The globular to fibrillar transition of proteins represents a key pathogenic event in the development of amyloid diseases. Although systemic amyloidoses share the common characteristic of amyloid deposition in the extracellular matrix, they are clinically heterogeneous as the affected organs may vary. The observation that precursors of amyloid fibrils derived from circulating globular plasma proteins led to huge efforts in trying to elucidate the structural events determining the protein metamorphosis from their globular to fibrillar state. Whereas the process of metamorphosis has inspired poe

Clarifying the mechanisms underlying shape alterations during insect metamorphosis is important for understanding exoskeletal morphogenesis. The large horn of the Japanese rhinoceros beetle Trypoxylus dichotomus is the result of drastic metamorphosis, wherein it appears as a rounded shape during pupation and then undergoes remodeling into an angular adult shape. However, the mechanical mechanisms underlying this remodeling process remain unknown. In this study, we investigated the remodeling mechanisms of the Japanese rhinoceros beetle horn by developing a physical simulation. We identified th

The effect of procercoids of T. nodulosus on the periods of metamorphosis, fecundity, survival and the capacity to infect C. strenuus at all copepodid stages (I-V) was studied experimentally. It has been first shown that early copepodid stages (I-III) can be easily infected and serve as first intermediate hosts of T. nodulosus. Under the effect of the parasite the metamorphosis periods of the I and 2-nd copepodid stages increase considerably even at the low infection intensity while at the IV and V copepodid stages--only at the high infection intensity. If the infection takes place at the IV c

Beyond the well-established roles in biomedical processes, extracellular vesicles (EVs) from Gram-negative bacteria are now identified as potent inducers of larval metamorphosis in marine invertebrates, a critical process for sustaining the health and function of global marine ecosystems. However, the specific inductive molecules within EVs remain largely unknown. Our findings reveal that cytochrome c (CytC), a previously unrecognized chemical signal packaged within EVs, induces larval metamorphosis. CytC maturation is essential for this process, as it enhances the efficiency of electron trans

The Mexican axolotl (Ambystoma mexicanum) is an established model for studying tetrapod regeneration and development. Notably, axolotls exhibit remarkable brain regeneration as adults, a trait rarely observed in other adult vertebrates. Adult axolotls can undergo metamorphosis, a process that induces dramatic remodeling of multiple organs and is accompanied by a gradual decline in regenerative capacity and lifespan. However, systematic studies on whole-brain cellular dynamics and molecular mechanisms in both adult and metamorphosed individuals remain lacking. Here, we profiled five representat

Gregarines are symbionts of the gut or other body cavities associated with numerous insect species. Knowledge about natural gregarine infection levels and across metamorphosis in holometabolous hosts is sparse. Individuals of the mustard leaf beetle (Phaedon cochleariae, Coleoptera: Chrysomelidae) were collected from eight populations and dissected to estimate the prevalence and load of infection with gregarines. In the laboratory, the effect of larval density (single individuals versus groups of five) on subsequent larval gametocyst load and adult infection with Gregarina cochlearium was test

Spodoptera litura commonly known as the cutworm, is among the most destructive lepidopteran pests affecting over 120 plants species. The powerful destructive nature of this lepidopteran is attributable to its high reproductive capacity. The testicular fusion that occurs during metamorphosis from larvae to pupa in S.litura positively influences the reproductive success of the offspring. In contrast, Bombyx mori, the silkworm, retains separate testes throughout its life and does not undergo this fusion process. Microscopic examination reveals that during testicular fusion in S.litura, the perito

Temperature is one of the important factors affecting the growth, development, and metamorphosis of amphibians. Endochondral ossification during metamorphosis plays a crucial role in amphibian survival and adaptation on land. In this study, we explored the effects of different temperature treatments on the growth, development, and endochondral ossification of Rana chensinensis tadpoles during metamorphosis. The results showed that high temperature exposure may affect the skeletal development of tadpoles during metamorphosis, such as reduction of bone length and ossification of limbs, thyroid g

Nitrite, as a part of nitrogen cycle, is one of the most common toxic compounds in aquatic ecosystems. Since skeletal development is an essential process during amphibian metamorphosis, exposure of larval amphibians to nitrite might disrupt skeletal development. To evaluate whether nitrite affects skeletal development of amphibian larvae, Bufo gargarizans larvae at Gs26 were exposed to 10, 100, 500 and 1000 μg/L nitrite-nitrogen (NO2-N) in the present study. The metamorphosis rate, body weight, body length, forelimb length and hindlimb length of B. gargarizans exposed to NO2-N were decreased.

Discovery of elevated concentrations of cadmium in the natural environment has increased awareness because of their potential threats. Amphibians are negatively affected due to their moderate sensitivity to cadmium. Here, we conduct acute and subchronic toxicity tests to examine whether, and to what extent, cadmium exposure disturbs metamorphosis, growth, and kinetic ability of Rana zhenhaiensis. We set different concentration treatment groups for the subchronic toxicity test (0, 10, 40, 160 μg Cd L-1). Our findings demonstrate that cadmium exposure reduces growth parameters and the cumulative

The amphibian metamorphosis assay (AMA; US Environmental Protection Agency [USEPA] test guideline 890.1100 and Organisation for Economic Co-Operation and Development test guideline 231) has been used for more than a decade to assess the potential thyroid-mediated endocrine activity of chemicals. In 2013, in the context of the Endocrine Disruptor Screening Program of the USEPA, a Scientific Advisory Panel reviewed the results from 18 studies and recommended changes to the AMA test guideline, including a modification to a fixed-stage design rather than a fixed-time (i.e., 21-d) design. We descri

For marine benthic animals, the selection of substrate by larvae is important for their survival, with early post-settlement mortality often being affected by the microenvironment where they settle. We tested the substrate preference of the larvae of the ascidian Phallusia philippinensis toward nine commercially available substrates. In the present assay, the larvae settled on one of four substrates for seven substrate combinations with different wettabilities; we counted the number of settled larvae on each of the four substrates, and Manly's selection indices were compared to determine the p

The hypothesis here presented tries to explain why organ regeneration is present in fish and amphibians (anamniotes) but is absent in reptiles, birds and mammals (amniotes). Anamniotes possess complex life cycles including larvae and metamorphosis stages, the latter representing a physiological form of organ destruction and regeneration coded in their genome that can be reactivated in adults in the form of regeneration. Part of the genome for larvae and metamorphosis phases was likely lost in amniotes with the evolution of direct development, the potentiation of the immune system and the incre

The Porifera (sponges) is one of the earliest phyletic lineages to branch off the metazoan tree. Although the body-plan of sponges is among the simplest in the animal kingdom and sponges lack nervous systems that communicate environmental signals to other cells, their larvae have sensory systems that generate coordinated responses to environmental cues. In eumetazoans (Cnidaria and Bilateria), the nervous systems of larvae often regulate metamorphosis through Ca(2+)-dependent signal transduction. In sponges, neither the identity of the receptor system that detects an inductive environmental cu

In some insect species, two sites of juvenile hormone (JH) synthesis have been reported: the very well documented corpora allata that secrete JH for "general use", and the reproductive system, in particular the male accessory glands, in which the function of the sometimes huge amounts of JH (e.g. in Hyalophora cecropia) remains to be clarified. A recent finding in Schistocerca gregaria, namely that suppression of the ecdysteroid peak preceding a molt by RNAi of the Halloween genes spook, phantom and shade does not impede normal molting, challenges the (never experimentally proven) classical co

The oceanic bathypelagic realm (1000-4000 m) is a nutrient-poor habitat. Most fishes living there have pelagic larvae using the rich waters of the upper 200 m. Morphological and behavioural specializations necessary to occupy such contrasting environments have resulted in remarkable developmental changes and life-history strategies. We resolve a long-standing biological and taxonomic conundrum by documenting the most extreme example of ontogenetic metamorphoses and sexual dimorphism in vertebrates. Based on morphology and mitogenomic sequence data, we show that fishes currently assigned to thr

The thyroid hormone plays an essential role in the metamorphosis in flatfish, during which external asymmetry (for example, eye relocation and pigmentation) is established. However, no information is available on the expression mechanisms of metamorphic asymmetry. Since the presence of malformed juveniles (having either ocular or blind side characteristics on both sides) was known in spotted halibut (Verasper variegatus), the effect of the timing of thyroid hormone treatment was investigated. When thyroxine (T4, 10 or 30 ng/ml) was administered to the hypothyroid larvae (continuously receiving

The insect larval fat body is the primary organ for detoxification of environmental pesticides and metabolic waste. During lepidopteran metamorphosis, the larval fat bodies undergo degradation and reprogramming for development, leaving the process of detoxification largely unknown at this stage. Using integrative approaches in the silkworm, Bombyx mori, we demonstrate that the pupal rectal sac, a greatly enlarged hindgut structure, serves as a central detoxification hub during the non-feeding metamorphic period. We show that it actively sequesters metabolic waste (uric acid) and, remarkably, c

The larva-to-pupa transition in honey bees (Apis mellifera) involves extensive neural remodeling, yet the molecular dynamics of brain development and their relationship with the surrounding microbial environment remain poorly characterized. This study integrated brain transcriptomic profiling with comb-associated metagenomic analysis to characterize stage-specific molecular signatures during metamorphosis. RNA sequencing of larval and pupal brains was combined with honeycomb shotgun metagenomics from the same sample. Brain transcriptomes exhibited marked stage-specific divergence. Pupae displa
