Exploring optimal folic acid supplementation levels for lactating-pregnant rabbit does with different litter size.

The lactation-pregnancy overlap in the industrialized 49-d breeding model increases nutritional demands for lactating-pregnant rabbit does. This study examined the effects of folic acid (FA) on the production performance and intestinal microflora of does with different litter sizes (LS, or number of kits). A total of 144 third-parity Hyplus does, aged 11 mo and weighing approximately 5.00 ± 0.07 kg, were divided into 4 treatment groups: control group (basal diet) and FA groups (basal diet + 15, 30, and 45 mg/kg FA). Does with LS of 5, 7, and 9 were selected after weaning. Our findings revealed…

Resolving and functional analysis of RNA editing sites in sheep ovaries and associations with litter size.

Sheep litter size is a critical trait in mutton production. While litter size regulation in relation to DNA transcription have been rigorously investigated, the function of RNA editing remains less explored. To elucidate the mechanisms controlling sheep fecundity at the RNA editing level and identify pivotal RNA editing sites, this study scrutinised RNA editing sites (RESs) in follicular and luteal phases of ovaries from sheep with high and low fecundity, and the functions of population-specific RESs were subsequently analysed. A total of 2 182 475 RESs, 74.61% of which were A-to-I and C-to-U …

Analysis of goat PPP6C mRNA profile, detection of genetic variations, and their associations with litter size.

The Protein Phosphatase 6 Catalytic Subunit (PPP6C) is evolutionarily a conserved gene in eukaryotes known to play a significant role in mammalian reproduction. This study aimed to investigate expression patterns of PPP6C and explore its association with litter size in Shaanbei white cashmere (SBWC) goats. Initially, we determined the mRNA expression levels of PPP6C in both male and female goats across multiple tissues. The results showed that PPP6C mRNA was expressed in multiple tissues, with higher levels in the testis and fallopian tubes, suggesting its involvement in goat reproduction. Add…

A novel nonsynonymous SNP in the OLR1 gene associated with litter size in Guizhou white goats.

Oxidized low-density lipoprotein receptor-1 (OLR1) encodes a low-density lipoprotein receptor belonging to the C-type lectin superfamily, which is closely related to reproduction. OLR1 is associated with fecundity in Awassi sheep. However, its effect on litter size has not been investigated in goats. In this study, OLR1 sequences and their mRNA expression levels in the gonadal axis of Guizhou white goats were evaluated to investigate the relationship between gene polymorphisms and litter size. In addition, the potential effects of a nonsynonymous substitution were evaluated using a bioinformat…

Influence of litter size on placental stereological characteristics and its relationship with neonatal outcome in dogs.

Although development of placental structure towards the endometrial layers is determined by effective tissue remodeling, expansion within the uterine surface is limited in polytocous species. We aimed to evaluate the influence of litter size on placental morphological characteristics and puppies' clinical outcome and physical development during neonatal period. Bitches and their neonates were assigned to: Large-Sized Litter Group (n = 8 bitches and n = 16 neonates; litter size exceeding breeds reference) and Small/Normal-Sized Litter Group (n = 7 bitches; n = 12 neonates). Placentas were colle…

Glucosamine inhibits decidualization of human endometrial stromal cells and decreases litter sizes in mice.

Embryo implantation in the uterus depends on decidualization of the endometrial stromal cells (ESCs), and glucose utilization via the pentose phosphate pathway is critical in this process. We hypothesized that the amino sugar glucosamine may block the pentose phosphate pathway via inhibition of the rate-limiting enzyme glucose-6-phosphate dehydrogenase in ESCs and therefore impair decidualization and embryo implantation, thus preventing pregnancy. Both human primary and immortalized ESCs were decidualized in vitro in the presence of 0, 2.5, or 5 mM glucosamine for 9 days. Viability assays demo

Maintenance of body temperature in the neonatal lamb: effects of breed, birth weight, and litter size.

To survive, the newborn lamb must be able to maintain body temperature and to stand and move to the udder to suck colostrum to fuel heat production. The objective of this study was to investigate whether neonatal lambs showing slow behavioral progress to standing and sucking also have an impaired ability to maintain body temperature. The time taken to stand and suck after birth, rectal temperatures, and blood samples were collected from 115 newborn single, twin, and triplet lambs of 2 breeds, Scottish Blackface and Suffolk, which are known to show variations in their neonatal behavioral progre

The influence of litter size on brain damage caused by hypoxic-ischemic injury in the neonatal rat.

Hypoxic ischemia is a common cause of brain injury in the human neonate. This can be mimicked in the neonatal rat, but produces variable injury. The present study investigated the influence of litter size on the severity and variability of damage caused by hypoxic-ischemic injury in neonatal rats. Groups of 7-d-old pups from birth-sized litters (13-15 pups), or from litters culled to 10 on postnatal d 2, and 8- and 9-d-old pups from birth-sized litters, were exposed to common carotid artery occlusion and then, 3 h later, hypoxia (2 h 15 min, 8% oxygen). Damage was assessed histologically 72 h …

[Endocrine pancreas function in rats in the early postnatal period. The effect of feeding and litter size].

The rat pancreatic endocrine region during the early postnatal period was evaluated depending on the litter size. The body and pancreatic weight lowers and the blood glucose content tends to rise alongside with the litter size increasing. Insulin levels are augmented and glucagon content is reduced in the pancreas of newborn rats with the large litter. The feeding of the animals is accompanied by rapid enhancement of the gland hormone concentration. The per cent content of insulin-secreting cells in Langerhans' islets of newborn rats is rather higher than that of adult animals, being dependent…

Longitudinal metabolic and progesterone dynamics in estrus-synchronized ewes: influence of pregnancy outcome and litter size.

Metabolic status plays a critical role in reproductive efficiency in small ruminants; however, longitudinal information describing metabolic adaptations from estrus synchronization through gestation and the immediate postpartum period remains limited. This study investigated temporal changes in selected serum metabolic and hormonal parameters in estrus-synchronized ewes and compared pregnant (PR) and non-pregnant (NPR) animals, with particular emphasis on the influence of litter size. A total of 235 ewes were synchronized during the breeding season. Of these, 150 animals (75 PR and 75 NPR) wer…

Cathepsin D knockdown regulates biological behaviors of granulosa cells and affects litter size traits in goats.

Cathepsin D (CTSD), the major lysosomal aspartic protease that is widely expressed in different tissues, potentially regulates the biological behaviors of various cells. Follicular granulosa cells are responsive to the increase of ovulation number, hence indirectly influencing litter size. However, the mechanism underlying the effect of CTSD on the behaviors of goat granulosa cells has not been fully elucidated. This study used immunohistochemistry to analyze CTSD localization in goat ovarian tissues. Moreover, western blotting was applied to examine the differential expression of CTSD in the …

Early life overnutrition induced by litter size manipulation decreases social play behavior in adolescent male rats.

Several studies have investigated the effects of artificial litter size adjustment on offspring development. Social play behavior is important for neurobehavioral development and is impaired in several developmental psychiatric disorders. This study therefore investigated the effect of litter size on play behavior in adolescent rats. On postnatal day (PND) 2, litters were adjusted to a small litter (SL) size of 3 pups per dam or normal litter (NL) size of 12 pups per dam. Maternal behaviors scored daily during the first week of lactation (PND2-8) revealed that arched nursing and pup licking be…

Postnatal overfeeding in rodents by litter size reduction induces major short- and long-term pathophysiological consequences.

Numerous studies have demonstrated that the early postnatal environment can influence body weight and energy homeostasis into adulthood. Rodents raised in small litters have been shown to be a useful experimental model to study the short- and long-term consequences of early overnutrition, which can lead to modifications not only in body weight but also of several metabolic features. Postnatal overfeeding (PNOF) induces early malprogramming of the hypothalamic system, inducing acquired persisting central leptin and insulin resistance and an increase in orexigenic signals. Visceral white adipose

Farrowing rates and litter size following transfer of vitrified porcine embryos into a commercial swine herd.

The objective was to determine farrowing rates and litter sizes that could be achieved in a typical farm-to-farm porcine embryo transfer program using vitrified blastocysts that were zona pellucida intact when cryopreserved. The embryos were transferred surgically on-farm into recipient sows that were managed throughout gestation and farrowing under the same conditions as other sows in the herd. Twenty recipient sows (mean parity 2.1) received a total of 568 embryos; seven received 203 embryos derived from donor sows, five received 139 embryos from gilts and eight received a mixture of 161 emb…

Reproductive performance in a diallel cross among lines of mice selected for litter size and body weight.

Genetic factors affecting female reproductive performance in lines of mice with a known history of selection were estimated from a 5 X 5 diallel cross. Lines were selected as follows: large litter size at birth (L+); large 6-wk body weight (W+); an index for large litter size and small 6-wk body weight (L+W-); the complementary index (L-W+) and randomly (K). Partitioning of direct and correlated responses for litter size, 6-wk body weight and related traits into average direct genetic (li) and average maternal genetic (mi) effects indicated that the magnitude of differences in li exceeded thos…

Functional and Genetic Insights into the Role of the NR4A1 Gene in the Litter Size of the Shaanbei White Cashmere Goat.

Nuclear receptor subfamily 4 group A member 1 (NR4A1) plays a crucial role in regulating various physiological processes. As gene mining for reproductive traits is essential, this study aimed to investigate the mRNA expression, genetic variation, and association of the NR4A1 gene with goat litter size. We examined the mRNA expression levels of the NR4A1 gene in eight different tissues of female Shaanbei White Cashmere (SBWC) goats (n = 6). Then, a novel 11-bp insertion/deletion (InDel) variant was genotyped in 1136 SBWC goats, 87 SNPs were identified through resequencing (n = 120), and selecti…
