Molting success of Ixodes scapularis varies among individual blood meal hosts and species.

The blacklegged tick (Ixodes scapularis) is an important vector of emerging human pathogens. It has three blood-feeding stages, as follows: … divided into a series of steps, as follows: finding a host, taking a blood meal, becoming infected, molting, and overwintering. The chance of succeeding in each of these steps may depend on the species identity of the blood meal host. We used a Bayesian method to estimate the molting success of larval I. scapularis collected from four commonly parasitized …

C. elegans peb-1 mutants exhibit pleiotropic defects in molting, feeding, and morphology.

Caenorhabditis elegans PEB-1 is a novel DNA-binding protein expressed in most pharyngeal cell types and outside the pharynx in the hypodermis, hindgut, and vulva. Previous RNAi analyses indicated that PEB-1 is required for normal morphology of these tissues and growth; however, the peb-1 null phenotype was unknown. Here we describe the deletion mutant peb-1(cu9) that not only exhibits the morphological defects observed in peb-1(RNAi) animals, but also results in penetrant larval lethality characterized by defects in pharyngeal function and molting. Consistent with a function in molting, we fou…

Transglutaminase-catalyzed reaction is important for molting of Onchocerca volvulus third-stage larvae.

Highly insoluble proteins, which are probably cross-linked, are common in the cuticle and epicuticle of filarial parasites and other nematode species. We have investigated the possible involvement of transglutaminase (TGase)-catalyzed reactions in the development of Onchocerca volvulus fourth-stage larvae (L4) by testing the effects of TGase inhibitors on the survival of third-stage larvae (L3) and the molting of L3 to L4 in vitro. The larvae were cultured in the presence of three specific TGase inhibitors: monodansylcadaverine, cystamine, and N-benzyloxycarbonyl-D,L-beta-(3-bromo-4,5-dihydroi…

Injuries and molting interference in a trilobite from the Cambrian (Furongian) of South China.

The risk of major malformations in the offspring of mothers with epilepsy receiving antiepileptic drugs is 4--8% compared to 2--4% in the general population. Risk factors include daily dose and polytherapy. Selected drugs have been found to be associated with a higher risk of specific malformations (congenital heart defects and cleft palate with phenytoin and barbiturates; neural tube defects with valproate and carbamazepine). Although some of these findings are unquestionable, several questions are still unsolved, depending the characteristics of the target populations, the small samples of p

Reproductive Biology of Triatoma brasiliensis (Hemiptera, Triatominae) During the Imaginal Molt.

The triatomines are vectors of the protozoan Trypanosoma cruzi, etiologic agent of Chagas disease. These insects are sexually active after the imaginal molt. Some aspects have been studied in Triatoma brasiliensis during the imaginal molt, such as autogeny in virgin females and the relationship between blood ingestion by fifth instar nymph and the realization of the imaginal molt. Thus, to aid in the understanding of reproductive biology and developmental physiology of these vectors, this article analyzes the spermatogenesis of T. brasiliensis during the imaginal molt. The analysis of the semi

High-zinc diets accelerate molting and recovery by remodeling the cecal microbiome in laying hens.

INTRODUCTION: Conventional fasting molting can restore laying performance but imposes substantial physiological stress and welfare concerns. High-zinc diets (2% ZnO) have been proposed as a less stressful alternative, but their mechanisms remain unclear. METHODS: This study compared fasting-induced molting with zinc oxide supplementation in 384 Lohmann Pink hens (65 wk). Serum, cecal contents, and ovaries were sampled across six stages from pre-molt to post-molt recovery. RESULTS: ZnO accelerated cessation of lay (5.75 vs. 8.87 d) and earlier recovery to 50% production (15.25 vs. 16.62 d) wi…

RNAi-Based Insecticide Target Screening: Four Genes Critical for Cuticular Pigmentation and Molting in Beetles.

Monochamus alternatus, a destructive pine-boring pest, urgently requires finding alternatives to chemical pesticides for its eco-friendly control, such as RNA interference (RNAi)-based pesticides. This study screened potential RNAi targets by investigating cuticle-associated genes involved in pigmentation and molting. Silencing tyrosine hydroxylase (TH) induced albinism in pupae and adults and reduced pupation rates with 74.8% adult corrected mortality, and TH inhibitors exhibited dose-dependent effects, with high doses causing complete larval mortality. Targeting ultraspiracle via two dsRNAs …

Breaking the fast: first report of dives and ingestion events in molting southern elephant seals.

Southern elephant seals (SES) experience a 'catastrophic molt', a costly event characterized by the renewal of both hair and epidermis that requires high peripheral vascular circulation. Molting animals are therefore constrained by high metabolic heat loss and are thought to fast and remain on land. To examine the ability of individuals to balance the energetic constraints of molting on land we investigate the stomach temperature and movement patterns of molting female SES. We find that 79% of females swam and 61% ingested water or prey items, despite the cost of cold-water exposure while molt…

Comparative transcriptomics reveals the immune dynamics during the molting cycle of swimming crab Portunus trituberculatus.

Molting is one of the most important biological processes of crustacean species, and a number of molecular mechanisms facilitate this complex procedure. However, the understanding of the immune mechanisms underlying crustacean molting cycle remains very limited. This study performed transcriptome sequencing in hemolymph and hepatopancreas of the swimming crab (Portunus trituberculatus) during the four molting stages: post-molt (AB), inter-molt (C), pre-molt (D), and ecdysis (E). The results showed that there were 78,572 unigenes that were obtained in the hemolymph and hepatopancreas of P. trit…

Label-free quantitative proteomic analysis of molting-related proteins of Trichinella spiralis intestinal infective larvae.

Molting is a key step for body-size expansion and environmental adaptation of parasitic nematodes, and it is extremely important for Trichinella spiralis growth and development, but the molting mechanism is not fully understood. In this work, label-free LC-MS/MS was used to determine the proteome differences between T. spiralis muscle larvae (ML) at the encapsulated stage and intestinal infective larvae (IIL) at the molting stage. The results showed that a total of 2885 T. spiralis proteins were identified, 323 of which were differentially expressed. These proteins were involved in cuticle str…

Ontogenesis, gender, and molting influence the venom yield in the spider Coremiocnemis tropix (Araneae, Theraphosidae).

The demand for spider venom increases along with the growing popularity of venoms-based research. A deeper understanding of factors that influence the venom yield in spiders would therefore be of interest to both commercial venom suppliers and research facilities. The present study addresses the influence of several factors on the venom yield by systematically analyzing the data obtained from 1773 electrical milkings of the Australian theraphosid spider Coremiocnemis tropix. Gender and ontogenesis were found to cause a major effect on the venom yield, as adult female C. tropix yielded signific

Beyond molting disruption: Tebufenozide modifies gut immunity and antiviral responses in Helicoverpa armigera (Noctuidae).

The insect growth regulator tebufenozide, an ecdysone agonist, is widely used to control Helicoverpa armigera through disrupting molting and development. However, its other effects on the insect physiology remain poorly understood. This study investigates how tebufenozide modulates gut immunity, microbial homeostasis, and antiviral defense mechanisms in H. armigera larvae. Using RT-qPCR, we analyzed the expression of key immune genes following exposure to lethal (LC₅₀) and sublethal (LC₁₀, LC₂₅) concentrations of tebufenozide. Our results demonstrate that tebufenozide induced the IMD pathway t…

Unraveling the regulatory mechanisms of fasting-induced molting on follicle development in laying hens.

Fasting-Induced Molting (FIM) has been shown to enhance the laying rate and extend the laying cycle of hens in the late laying period. However, the precise regulatory mechanism underlying FIM's effects on follicle development remains unknown. This study aimed to elucidate these mechanisms by utilizing RNA-seq to analyze the gene expression changes during FIM and identify the key genes involved in follicle development, thereby uncovering the molecular pathways through which FIM exerts its regulatory effects. A total of 96 Houdan hens, aged 326 days, were selected for FIM treatment. The results …

Metalloproteases with EGF, CUB, and thrombospondin-1 domains function in molting of Caenorhabditis elegans.

Functional analysis using RNAi was performed on eleven genes for metalloproteases of the M12A family in Caenorhabditis elegans and the interference of the C17G1.6 gene (nas-37) was found to cause incomplete molting. The RNAi of the C26C6.3 gene (nas-36) also caused a similar molting defect but not so severely as that of the nas-37 gene. Both the genes encode an astacin-like metalloprotease with an epidermal growth factor (EGF)-like domain, a CUB domain, and a thrombospondin-1 domain, in this order. The promoter-driven green fluorescent protein (GFP) expression analysis suggested that they are …

Dirofilaria immitis: further characterization of the transglutaminase enzyme and its role in larval molting.

We recently reported the cDNA cloning and functional characterization of a novel transglutaminase (TGase) from the dog filarial parasite Dirofilaria immitis. D. immitis TGase (DiTG) has no sequence similarity with any other known TGase, but has significant similarity to protein disulfide isomerase (PDI)-related endoplasmic reticulum protein ERp60. In the present study. we further characterized the recombinant DiTG (rDiTG) and studied its role in the molting process of third-stage larvae. The enzymatic activity of rDiTG requires Ca2+, and the maximum activity was observed at a calcium concentra…
