The small G protein H-Ras in the mesolimbic system is a molecular gateway to alcohol-seeking and excessive drinking behaviors.

Uncontrolled consumption of alcohol is a hallmark of alcohol abuse disorders; however, the central molecular mechanisms underlying excessive … ether, these results position H-Ras as a central molecular mediator of alcohol's actions within the mesolimbic system and put forward the potential value of the enzyme as a novel target to treat alcohol use disorders.

Physical Exercise Promotes Beneficial Changes on Neurotrophic Factors in Mesolimbic Brain Areas After AMPH Relapse: Involvement of the Endogenous Opioid System.

Addiction is a serious public health problem, and the current pharmacotherapy is unable to prevent drug use reinstatement. Studies have focused on physical exercise as a promising coadjuvant treatment. Our research group recently showed beneficial neuroadaptations in the dopaminergic system related to amphetamine-relapse prevention involving physical exercise-induced endogenous opioid system activation (EXE-OS activation). In this context, additional mechanisms were explored to understand the exercise benefits on drug addiction. Male rats previously exposed to amphetamine (AMPH, 4.0 mg/kg) for

Neurofilament proteins and the mesolimbic dopamine system: common regulation by chronic morphine and chronic cocaine in the rat ventral tegmental area.

The ventral tegmental area (VTA) and its dopaminergic projections appear to mediate some of the rewarding properties of opiates, cocaine, and other drugs of abuse. In a previous study, we demonstrated that chronic morphine and cocaine exert common actions on tyrosine hydroxylase, the rate-limiting enzyme in catecholamine biosynthesis, in this dopaminergic brain reward region (Beitner-Johnson and Nestler, 1991). In the present study, we investigated the effects of chronic morphine and cocaine on other phosphoproteins in the VTA by back phosphorylation and two-dimensional electrophoretic analysi

Sex differences in alcohol and amphetamine-like compound consumption and their relationship with mu opioid receptor density in the pallidal-mesolimbic system.

Excessive alcohol (ETOH) consumption is associated with alterations in the opioid system, including β-endorphin deficiency and differential expression of the mu opioid receptor (MOR) in brain areas involved in reward. This system also contributes to the rewarding properties of psychostimulants, such as cocaine and amphetamines; however, its role in this regard has been little studied, particularly with respect to sex differences. The aim of this study was to determine whether male and female Wistar rats with high ETOH consumption also exhibit higher intake of the psychostimulant phentermine (P

Prenatal Methyl Nutrient Availability Shapes Mesolimbic Dopaminergic Circuitry and Systemic Inflammation in Wistar Rat Offspring of Both Sexes.

The dopaminergic system constitutes a principal element of the reward neurocircuitry and is responsive to influences encountered early in life. Methylation potential, shaped by nutrients involved in one-carbon metabolism, intersects with immune signaling and dopaminergic receptor, creating a broader metabolic network through which early life nutrition may alter neurobehavioral outcomes. We have shown that methyl nutrient imbalance between folic acid and choline during pregnancy produces offspring with higher body weight and perturbed methylation potential. However, whether these effects reflec

Investigation of the impact of rs3025343 near the dopamine ß-hydroxylase (DBH) gene on reward-related responses within the mesolimbic dopamine system.

OBJECTIVES: Recent research focused on the impact of Neandertal alleles on dopamine-related brain traits. A joint measure of these alleles was reported to be associated with lower dopamine synthesis, presumably leading to reduced activity of the reward system. To isolate single variants, we selected rs3025343 near the dopamine beta hydroxylase gene, which contains a likely introgressed allele. rs3025343 is an established variant for reward system-related psychiatric traits. METHODS: We hypothesised that the rs3025343 minor allele reduces responses to conditioned reward stimuli in the mesolimb

Sex-specific alterations in CRF and dopaminergic markers in the lateral septum and mesolimbic system following chronic high-fat diet exposure in rats.

BACKGROUND: Obesity is a global health concern closely associated with sedentary lifestyles and the consumption of hypercaloric, hyperpalatable diets. While these factors contribute to excessive food intake and weight gain, the long-term impact of obesogenic diets on neural circuits regulating energy balance remains insufficiently understood. Previous research has identified the lateral septum (LS) as a critical structure involved in stress responses and addiction, with corticotropin-releasing factor (CRF) and dopaminergic signalling playing key roles. However, the influence of chronic high fa

Involvement of the K+-Cl- co-transporter KCC2 in the sensitization to morphine-induced hyperlocomotion under chronic treatment with zolpidem in the mesolimbic system.

Benzodiazepines are commonly used as sedatives, sleeping aids, and anti-anxiety drugs. However, chronic treatment with benzodiazepines is known to induce dependence, which is considered related to neuroplastic changes in the mesolimbic system. This study investigated the involvement of K(+) -Cl(-) co-transporter 2 (KCC2) in the sensitization to morphine-induced hyperlocomotion after chronic treatment with zolpidem [a selective agonist of γ-aminobutyric acid A-type receptor (GABAA R) α1 subunit]. In this study, chronic treatment with zolpidem enhanced morphine-induced hyperlocomotion, which is …

Time-dependent increases in brain-derived neurotrophic factor protein levels within the mesolimbic dopamine system after withdrawal from cocaine: implications for incubation of cocaine craving.

Using a rat model of drug craving, we found that the responsiveness to cocaine cues progressively increases or incubates over the first 60 d of cocaine withdrawal. Here we studied whether alterations in brain-derived neurotrophic factor (BDNF) protein levels within the mesolimbic dopamine system are associated with this incubation phenomenon. BDNF is involved in synaptic plasticity and was found to enhance responding for cues associated with natural rewards. Rats were trained to press a lever to receive intravenous cocaine or oral sucrose for 6 hr/d for 10 d; each earned reward was paired with

Effects of MC-100093, GLT-1 enhancer, in mesolimbic brain regions of C57BL/6 mouse exposed to hydrocodone high-dose challenge using comprehensive laboratory monitoring system.

Chronic exposure and overdose of opioid cause neurotoxicity through alterations of several neurotransmitters, including glutamate. Upregulating glutamate transporter 1 (GLT-1) has been shown to attenuate the behavioral and neurochemical effects of chronic exposure to drugs of abuse. This study investigated the effects of GLT-1 enhancers-MC-100093 and ceftriaxone-on locomotor activity, respiratory exchange, and glutamate transporters (GLT-1, cystine/glutamate transporter, xCT), brain derived neurotrophic factor (BDNF), and neuroinflammatory biomarkers (TNF-α and HMGB1) in nucleus accumbens (NAc

Baseline Functional Connectivity of the Mesolimbic, Salience, and Sensorimotor Systems Predicts Responses to Psychological Therapies for Chronic Low Back Pain With Comorbid Depression: A Functional MRI Study.

INTRODUCTION: Chronic low back pain (CLBP) is a prevalent and debilitating condition. Cognitive behavioral therapy (CBT) can improve coping mechanisms for CLBP and pain-related outcomes. However, the mechanisms by which they do so remain undetermined. We explored the neural correlates of CLBP symptoms and CBT action using functional magnetic resonance imaging (fMRI) in women with CLBP and comorbid depression. METHODS: Forty individuals underwent fMRI followed by 8 weeks of either treatment as usual (TAU) or one of two CBT in addition to TAU: acceptance and commitment therapy (ACT) or behavior

Dopamine and schizophrenia.

The antipsychotic actions and extra-pyramidal side-effects of neuroleptic drugs are strongly correlated with their ability to block central dopaminergic transmission. It is argued that the former are more closely related to actions on dopaminergic mechanisms in the "mesolimbic dopamine" system, and the latter to similar actions in the striatum. Although the amphetamine psychosis closely resembles paranoid schizophrenia and may be due to excess dopamine release, clinical, biochemical, and endocrine studies suggest that dopaminergic overactivity is not a necessary concomitant of schizophrenic il

A morphometric study of the dopamine-containing cell groups in the mesencephalon of normals, Parkinson patients, and schizophrenics.

A quantitative study of the melanin-containing dopaminergic neurons of the nigrostriatal system (A 9) and of the mesolimbic system (A 10) was carried out on Nissl-stained serial sections of nine normal brains, six age-matched brains of schizophrenics, five brains of paralysis agitans, and six brains of postencephalitic parkinsonism. By contrast with most laboratory animals the A 10 cell group is not well developed in the human brain. Both in Parkinson's disease with a known hypoactivity of dopamine neurons and in schizophrenia with a postulated hyperfunction of these systems, pathological alte…

NMDA Receptors in Accumbal D1 Neurons Influence Chronic Sugar Consumption and Relapse.

Glutamatergic input via NMDA and AMPA receptors within the mesolimbic dopamine (DA) pathway plays a critical role in the development of addictive behavior and relapse toward drugs of abuse. Although well-established for drugs of abuse, it is not clear whether glutamate receptors within the mesolimbic system are involved in mediating chronic consumption and relapse following abstinence from a non-drug reward. Here, we evaluated the contribution of mesolimbic glutamate receptors in mediating chronic sugar consumption and the sugar-deprivation effect (SDE), which is used as a measure of relapse-l…

Retrieval Demands Adaptively Change Striatal Old/New Signals and Boost Subsequent Long-Term Memory.

The striatum is a central part of the dopaminergic mesolimbic system and contributes both to the encoding and retrieval of long-term memories. In this regard, the co-occurrence of striatal novelty and retrieval success effects in independent studies underlines the structure's double duty and suggests dynamic contextual adaptation. To test this hypothesis and further investigate the underlying mechanisms of encoding and retrieval dynamics, human subjects viewed pre-familiarized scene images intermixed with new scenes and classified them as indoor versus outdoor (encoding task) or old versus new…

A dimmer switch for reward: the vagus sets the gain.

The vagus nerve, among its various functions, carries gut nutrient signals to brain reward circuits. In a recent study, Onimus and colleagues have shown it is more than a simple relay: vagal integrity is required for maintaining dopamine release and the circuit structure of the mesolimbic system to mount reward responses. These findings reveal the vagus as a tonic gatekeeper of motivation and reward.

Glycine receptors regulate dopamine release in the rat nucleus accumbens.

BACKGROUND: The mesolimbic dopamine (DA) system seems to be centrally involved in regulating reward-related behavior and consequently has been implicated in addictive processes, such as alcoholism and drug addiction. This DA system has also been implicated in psychosis and in regulating hedonia/anhedonia, important components of mania and depression. Given the potentially great importance of the mesolimbic DA system for several psychiatric disorders, it is of major interest to delineate the mechanisms and dynamics underlying DA regulation and release. Recently strychnine-sensitive glycine rece
