Three new carbon-skeleton triterpenoids, named daphniphyanoids A-C (1-3), featuring an uncommon A-ring rearranged 5/6/6/6/5-pentacyclic nucleus, along with two biogenetically related known lupane-type triterpenoids (4 and 5), have been isolated from Daphniphyllum calycinum Benth. The determination of their structures was based on the extensive spectroscopic analysis, X-ray single crystal diffraction analysis, and chemical conversion. A plausible biosynthetic pathway for compounds 1-5 was proposed, involving a pinacol rearrangement, Wagner-Meerwein rearrangement, oxidation, and esterification.

Chronic inflammation of the tissues surrounding and supporting the teeth is known as periodontal disease. Human gingival fibroblasts (HGFs) are among the greatest prevalent cells in gingival tissue and play a crucial role in oral infections. Data from the GSE dataset revealed that the inflammatory cytokines IL-1β, IL-6, and IL-8 were up-regulated in periodontitis patients compared to healthy individuals. Betulin, a lupane-type pentacyclic triterpene alcohol extracted from the birch trees, inhibits lipopolysaccharide (LPS; component of gram-negative bacteria)-induced synthesis of IL-1β, IL-6, a

This study investigates the bioactive properties of different extracts of cardoon leaves in rescuing neuronal development arrest in an in vitro model of Rett syndrome (RTT). Samples were obtained from plants harvested at different maturity stages and extracted with two different methodologies, namely Naviglio® and supercritical carbon dioxide (scCO2). While scCO2 extracts more hydrophobic fractions, the Naviglio® method extracts phenolic compounds and less hydrophobic components. Only the scCO2 cardoon leaves extract obtained from plants harvested in spring induced a significant rescue of neur

BACKGROUND: Extensive research on Lupeol's potential in cancer prevention highlights its ability to target various cancer-related factors. It regulates proliferative markers, modulates signaling pathways, including PI3K/AKT/mTOR, and influences inflammatory and apoptotic mechanisms. Additionally, Lupeol demonstrates selectivity in killing cancer cells while sparing normal cells, thus minimizing the risk of toxic effects on healthy tissues.
HYPOTHESIS: Therefore, we aimed to explore Lupeol's potential roles as a chemotherapeutic agent and as a sensitizer to chemotherapy by reviewing various an

Lupeol is a lupane-type triterpene isolated from Sorbus commixta, an oriental medicine used to treat arthritis and inflammatory diseases. However, the antiosteoporotic effects of S. commixta or any of its constituents have not been studied yet. In the present study, we have examined the effect of lupeol (a major active triterpenoid isolated from S. commixta) on osteoclastogenesis and sought to elucidate its underlying molecular mechanisms. We evaluated whether lupeol antagonized osteoclast differentiation and bone resorption. Lupeol markedly inhibited osteoclast differentiation and bone resorp

Oleanolic acid (OA) and Lupeol (LU) belong to the class of natural triterpenes and are endowed with a wide range of biological activities, including cytotoxicity toward several cancer cell lines. In this context, we investigated a set of compounds obtained from the two natural precursors for the cytotoxicity against leukemia HL60 cells and the multidrug-resistant (MDR) variant HL60R. Six new semi-synthetic triterpenes have been synthetized, fully characterized, and were investigated together with other triterpenes compounds for their pharmacological mechanism of action. The interaction of the

Osteoarthritis (OA) is a widespread joint condition often linked to aging and obesity, resulting in pain, joint dysfunction, and disability. Betulin, a lupane-type pentacyclic triterpene alcohol extracted from birch trees, exhibits anti-inflammatory properties; however, its anti-inflammatory effects in OA remain largely unknown. Our high-throughput cytokine array data exhibit that betulin inhibits two key inflammatory factors, CHI3L1 and ICAM-1, in OA synovial fibroblasts (OASFs). Results from the GEO database and our clinical tissues confirm that CHI3L1 and ICAM-1 levels are markedly higher i

Betulin is a natural pentacyclic triterpenoid, possessing a lupane-structure, with a wide range of pharmacological activities. Its weak hydrosolubility hinders the biological activity of the compound and its derivatives. To circumvent this problem, we synthesized and tested in vitro three d-glycosaminosides of betulin. The structure of betulin was modified by incorporation of 2-amino-2-deoxy-d-gluco- and -d-galactopyranosyl moieties to its C-3 position. So far betulinyl glycosides containing these amino-sugars have not been reported in the literature. The structure of the studied derivatives w

Five districts in the Matabeleland region, an arid western area of Zimbabwe, were investigated for the status of Ca, P, Na, Cu and Zn in soil, forage and cattle during the wet and dry seasons over a period of one year. The cattle came from the natural grazing lands and were not supplemented at the time of sampling. Some deficiencies in soil Zn and P were found in the districts of Lupane and Bulilimamangwe, respectively. Dry season soil Ca, Cu and P concentrations were significantly higher (p < 0.05) than rainy season values owing to leaching in all five districts. Most forage samples had miner
