Difference in effect of sodium selenite on mercury distributions after duodenal administration of mercuric chloride and mercuric oxide.

To obtain evidence for difference in the absorptive forms of HgCl2 and HgO from duodenum, the Hg distributions after the duodenal administration of HgCl2 and HgO and the effect of Na2SeO3 on their Hg distributions were compared. The Hg concentrations in erythrocytes and liver 2 hr after the administration of HgCl2 were significantly lower and higher than those of HgO, respectively, but their Hg distributions at 3.5 or 5 hr became identical. The administration of Na2SeO3 (intravenously) 2 hr after the administration of HgCl2 and HgO resulted in significant differences in their Hg concentrations

Hemolytic effects of sodium selenite and mercuric chloride in human blood.

Many works have reported the interaction between selenium and mercury in the mammalian body and that chalcogen seems to have a protective effect against mercury toxicity. The aim of this study was to investigate the hemolytic effects of sodium selenite and/or mercuric chloride in human blood under in vitro conditions. For this, total venous blood from healthy subjects (males and females) was heparinized and incubated at 37 degrees C for 90 min with different concentrations of sodium selenite and/or mercuric chloride. The hemolytic effects of compounds were evaluated by measuring plasma hemoglo

Lipid peroxidation in liver of rats administrated with methyl mercuric chloride.

Parenteral administration of methyl mercuric chloride (MMC, CH3HgCl) to rats enhanced lipid peroxidation in liver of rats, as measured by the thiobarbituric acid reaction for malondialdehyde (MDA) in fresh tissue homogenates. After sc injection of CH3HgCl (5 mg/kg body wt), MDA concentration in liver became significantly increased at 24 h and further increased at 48 h. Dose-response studies were carried out with male albino rats of the Fisher-344 strain (body wt 170-280 g) injected with 3 or 5 mg Hg/kg as CH3HgCl and sacrificed after 24 h. In time-response studies, animals were administered 5

Comparison of the interaction of methyl mercury and mercuric chloride with murine macrophages.

The toxicity of organic methyl mercury was studied on murine macrophages in cell culture and compared to that of inorganic mercuric chloride. Long-term treatment of macrophage cultures with methyl mercury resulted in decreased cell viability in a concentration-dependent fashion. Experiments showed that 20 microM methyl mercury was highly toxic, causing cell death within a few days, while cultures exposed to lower levels were less severely affected. Comparison of the toxicity of organic and inorganic mercury by cell viability showed no difference between equimolar concentrations of methyl mercu

Effects of methylmercury and mercuric chloride on preimplantation mouse embryos in vivo.

This report compares the effects of methylmercuric chloride (MMC) and mercuric chloride (MC) on the development of mouse preimplantation embryos in vivo. Female mice were injected with a single intravenous dose of 0.5-20.0 mg Hg/kg MMC or 0.5-2.5 mg Hg/kg MC on day 0 of gestation. The embryos were recovered by flushing excised oviduct and uterus on day 3.5 of pregnancy, and were examined for abnormalities. In the groups treated with doses of 0.5 and 1.0 mg Hg/kg of both compounds, the rates of abnormal embryos were not significantly different from that in the control group. The 50% effective d

Renal function and mercury level in rats with mercuric chloride nephrotoxicity.

The levels of Hg++ in different subcellular compartments of rat kidney cortex were determined after a single subcutaneous injection of 203HgCl2, 4.7 mg/kg body weight. By 30 min after injection, cortical Hg++ level was 46.3 +/- 6.8 micrograms/g protein. A maximal cortical Hg++ value of 429 +/- 49 micrograms/g protein was reached 4 h after injection. Hg++ associated with the brush border membrane showed a progressive increase during the first 15 h after injection, but at each time interval was significantly less than cortical Hg++. The 45-hour values for cortex and brush border were equivalent.

Glomerulonephritis with - and 1C-globulin deposits induced in rats by mercuric chloride.

Repeated subcutaneous injections of 0.15 or 0.25 mg/100 g of body weight of HgCl(2) may induce a membranous glomerulonephritis in rats. This glomerulonephritis with subepithelial deposits is similar, by electron microscopy and immunohistochemistry, to experimental chronic glomerulonephritis caused by immune complexes and to the human membranous glomerulonephritis.

Effect of mercuric chloride on various hydroxyproline fractions in rat serum.

Mercuric chloride (HgCl2) disturbs the collagen metabolism in the body which is reflected by altered hydroxyproline fractions in the serum. The aim of the present investigation was to study the effect of HgCl2 treatment on various hydroxyproline (Hyp) fractions in rat serum and the effect of 2,3-dimercapto-1-propane sulfonic acid (DMPS) treatment on serum Hyp fractions in HgCl2 treated rats. Other parameters studied included body weight, food intake, water intake and kidney weight. Doses of HgCl2 used were 0.1, 0.5, 1.0, 2.0, 3.0 mg/kg body weight and that of DMPS was 100 mg DMPS/kg body weigh

In vitro effects of mercuric chloride (HgCl2) on human mononuclear cells.

Due to the release of the toxic compounds of mercury from amalgam fillings, dental amalgam has been questioned as an adequate restoration material for tooth fillings. HgCl2 has been found to be mitogenic for human blood lymphocytes in vitro. However, activation required much higher concentrations than are ever found in vivo. This study has been initiated to evaluate further the influence of HgCl2 on human immunocompetent cells in vitro. It is found that HgCl2 in a narrow concentration range has the ability to preferentially stimulate the CD4+ T cell subset to blast transformation and DNA synth

Mercuric chloride-induced tubulonecrosis in the rat kidney: the recovery phase.

Regeneration of the renal tubules in rats, after death of the cells of the pars recta caused by a dose of 1.5 mg HgCl2/kg, was examined by histological and radioautographic methods. The tubules regenerated from surviving squamoid cells at both ends of the necrotic segments, which often appeared to arise from epithelial cells that had cast off superficial "dead cytoplasm" to leave the basal parts containing the nucleus in a rim of cytoplasm. The tubular epithelium was reconstituted between the 2nd and 5th days by proliferation and sliding extension of the squamoid cells along the tubules and pr

Demonstration of antinuclear antibodies in mercuric chloride-induced glomerulopathy in the rat.

Serial administration of mercuric chloride to PVG/C rats induced a glomerulopathy associated with immune complex deposition along the glomerular basement membrane and in the mesangial area. These deposits could be demonstrated by immunofluorescence and electron microscopy 5 to 8 weeks after the first injection of mercuric chloride. At this time antinuclear antibodies could be demonstrated in the sera and in the eluates from the renal cortices from diseased animals. These findings suggest a possible role for antibodies directed against nuclear antigen in the pathogenesis of this type of experim

The mechanism of action of mercuric chloride on the isolated perfused rat kidney.

Studies on 80 rat kidneys, perfused at constant flow, showed that mercuric chloride produced a marked increase in perfusion pressure within five minutes of administration. Adrenergic blocking agents (phentolamine and propranolol), angiotensin sensitivity depletion, bradykinin and low concentrations of mannitol had no effect on this increase in resistance. Perfusion of the kidney with 5 percent mannitol solution, however, significantly reduced the increase of vascular resistance induced by mercuric chloride. Evidence is presented that mercuric chloride may evoke an increase in vascular resistan

Differential effects of methylmercuric chloride and mercuric chloride on oxidation and iodination reactions catalyzed by thyroid peroxidase.

Thyroid peroxidase (TPO), the major enzyme in the thyroid hormone synthesis, multifunctionally catalyzes (1) iodide oxidation, (2) iodination of the precursor protein, and (3) a coupling reaction of iodotyrosyl residues. The present study was carried out to examine the mercurial effects on the iodination, the second step of TPO. Purified porcine thyroglobulin or bovine serum albumin as acceptor protein was iodinated with [125I]NaI and H2O2 by purified porcine TPO. Iodinated protein was separated by acid precipitation on membrane filter or paper chromatography. Both CH3HgCl and HgCl2 dose-depen

Lotus seedpod proanthocyanidins protect against neurotoxicity after methyl-mercuric chloride injury.

In the present study, to investigate the prevention mechanism of proanthocyanidins from lotus seedpod (LSPCs) on methyl mercuric chloride (MMC) induced neurotoxicity, neuron/astrocyte cells were co-cultured to simulate the microenvironment in vivo to the greatest extent. The results showed that, compared with MMC group, pretreatment with LSPCs not only improved cell survival rate, decreased the release of lactate dehydrogenase (LDH), decreased the intracellular reactive oxygen species (ROS) level, and prevented the increase of intracellular [Ca2+]i, but also significantly increased the total a

Anti-phospholipid antibodies in the mercuric chloride treated brown Norway rat.

Anti-phospholipid antibodies (aPL), in the form of anti-cardiolipin antibodies (aCL) and/or lupus anticoagulant (LAC), are found in a number of disorders, including systemic lupus erythematosus. Autoimmune aPL are associated with clinical manifestations that may include vascular thrombosis, recurrent fetal loss, thrombocytopenia, livedo reticularis and neurological abnormalities. aPL found in the context of infections such as syphilis are not usually associated with clinical complications. Here we report the presence of aCL in Brown Norway (BN) rats treated with mercuric chloride (HgCl2), whic
