[Linear Darier disease in two siblings. An example of loss of heterozygosity].

BACKGROUND: Darier's disease or keratosis follicularis is an autosomal dominant acantholytic disorder that frequently arises as a result of spontaneous mutation. It is either a generalized or localized condition due to a mutation in the SERCA2 12q23-q24,1 resulting in a faulty organization of the tonofilaments. We present two siblings affected with the linear form of this disorder and discuss these cases as an example of the genetic mechanism of loss of heterozygosity. CASE REPORTS: A 7 year-old girl was referred for evaluation of linear lesions present since the first year of age. Examinatio

Allelotype analysis of oesophageal adenocarcinoma: loss of heterozygosity occurs at multiple sites.

Deletions of tumour-suppressor genes can be detected by loss of heterozygosity (LOH) studies, which were performed on 23 cases of adenocarcinoma of the oesophagus, using 120 microsatellite primers covering all non-acrocentric autosomal chromosome arms. The chromosomal arms most frequently demonstrating LOH were 3p (64% of tumours), 5q (45%), 9p (52%), 11p (61%), 13q (50%), 17p (96%), 17q (55%) and 18q (70%). LOH on 3p, 9p, 13q, 17p and 18q occurred mainly within the loci of the VHL, CDKN2, Rb, TP53 and DCC tumour-suppressor genes respectively. LOH on 5q occurred at the sites of the MSH3 mismat

Prediction of recurrence of nonfunctioning pituitary tumours by loss of heterozygosity analysis.

BACKGROUND: Postsurgical regrowth or recurrence of nonfunctioning pituitary adenomas (NFAs) is not uncommon and often requires further surgery or radiotherapy (DXT). Routine postoperative DXT increases the incidence of hypopituitarism, which is associated with increased morbidity and mortality. Identification of genetic abnormalities in the tumour tissue, which can predict recurrence, may allow targeting DXT to the most appropriate patients. DESIGN AND METHODS: We have performed loss of heterozygosity (LOH) analysis on 96 NFAs of which 43 (45%) were recurrent and 53 (55%) were nonrecurrent tu

Loss of heterozygosity on chromosome 5 in adults with acute lymphoblastic leukemia.

Cytogenetic abnormalities are among the most important pretreatment predictors of outcome in patients with acute lymphoblastic leukemia (ALL). Deletions of genetic material can result in loss of tumor suppressor genes or other translation products that are crucial in maintaining an orderly cell cycle sequence or viability of the apoptotic cascade. Chromosome 5 contains many genes that are relevant in hematopoiesis. Deletions of chromosome 5 or parts thereof are found frequently in myelodysplastic syndromes (MDS) and acute myeloid leukemias (AML) where they are associated with a poor prognosis.

Frequent loss of heterozygosity for markers on chromosome arm 10q in chondrosarcomas.

Many tumors exhibit loss of heterozygosity (LOH) for polymorphic markers in regions of the genome that contain genes whose normal function can suppress tumor growth. Mapping of regions of LOH can help identify putative tumor suppressor loci that play a role in the pathogenesis of a disease. We evaluated 18 chondrosarcomas for LOH at 17 short tandem-repeat polymorphism loci on chromosome 10. Sixty-seven percent of the tumors (12/18) showed LOH for at least one marker and in most of these tumors the region of loss spanned all or large portions of the chromosome. By determining the smallest segme

[Expression, loss of heterozygosity, and methylation of GNAT1 gene in nasopharyngeal carcinoma].

BACKGROUND & OBJECTIVE: In nasopharyngeal carcinoma (NPC), chromosome 3p21.3 is a high frequency deletion region. Evidences from both loss of heterozygosity (LOH) and functional studies showed that there may exists NPC-related tumor suppressor genes on 3p21.3. This study was to investigate the expression, LOH, and methylation of GNAT1 gene, which locates at 3p21.3, in NPC. METHODS: The expression of GNAT1 in 33 specimens of primary NPC and 15 specimens of chronic nasopharyngitis tissue was detected by reverse transcription-polymerase chain reaction (RT-PCR). LOH and promoter methylation statu

Loss of heterozygosity in ampulla of Vater neoplasms during adenoma-carcinoma sequence.

BACKGROUND: Ampulla of Vater cancers arise from precancerous lesions and existence of an adenoma-carcinoma sequence is based on morphological observations. MATERIALS AND METHODS: We studied the loss of heterozygosity (LOH) in 22 adenomas, 32 carcinomas and 10 metastatic lesions using nine dinucleotide-repeated sequences in 3p, 8p, 8q, 9p, 10q, 13q, 17p, 17q, 18q. RESULT: High LOH frequencies (> 50%) of 9p (IFNA) and 17p (TP53) were observed in adenomas and carcinomas. The frequency of LOH is higher in adenoma (55.6%) than in carcinoma (40%) for 8p (D8S261), but it is the same in cases having

Loss of heterozygosity of chromosome 14q in low- and high-grade meningiomas.

Abnormalities of chromosome 22q have been well studied as a major molecular genetic event in meningiomas. Chromosome 14q loss has also been shown to be a common phenomenon. However, only a few studies have reported molecular genetic changes of this chromosome in meningioma. In this study, we examined 41 sporadic meningiomas of different histological subtypes and grades with 15 polymorphic microsatellite markers covering a wide region on chromosome 14q. Overall, 37% (15 of 41) of cases showed loss of heterozygosity for one or more allelic markers. Thirty percent (10 of 33) of benign tumors show

Tumour TIF1 mutations and loss of heterozygosity related to cancer-associated myositis.

OBJECTIVES: To analyse the influence of genetic alterations and differential expression of transcription intermediary factor 1 (TIF1) genes in the pathophysiology of cancer-associated myositis (CAM). METHODS: Paired blood and tumour DNA samples from patients with anti-TIF1γ-positive CAM and from controls were analysed by whole-exome sequencing for the presence of somatic mutations and loss of heterozygosity (LOH) in their TIF1 genes. The genesis and maintenance of the autoimmune process were investigated immunohistochemically by studying TIF1γ expression in the different tissues involved in C

Loss of heterozygosity 4q24 and TET2 mutations associated with myelodysplastic/myeloproliferative neoplasms.

Chromosomal abnormalities are frequent in myeloid malignancies, but in most cases of myelodysplasia (MDS) and myeloproliferative neoplasms (MPN), underlying pathogenic molecular lesions are unknown. We identified recurrent areas of somatic copy number-neutral loss of heterozygosity (LOH) and deletions of chromosome 4q24 in a large cohort of patients with myeloid malignancies including MDS and related mixed MDS/MPN syndromes using single nucleotide polymorphism arrays. We then investigated genes in the commonly affected area for mutations. When we sequenced TET2, we found homozygous and hemizyg

Loss of heterozygosity on chromosome 16p13.3 in hamartomas from tuberous sclerosis patients.

Tuberous sclerosis (TSC) is an autosomal dominant condition with characteristic skin lesions, mental handicap, seizures and the development of hamartomas in the brain, heart, kidneys and other organs. Linkage studies have shown locus heterogeneity with a TSC gene mapped to chromosome 9q34 and a second, recently identified on 16p13.3. We have analysed DNA markers in eight hamartomas and one tumour from TSC patients and found allele loss on 16p13.3 in three angiomyolipomas, one cardiac rhabdomyoma, one cortical tuber and one giant cell astrocytoma. We suggest that the TSC gene on 16p13.3 functio

Loss of heterozygosity of CYP2D6 enhances the sensitivity of hepatocellular carcinomas to talazoparib.

BACKGROUND: Loss of heterozygosity (LOH) diminishes genetic diversity within cancer genomes. A tumour arising in an individual heterozygous for a functional and a loss-of-function (LoF) allele of a gene occasionally retain only the LoF allele. This can result in deficiency of specific protein activities in cancer cells, creating unique differences between tumour cells and normal cells of the individual. Such differences may constitute vulnerabilities that can be exploited through allele-specific therapies. METHODS: To discover frequently lost genes with prevalent LoF alleles, we mined the 100

Altered expression and loss of heterozygosity of the LOT1 gene in ovarian cancer.

OBJECTIVES: Previously, we demonstrated that the LOT1 (PLAGL1/ZAC1) gene encodes a zinc-finger transcription factor and has growth suppressive effects in carcinoma cell lines. The gene is localized on chromosome 6q24-25, a common site for loss of heterozygosity (LOH) in many solid tumors including ovarian cancer. In this study, we evaluated the LOT1 gene expression and allelic deletion in the tumor tissues in order to provide additional evidence to support the gene's potential role in cancer. METHODS: The LOT1 gene expression was analyzed in malignant ovarian epithelium obtained from frozen h

Frequent loss of heterozygosity in the region of D1S450 at 1p36.2 in myelodysplastic syndromes.

To understand the underlying mechanisms in myelodysplastic syndromes (MDS) by identifying target tumor suppressor genes, we performed a detailed deletional mapping of the short arm of chromosome 1 in 38 paired samples of bone marrow and peripheral blood obtained from individuals with MDS by PCR amplification of a total of 23 highly informative microsatellite sequences. We identified the commonly deleted region between D1S508 and D1S244. LOH of this region was found in five patients (13%). In addition, LOH at 1p was associated with a poor clinical outcome, suggesting that the deletion of a gene

Loss of heterozygosity in adenomyosis on hMSH2, hMLH1, p16Ink4 and GALT loci.

Adenomyosis is a gynecological condition in which tissue histologically similar to that in endometrium is found within the myometrium in the uterus. Although, lesions of both adenomyosis and endometriosis are identical to their sources with respect to structure and function, they are generally regarded as separate and distinct nosologic processes. In this study, we used 17 microsatellite markers, in four tetraplex and one single PCR assay, to determine the incidence of loss of heterozygosity (LOH) in 31 cases of adenomyosis. The markers used are located close to tumor suppressor genes, DNA rep

Loss of heterozygosity at 12P12-13 in primary and metastatic prostate adenocarcinoma.

PURPOSE: Our laboratory has recently identified a 1 to 2 Mb homozygous deletion at 12p12-13 in a prostate cancer specimen and determined that the p27/kip1 gene lies within the deletion. While immunohistochemical analysis has implicated p27/kip1 in prostate carcinoma, no previous studies had identified genetic abnormalities at this locus. Here, we examined primary and metastatic prostate tumors to determine if allelic loss occurs at this locus in localized disease and if it increases the risk of metastatic, high stage or high-grade disease. MATERIALS AND METHODS: DNA was extracted from prostat

Loss of heterozygosity on the long arm of chromosome 11 in nasopharyngeal carcinoma.

Loss of heterozygosity (LOH) on the long arm of chromosome 11 had been reported in many types of solid tumors. In this study, we investigated the LOH patterns of chromosome 11 on 52 primary nasopharyngeal carcinomas using 10 microsatellite polymorphic markers. The results revealed that 28 of the 52 cases (53.8%) demonstrated LOH on at least one of the nine 11q microsatellite loci studied. The highest frequencies of LOH were found at the two loci D11S2000 (36.1%) and D11S934 (34.5 %), both located at 11q22-24. Two distinct regions of deletion were found at 11q, with the first one defined by INT
