chloroform: looking for a lipid-mediated mechanism of anesthesia.

The molecular mechanism of general anesthesia is still a controversial issue. Direct effect by linking of anesthetics to proteins and indirect action on the lipid membrane properties are the two hypotheses in conflict. Atomistic simulations of different lipid membranes subjected to the effect of small volatile organohalogen compounds are used to explore plausible lipid-mediated mechanisms. Simulations of homogeneous membranes reveal that electrostatic potential and lateral pressure transversal profiles are affected differently by chloroform (anesthetic) and carbon tetrachloride (non-anesthetic

Involvement of Lipids and Lipid Mediators in Inflammation and Atherogenesis.

Atherosclerosis is the pathophysiological basis for major diseases, such as coronary heart disease, cerebral stroke, and peripheral arterial disease, which have become epidemic in modern Western society. Atherosclerosis has a complex nature that involves mutually related immune and metabolic mechanisms. Many cells of the vascular wall and peripheral bloodstream, including endothelial cells, monocytes and macrophages, platelets, and others, are involved in the development and progression of atherosclerosis. These cells perform a number of innate immune functions, disorders of which are associat

Lipids and lipid regulators in intraocular pressure homeostasis.

Increased intraocular pressure is the strongest correlated modifiable risk factor for developing primary open-angle glaucoma (POAG). Lipids have long been known to be a major constituent of aqueous humor. Lipid mediators, prostaglandins for example, are the first-line treatment for glaucoma. Innovative technologies have made the investigation of lipids in small quantities possible, and interest in identifying lipids as new pharmacological targets has grown in ophthalmology. There is expanding evidence to suggest that lipids and their active metabolites play a role in POAG pathophysiology, as d

Dietary lipids, gut microbiota and lipid metabolism.

The gut microbiota is a central regulator of host metabolism. The composition and function of the gut microbiota is dynamic and affected by diet properties such as the amount and composition of lipids. Hence, dietary lipids may influence host physiology through interaction with the gut microbiota. Lipids affect the gut microbiota both as substrates for bacterial metabolic processes, and by inhibiting bacterial growth by toxic influence. The gut microbiota has been shown to affect lipid metabolism and lipid levels in blood and tissues, both in mice and humans. Furthermore, diseases linked to dy

Stimulation of splenocytes in C3H/HeJ mice with Porphyromonas gingivalis lipid A in comparison with enterobacterial lipid A.

Porphyromonas gingivalis 381 lipid A induced strong mitogenic response in splenic B cells separated from LPS-non-responsive C3H/HeJ mice as well as LPS-responsive C3H/HeN mice, by using a magnetic cell sorting system. The lipid A also exhibited mitogenic activity in splenic T cells. P. gingivalis lipid A induced lower production of interleukin-1 beta (IL-1 beta) in splenic macrophage cultures and exhibited a comparable IL-6 producing activity in splenic B cells of C3H/HeN mice as compared to Escherichia coli-type synthetic lipid A (compound 506) and monophosphoryl lipid A from Salmonella minne…

The interaction of TRPV1 and lipids: Insights into lipid metabolism.

Transient receptor potential vanilloid 1 (TRPV1), a non-selective ligand-gated cation channel with high permeability for Ca2+, has received considerable attention as potential therapeutic target for the treatment of several disorders including pain, inflammation, and hyperlipidemia. In particular, TRPV1 regulates lipid metabolism by mechanisms that are not completely understood. Interestingly, TRPV1 and lipids regulate each other in a reciprocal and complex manner. This review surveyed the recent literature dealing with the role of TRPV1 in the hyperlipidemia-associated metabolic syndrome. Bes

Analysis of lipid profile in lipid storage myopathy.

Lipid dysmetabolism disease is a condition in which lipids are stored abnormally in organs and tissues throughout the body, causing muscle weakness (myopathy). Usually, the diagnosis of this disease and its characterization goes through dosage of Acyl CoA in plasma accompanied with evidence of droplets of intra-fibrils lipids in the patient muscle biopsy. However, to understand the pathophysiological mechanisms of lipid storage diseases, it is useful to identify the nature of lipids deposited in muscle fiber. In this work fatty acids and triglycerides profile of lipid accumulated in the muscle…

Replacing Helper Lipids With Cationic Lipids Enhances mRNA Lipid Nanoparticles Stability in Solution.

Messenger RNA lipid nanoparticles (mRNA-LNPs) have achieved remarkable success in clinical vaccination efforts to curb the COVID-19 pandemic, and have attracted tremendous interest from both industry and academia to broaden their biomedical applications. However, their storage and transportation rely heavily on the cold chain to prevent rapid degradation in non-frozen solutions, posing significant challenges for logistics and accessibility. Therefore, enhancing the in-solution stability of mRNA LNPs is crucial. Herein, the role of helper phospholipids is investigated in storage stability of mR

Efficacy and safety of a lipid-containing artificial tear compared with a non-lipid containing tear: a randomized clinical trial.

BACKGROUND: Dry eye disease (DED) is a prevalent condition affecting over 16 million patients in the USA. DED and the symptoms of ocular discomfort are debilitating and a significant burden on patients. If left untreated, DED can progress to cause severe pathology. Treatment is often initiated by patients without consulting a healthcare professional. This study investigated the safety and efficacy of a novel lipid-containing eye drop (BTC), which might better mimic the components of natural tears. METHODS: This was a multicenter, randomized, double-masked, active control, two arm, parallel gr

A phospholipid-like charge-reversible lipid for PEG-free, stable, and low-inflammatory mRNA lipid nanoparticles.

The development of lipid nanoparticles (LNPs) facilitated optimal usage of mRNA drugs to achieve clinical success during the COVID-19 pandemic. Conventional LNPs rely on ionizable lipids for mRNA encapsulation and endosomal escape. However, the high hydrophobicity of ionizable lipids leads to aggregation of these LNPs under physiological conditions, requiring polyethylene glycol (PEG) modification for stabilization. Ionizable lipids and PEG induce inflammation and hypersensitivity reactions, respectively, after intramuscular administration. Therefore, developing low-inflammatory LNPs is essent

Lipid Metabolism as a Therapeutic Target in Arrhythmogenic Cardiomyopathy: Emerging Therapeutic Implications of Lipid-Lowering Therapy.

Arrhythmogenic cardiomyopathy (ACM) is a hereditary myocardial disorder characterized by progressive replacement of ventricular myocardium with fibro-fatty tissue, leading to life-threatening ventricular arrhythmias and heart failure. Historically, management has focused on arrhythmia suppression and prevention of sudden cardiac death (SCD). However, recent advances in understanding disease mechanisms have redefined ACM as a structural-metabolic cardiomyopathy. Central to this evolving paradigm is the "adipogenic switch", driven by desmosomal dysfunction, impaired Wnt/β-catenin signaling, and

Is Lipopolysaccharide-Induced Lipid Metabolism Disorder in Testis of Rats a Consequence of Plasma Lipid Changes?

PURPOSE: The systemic infection and inflammation can result in testes injury, whereas the exact mechanism is unknown. The lipid metabolism has a dual impact on controlling metabolism and inflammation, which is a potential pathway. The objective of this study was to determine if changes in plasma lipids during lipopolysaccharide (LPS)-induced systemic inflammation affect the dysregulation of testes lipid metabolism. MATERIALS AND METHODS: LPS (5 mg/kg) was used to induce systemic inflammation in rats after a single intraperitoneal injection. After 4 weeks, the rats were sacrificed, and the ser

Lipid traits and lipid-lowering drug target genes and risk of melanoma: a mendelian randomization study.

Our study aimed to investigate the role of lipids in melanoma risk and the effect of lipid-lowering drug targets on melanoma. Using Mendelian Randomization analysis, we examined the genetic agents of nine lipid-lowering drugs and their association with melanoma risk. We found that genetically proxied inhibition of HMGCR, ABCG5/ABCG8, and ANGPTL3 was associated with a reduced risk of melanoma. On the other hand, inhibition of LPL and Apo-B100 was significantly associated with an increased risk of melanoma. Sensitivity analyses did not reveal any statistical evidence of bias from pleiotropy or g

A Systematic Review of Lipid Management in Secondary Prevention and Comparison of International Lipid Management Pathways.

Acute coronary syndrome remains a major cause of morbidity and mortality despite significant improvements in its prevention and management. Lipid management and other risk factors such as hypertension, diabetes, obesity, smoking and sedentary lifestyle stratification is the key to minimising this risk. Lipid management is an important part of secondary prevention and patients are historically undertreated after post-acute coronary syndrome. We performed a narrative review on observational studies on lipid management pathways post ACS on PubMed, Google Scholar, Journal Storage and ScienceDirect

Acute atherosis and diffuse lipid infiltration of the placental bed: A review of historical lipid studies.

Based on a variety of tissue samples, including Caesarean hysterectomy specimens with the placenta in situ, a detailed map of uteroplacental vascular lesions was established in over a century of research. One such lesion is acute atherosis of unremodelled basal and uteroplacental arteries, defined by the presence of fibrinoid necrosis, subendothelial macrophage foam cells, and perivascular lymphocytic infiltration. Two studies conducted over 50 years ago used Oil Red O staining of frozen tissue sections to visualise lipid infiltration of placental bed vessels and document the presence of lipid

A waxy path: Lipid dynamics during tuberculosis.

Infectious pathogens must adapt to the metabolic landscapes of their hosts, often exploiting host-derived nutrients while mitigating the stresses these resources impose. Among these challenges, host lipids are both a rich energy source and a potential liability, requiring tight regulation during infection. Mycobacterium tuberculosis, the causative agent of tuberculosis and the leading cause of death from a single infectious agent, exemplifies an extreme adaptation to lipid-rich environments. Recent studies have uncovered coordinated host and bacterial pathways that mediate fatty acid transfer,
