Epidemiology of Mansonella perstans in the middle belt of Ghana.

BACKGROUND: Mansonellosis was first reported in Ghana by Awadzi in the 1990s. Co-infections of Mansonella perstans have also been reported in a small cohort of patients with Buruli ulcer and their contacts. However, no study has assessed the exact prevalence of the disease in a larger study population. This study therefore aimed to find out the prevalence of M. perstans infection in some districts in Ghana and to determine the diversity of Culicoides that could be potential vectors for transmission. METHODS: From each participant screened in the Asante Akim North (Ashanti Region), Sene West a…

Epidemiologic survey of Mansonella ozzardi in Corail, Haiti.

An epidemiologic (cross-sectional study) survey on 462 inhabitants in Corail, Haiti showed that 16.5% were infected with Mansonella ozzardi. This finding was determined from a single 20-μL sample of finger prick blood from each person. Among children, < 2% had a detectable microfilaremia. In persons > 15 years of age, the prevalence of infection for males and females was 23% and 21%, respectively. In general, the microfilaremias were low and 70% of positive persons had < 10 microfilariae per 20 μL of blood; only 5% had > 50 microfilariae. This study shows that persons living near mangrove mars…

Mansonella ozzardi in Haiti II. Arthropod vector studies.

The present vector studies show that the biting midge, Culicoides furens, serves as an intermediate host for Mansonella ozzardi in Haiti. A total of 3,430 C. furens were collected as they engorged on infected individuals. Under the maintenance conditions described herein, the microfilaria required 9 days to reach the infective stage. Approximately 43% of the midges survived, and from these 1,128 infective stage larvae were collected. This represents an overall infection rate of 0.76 larva per midge. Typically, there was a directly proportional relationship between the infection rate in midges …

Zoonotic Mansonella ozzardi in Raccoons, Costa Rica, 2019-2022.

Mansonella ozzardi, a filarioid parasite, causes human mansonellosis in the Americas. We identified raccoons (Procyon lotor) as wildlife reservoirs of M. ozzardi in Costa Rica. Noting the sympatry of free-ranging raccoons and humans, we conclude that mansonellosis is a considerable public health risk in the region.

High Prevalence of Mansonella perstans Filariasis in Rural Senegal.

Large parts of African and American countries are colonized by Mansonella, a very common but poorly described filarial nematode. Bloodsucking flies of the genus Culicoides are suspected to be the vector of Mansonella perstans, but no study in Senegal has confirmed that Culicoides can transmit the parasite. Designed specific real-time quantitative polymerase chain reaction (qPCR) can be used to identify microfilaria in stained blood smears. This study was performed in July and December 2010 in the southeastern Senegal, which is known to be endemic for M. perstans. We analyzed 297 blood smears f…

Mansonella perstans causing symptomatic hypereosinophilia in a missionary family.

Mansonella perstans is rarely pathogenic. The rare reports of symptomatic cases, however, include severe complications. Three cases of symptomatic hypereosinophilia with multi-organ involvement are described in a missionary family returning from tropical Africa. Pathogenicity may be related to the induction of hypereosinophilia rather than direct host-parasite interactions.

Efficacy of repeated doses of ivermectin against Mansonella perstans.

Treatment of Mansonella perstans infection, although seldom necessary, is difficult. In a 3 year's trial of normal and high-dose annual and 3-monthly ivermectin treatment against Onchocerca volvulus, the effects on M. perstans were recorded and related to the cumulative dose received. The World Health Organization's African Programme for Onchocerciasis Control may thus reduce the endemicity of M. perstans.

[Mansonella perstans isolated on aspiration puncture of a salivary gland].

Chronic parasitosis due to nematode worms (filariae) in tissue are very common in tropical ecosystems; their larvae (microfilariae) have been isolated in lymph vessels, skin, and blood. The case reported here is of a Nigerian patient suffering severe renal failure and admitted owing to the presence of a right submandibular gland abscess. In the FNAP, the presence of a Mansonella perstans microfilaria was identified. Post-surgery examination of pathology samples from the gland reported an acute inflammatory infiltrate including abundant eosinophils and microfilariae. There are some reports in t…

Development of Mansonella perstans in the surrogate vector Aedes aegypti.

The biology of the human filaria Mansonella perstans has been poorly studied due in part to the lack of experimental animal models in which its life-cycle could be reproduced. In nature Culicodes spp. (and probably simuliids) are the vectors involved in disease transmission. In the present work Aedes aegypti black-eyed strain was experimentally infected with M. perstans microfilariae by intrathoracic inoculation of 8 to 15 parasites contained in 0.4-0.6 microliters of RPMI 1640. Concentration of microfilariae was achieved by means of a ficoll separation technique. A. aegypti were maintained at…

Adult Mansonella perstans in the abdominal cavity in nine Africans.

Adult Mansonella perstans infected the abdominal cavity of nine patients seen at Karawa Hospital in the Ubangi territory of Zaire. In four patients the worms were removed at laparotomy, and in the other five they were removed at autopsy. Twelve adult worms were identified in the nine patients. None of the worms caused symptoms or contributed to the patient's death. Worms were in the hernial sac in three patients, and one each was in connective tissue beside a reactive mesenteric lymph node, in peripancreatic connective tissue, in perirenal connective tissue, in hepatic portal connective tissue…

Assessment of Culicoides barbosai as a vector of Mansonella ozzardi in Haiti.

Studies in the southern peninsula of Haiti showed that the biting midge, Culicoides barbosai, was capable of supporting the development of Mansonella ozzardi to the infective stage. The known vector, Culicoides furens, also was encountered. Both species showed distinct biting site preferences, i.e., 98% of the midges that engorged on the arms and head were C. barbosai whereas C. furens was collected mostly from the lower legs. Nine days after engorgement, 19 infective larvae were recovered from 13 C. barbosai versus six larvae from four C. furens. It was judged that C. barbosai may be as impor…

Mansonella ozzardi: a redescription with comments on its taxonomic relationships.

Mansonella ozzardi is redescribed from adult worms collected from the subcutaneous tissues of patas monkeys (Erythrocebus patas) infected with a Haitian strain of the filaria. The worms are small and very slender; females measure about 49 mm in length by 0.15 mm in diameter and males, 26 by 0.07 mm. Haitian (Caribbean) and Colombian (Amazon) forms of the filaria are morphologically identical, as are their microfilariae. Mansonella is most closely related to Tetrapetalonema. Based on taxonomic priority the latter becomes a synonym of Mansonella. As a consequence, T. perstans and T. streptocerca…

Identification and characterisation of Mansonella perstans in the Volta Region of Ghana.

Mansonella spp. have been reported to have a wide global distribution. Despite the distribution and co-occurrence with other filarial parasites like Wuchereria bancrofti, Onchocerca volvulus and Loa loa, it is given little attention. There are few surveillance programmes for assessing the distribution of mansonellosis, due to the associated mild to no symptoms experienced by infected people. However, addressing this infection is critical to the onchocerciasis control program as current rapid diagnostic tools targeting O. volvulus have the tendency to cross react with Mansonella species. In thi…

Improvement of a PCR test to diagnose infection by Mansonella ozzardi.

INTRODUCTION: Mansonelliasis is caused by Mansonella ozzardi. It is widespread in the Amazon region, with a high prevalence. The common exam of thick blood smears stained with Giemsa shows low efficacy levels and has been an obstacle to diagnosing individuals with low blood parasitemia. METHODS: In order to increase diagnosis efficacy, the PCR technique was improved. RESULTS AND CONCLUSIONS: PCR demonstrated the best performance, with sensitivity and negative predictive values (NPV) of 100%, followed by blood filtration through membrane filters, which showed a sensitivity of 88.9% and a NPV …

Analysis of the 24-h microfilarial periodicity of Mansonella perstans.

The Mansonella perstans microfilarial (mf) periodicity in peripheral blood was analysed in a group of 32 healthy individuals from an endemic community in Uganda. The majority of individuals had maximum mf intensity during the first and minimum during the last 12 h of the day. Mean mf ratios (i.e. time-specific counts in relation to mean count for the individual) indicated a weak but significant diurnal periodicity with peak intensity around 0800 h. Trigonometric analysis of the ratios, assuming a harmonic wave pattern of periodicity, gave a periodicity index of 11.1 and a peak time of 0645 h. …

No evidence of Wolbachia endosymbiosis with Loa loa and Mansonella perstans.

Endosymbiotic Wolbachia bacteria from different filarial species, including major pathogens of humans such as Wuchereria bancrofti, Brugia malayi and Onchocerca volvulus, seem to play an important role in the development, viability and fertility of these worms. Wolbachia trigger inflammatory host responses as well as adverse reactions against standard treatment regimens and are therefore under investigation as novel treatment targets. We investigated whether Wolbachia are also endosymbiotic in Loa loa and Mansonella perstans. In both male and female adult L. loa, we found no evidence of bacter…
