ROMP-based Glycopolymers with High Affinity for Mannose-Binding Lectins.

Well-defined, highly reactive poly(norbornenyl azlactone)s of controlled length (number-average degree of polymerization DPn¯ = 10 to 1,000) were made by ring-opening metathesis polymerization (ROMP) of pure exo-norbornenyl azlactone. These were converted into glycopolymers using a facile postpolymerization modification (PPM) strategy based on click aminolysis of azlactone side groups by amino-functionalized glycosides. Pegylated mannoside, heptyl-mannoside, and pegylated glucoside were used in the PPM. Binding inhibition of the resulting glycopolymers was evaluated against a lectin panel (Bc2

Mannose binding lectin gene polymorphism and the severity of chronic periodontitis.

OBJECTIVE: Mannose binding lectin (MBL) is a key molecule in first line defense against various microorganisms. Polymorphism of the MBL gene greatly affects serum MBL concentration, and is known to be associated with occurrence of infectious diseases. We aimed to determine whether there is a relationship between occurrence or severity of chronic periodontitis (CP) and polymorphism of the MBL gene. PATIENTS AND METHODS: Ninety-eight CP patients and 63 healthy subjects with no periodontitis were typed for the codon 54 polymorphism of the MBL gene by PCR-restriction fragment length polymorphism

Mannose-binding lectin as a risk factor for acute coronary syndromes.

BACKGROUND: Mannose-binding lectin (MBL) is a multifunctional protein involved in innate immunity. We tested whether MBL and elevated viral and bacterial antibodies were risk factors for acute coronary events. DESIGN: Controlled cohort study. METHODS: A total of 354 patients with unstable angina pectoris (UA) or acute myocardial infarction (AMI) were compared with 334 paired controls. RESULTS: Enterovirus titres were associated with increased risk of UA (odds ratio 10.04, P<0.001) and AMI (odds ratio 3.18, P=0.003), but titres did not correlate with either MBL concentration or genotype. Chl…

Interaction of human mannose-binding lectin (MBL) with Yersinia enterocolitica lipopolysaccharide.

The lipopolysaccharide (LPS) is involved in the interaction between Gram-negative pathogenic bacteria and host. Mannose-binding lectin (MBL), complement-activating soluble pattern-recognition receptor targets microbial glycoconjugates, including LPS. We studied its interactions with a set of Yersinia enterocolitica O:3 LPS mutants. The wild-type strain LPS consists of lipid A (LA) substituted with an inner core oligosaccharide (IC) which in turn is substituted either with the O-specific polysaccharide (OPS) or the outer core hexasaccharide (OC), and sometimes also with the enterobacterial comm…

Serum Mannose-Binding Lectin Levels in Asthma and Allergic Bronchopulmonary Aspergillosis.

Mannose-binding lectin (MBL) contributes to innate immunity against Aspergillus fumigatus. We assessed the role of reduced MBL levels in asthma patients with and without allergic bronchopulmonary aspergillosis (ABPA). In this prospective cross-sectional study, we enrolled 154 adults: 48 with asthma, 27 with Aspergillus-sensitized asthma (ASA, A. fumigatus-IgE ≥ 0.35 kUA/L), and 79 with ABPA. Serum MBL levels were quantified using enzyme-linked immunosorbent assay. Reduced MBL levels were defined using three thresholds (< 200, < 359, and < 500 ng/mL). The prevalence of reduced MBL levels did no…

Mannose-binding lectin gene (MBL-2) polymorphism in oral lichen planus.

TNF-α may be associated with the etiopathogenesis of oral lichen planus (OLP), and it has been suggested that polymorphism of mannose-binding lectin (MBL) increases the in vitro production of TNF- α. The aim of the present study was to assess the relevance of genetic diversity of MBL in OLP. The study sample comprised 90 individuals, 45 OLP patients and 45 healthy volunteers. MBL-2 gene was amplified using real-time PCR. Frequency of A/A genotype was 55.6% in OLP and 53.3% in healthy volunteers. Likewise, A/0 heterozygote genotype was found in 42.2% and 35.6%; 2.2% and 11.1%, had the recessive…

Association between mannose-binding lectin gene polymorphism and pediatric cytomegalovirus infection.

Mannose-binding lectin (MBL) is an important constituent of the human innate immune system, and can bind to a wide range of pathogens, including viruses such as influenza A, HIV, herpes simplex 2, and SARS-CoV. MBL deficiency results from single nucleotide polymorphisms (SNPs) in exon 1, and the promoter region of the human MBL2 gene has been found to be associated with susceptibility to a number of infections. However, studies on the interactions between MBL and CMV infection are limited. In this study, we investigated 104 children suffering from HCMV infection, in an effort to find any assoc…

Mannose-binding lectin genotypes and pre-eclampsia: a case-control study.

Both immunological and placental factors are involved in the pathogenesis of pre-eclampsia. The complement factor mannose-binding lectin (MBL) is associated with adverse pregnancy outcomes and has been suggested to play a role in abnormal placentation. We investigated whether MBL genotypes are associated with the systemic maternal syndrome pre-eclampsia. MBL2 gene polymorphisms were determined in a case-control study including 157 women with pre-eclampsia (case subjects) and 157 women with uncomplicated pregnancies (control subjects). Considering MBL polymorphisms, case and control subjects we…

Mannose-binding lectin and infection following allogeneic hemopoietic stem cell transplantation.

Major infection remains a major barrier to the success of allogeneic hemopoietic stem cell transplantation (SCT). There is growing interest in the importance of innate immunity in host defense, particularly when adaptive immunity is compromised. Furthermore, many host defense genes are polymorphic, and immunogenetic factors are known to influence the risk of other transplant complications, such as graft-versus-host disease. Mannose-binding lectin (MBL) has emerged as an important innate host defense molecule. MBL binds a wide range of pathogens independently of antibody and activates complemen…

Association of mannose-binding lectin 2 gene polymorphisms with Guillain-Barré syndrome.

Complement activation plays a critical role in the pathogenesis of Guillain-Barré syndrome (GBS), a debilitating immune-mediated neuropathy. Mannose-binding lectin (MBL) is a complement activation factor of lectin pathway which as genetic host factor may influence the susceptibility or severity of GBS. We investigated the frequency of MBL2 promoter (- 550H/L and - 221X/Y) and functional region (exon 1 A/O) polymorphisms and their association with disease susceptibility, clinical features and serum MBL among GBS patients (n = 300) and healthy controls (n = 300) in Bangladesh. The median patient…

Plasma levels of mannose-binding lectin and future risk of venous thromboembolism.

BACKGROUND: Animal and observational studies have suggested a pathophysiological role for complement in venous thromboembolism (VTE), but the initiating mechanisms are unknown. Mannose-binding lectin (MBL) bound to altered host cells leads to activation of the lectin complement pathway, and both high and low MBL levels have been implicated in the pathophysiology of cardiovascular disease. OBJECTIVES: To investigate the association between plasma MBL levels and future risk of incident VTE. METHODS: We conducted a nested case-control study in 417 VTE patients and 849 age-matched and sex-matche…

Role of mannose-binding lectin in nosocomial sepsis in critically ill neonates.

We investigated the association of mannose-binding lectin (MBL) serum levels with nosocomial sepsis (NS), their changes overtime during infection, their relation with pathogens, with the MBL2 genotype and their relationship with mortality. In a prospective observational study, we included 365 critically ill neonates: 261 had no infection and 104 had at least 1 septic event. The median MBL serum concentration was significantly lower in infected than in noninfected neonates (p < 0.001). Low MBL levels on admission increased the risk of infection, independently from gestational age and invasive p…

Meningococcal disease in a kidney transplant recipient with mannose-binding lectin deficiency.

We describe the case of a kidney transplant recipient who developed meningococcemia, without meningeal signs, 2 months after transplantation. Plasma levels of complement components C3, C4, and CH 50 were within the normal range. However, using a method to screen for the functional activity of all 3 pathways of complement, no activation via the mannose-binding lectin (MBL) pathway could be detected (0%). A subsequent quantification of MBL pathway components revealed normal levels of MASP 2 but undetectable amounts of MBL. To our knowledge, this is the first report of meningococcal disease after…

Association between mannose-binding lectin levels and graft survival in kidney transplantation.

The mannose-binding lectin (MBL) pathway of complement is activated by pattern recognition. Genetic MBL variants are frequent and associated with low MBL serum levels. Higher MBL levels may be associated with more complement-mediated damage resulting in inferior graft survival. Pre-transplant serum samples from 266 consecutive deceased donor kidney transplant recipients were analyzed for MBL concentration by ELISA. Subsequently the cohort was analyzed for transplant-related outcome. There was no significant difference in incidence of delayed graft function in recipients with a low MBL level (<…

Ultraviolet-B recruits mannose-binding lectin into skin from non-cutaneous sources.

Mannose-binding lectin (MBL) is an integral part of the innate immune system and functions as an opsonin by binding to pathogens and certain apoptotic cells to promote their uptake by phagocytes. We recently identified an association of low-producing MBL polymorphisms with adult dermatomyositis (DM). Our model is that MBL deficiency leads to a defect in the clearance of apoptotic debris in the skin, thereby predisposing to photosensitive autoimmune disease. In this study, we sought to determine whether MBL binds within the epidermis, and to determine its source, and potential function of this …

[Acute respiratory tract infections and mannose-binding lectin insufficiency in small children].

INTRODUCTION: According to hospital-based studies, increased susceptibility to certain infections is associated with genotypes that cause low serum levels of the protein mannose-binding lectin (MBL). However, the contribution of MBL insufficiency to the incidence of common childhood infections on a population basis is unknown. To investigate the effect of MBL insufficiency on the risk of acute respiratory infections (ARI) in unselected children, we performed a prospective population-based study of ARI in young children in Sisimiut, Greenland. MATERIAL AND METHODS: An open cohort of children a…
