Complete femoral nerve resection with soft tissue sarcoma: functional outcomes.

BACKGROUND: The functional consequences of resecting the femoral nerve in conjunction with soft tissue sarcoma management are not well described. In comparison, sciatic nerve involvement by sarcoma was once considered an indication for amputation, but sciatic resection is now commonly performed as part of a limb-salvage approach. We compared functional outcomes following resection of either the femoral or sciatic nerve in patients with soft tissue sarcoma. We also compared both groups with patients with large thigh sarcomas without nerve involvement. METHODS: The prospectively collected datab

Hydrogel derived from porcine decellularized nerve tissue as a promising biomaterial for repairing peripheral nerve defects.

UNLABELLED: Decellularized matrix hydrogels derived from tissues or organs have been used for tissue repair due to their biocompatibility, tunability, and tissue-specific extracellular matrix (ECM) components. However, the preparation of decellularized peripheral nerve matrix hydrogels and their use to repair nerve defects have not been reported. Here, we developed a hydrogel from porcine decellularized nerve matrix (pDNM-G), which was confirmed to have minimal DNA content and retain collagen and glycosaminoglycans content, thereby allowing gelatinization. The pDNM-G exhibited a nanofibrous st

The preparation and comparison of decellularized nerve scaffold of tissue engineering.

To integrate tissue engineering concepts into strategies to repair spinal cord injury (SCI) has been a hotspot in recent years, and the choice of scaffolding material is crucial to tissue engineering. Recently, decellularized nerve scaffold becomes a central concern due to its peculiar superiority. In this study, the decellularized nerve scaffold was prepared with three different methods and a comparison was made to acquire an ideal scaffold materials. All sciatic nerves from Sprague-Dawley (SD) rats were randomly divided into four groups: A: normal control group, B: TritonX-100 with sodium de

[The role of sensory nerves in the development of inflammation of oral tissues].

Experimental stimulation and clinical procedures applied on the tooth crown cause vascular reactions in the dental pulp of cats and rats. These reactions depend on the activation of trigeminal afferent nerves and release of neuropeptides. A brief stimulation causes vasodilation, which is mainly mediated by calcitonin gene-related peptide (CGRP). A longer stimulation results in plasma extravasation which is mediated mainly by substance P (SP) and prostaglandins in the pulp. In adjacent oral tissues the mechanisms following stimulation or local irritation are more complex and other mediators are

Vasculogenic tissue nanotransfection accelerates functional recovery after peripheral nerve injury.

Peripheral nerve injuries (PNIs) remain a major clinical challenge, often leaving patients with lifelong sensory, motor, and functional impairments despite surgical repair. While gene and cell therapies hold promise, their translation has been hampered by the lack of safe, efficient, and targeted delivery strategies. Here, we introduce vasculogenic tissue nanotransfection (TNT) as a nonviral, reprogramming-based therapeutic platform to enhance nerve regeneration to augment surgical reconstruction. This one-time, millisecond-scale intervention reprograms resident cells in situ toward a vasculog

Fat tissue alters quadriceps response to femoral nerve magnetic stimulation.

OBJECTIVE: We investigate the influence that inguinal fat has on maximal quadriceps response to femoral nerve magnetic stimulation. METHODS: Vastus lateralis M-wave amplitude and quadriceps twitch peak torque were measured (i) in lean and overweight subjects and (ii) when slices of pig fat ranging from 2 to 18 mm in thickness were placed between the coil and the nerve. RESULTS: In overweight group, the maximal response could not be elicited when intensity was ≤90% and ≤85% of maximal power for twitch torque and M-wave amplitude, respectively. The maximal response was obtained at 80% of maxim

Reduced nerve growth factor in Rett syndrome postmortem brain tissue.

To determine whether reduced nerve growth factor (NGF) and/or its high affinity receptor, trkA, play a role in the pathophysiology of Rett syndrome (RS), we used immunohistochemistry in paraffin-embedded human autopsy brain tissue, to quantify NGF and trkA levels within the frontal cortex of 9 RS females and 10 female controls of similar age. The results showed a significant reduction of NGF expression in RS patients (p < 0.001). Specifically, all RS brains exhibited NGF levels at or below the minimum level observed in controls. In 3 RS brains there was no NGF detected. TrkA expression was als

Imaging MS of Rodent Ocular Tissues and the Optic Nerve.

The visual system is comprised of many specialized cell types that are essential for relaying sensory information about an animal's surroundings to the brain. The cells present in ocular tissue are notoriously delicate, making it particularly challenging to section thin slices of unfixed tissue. Maintaining the morphology of the native tissue is crucial for accurate observations by either conventional staining techniques or in this instance matrix-assisted laser desorption ionization (MALDI IMS) or imaging using mass spectrometry. As vision loss is a significantly debilitating condition, study

[Tissue therapy of trigeminal neuralgia with the implantation of preserved nerve].

Cadaveric trigeminal and sciatic nerves or Gasser's ganglion were transplanted subcutaneously below the scapular angle twice a week to 71 patients with trigeminal neuralgia. The procedure is simple. The operation has been fairly effective: 42.3% of patients have been cured (a follow-up of more than 5 years), in 32.4% a marked effect has been achieved persisting for 3-4 years, the condition of 22.5% of patients has improved for 16-18 months, and in only 2.8% of cases the alleviation lasted but 3 months.

Effect of bupivacaine and adjuvant drugs for regional anesthesia on nerve tissue oximetry and nerve blood flow.

BACKGROUND: Nerve blood flow has a critical role in acute and chronic pathologies in peripheral nerves. Influences of local anesthetics and adjuvants on tissue perfusion and oxygenation are deemed as relevant factors for nerve damage after peripheral regional anesthesia. The link between low tissue perfusion due to local anesthetics and resulting tissue oxygenation is unclear. METHODS: Combined tissue spectrophotometry and laser-Doppler flowmetry were used to assess nerve blood flow in 40 surgically exposed median nerves in pigs, as well as nerve tissue oximetry for 60 min. After baseline mea

Dynamic molecular landscape in dorsal root ganglion for peripheral nerve regeneration promoted by tissue engineered nerve graft.

INTRODUCTION: The limited regenerative capacity of the nervous system represents a significant clinical challenge in the context of peripheral nerve injuries. An innovative strategy for sciatic nerve repair has been developed using tissue-engineered nerve grafts (TENGs) composed of skin-derived precursor Schwann-like cells (SKP-SCs) and a silk fibroin-chitosan scaffold. However, the reason why SKP-SCs-TENG demonstrated superior enhanced nerve regeneration compared to the autograft and scaffold groups remains unclear. OBJECTIVES: The present work aims to elucidate the superiority and molecular

Native paraneurial tissue and paraneurial adhesions alter nerve strain distribution in rat sciatic nerves.

Paraneurial adhesions have been implicated in the pathological progression of entrapment neuropathies. Surgical decompression of adhesions is often performed, with the intent of restoring nerve kinematics. The normal counterpart of adhesions, native paraneurium, is also thought to influence nerve deformation and mobility. However, influences of native or abnormal paraneurial structures on nerve kinematics have not been investigated. We measured regional strains in rat sciatic nerves before and immediately after decompression of native paraneurial tissue, and before and after decompression of a

Tissue Plasminogen Activator Loaded PCL Nanofibrous Scaffold Promoted Nerve Regeneration After Sciatic Nerve Transection in Male Rats.

According to the studies, damages to the peripheral nerve as a result of a trauma or acute compression, stretching, or burns accounts for a vast range of discomforts which strongly impressed the patient's life quality. Applying highly potent biomolecules and growth factors in the damaged nerve site would promote the probability of nerve regeneration and functional recovery. Tissue plasminogen activator (tPA) is one of the components that can contribute importantly to degenerating and regenerating the peripheral nerves following the injuries occurred and the absence of this biomolecule hinders

Soft-Tissue Contouring and Nerve Management During Lower-Limb Osseointegration Surgery.

BACKGROUND: Osseointegration provides a direct prosthesis interface for lower-limb amputees, many of whom are poor candidates for traditional socket-suspended prostheses. These skeletally anchored implants eliminate the skin-prosthesis interface, reducing complications such as soft-tissue breakdown, ulceration, and pain1. Soft-tissue contouring and management of peripheral nerves are essential aspects of osseointegrated implantation surgery for both transfemoral and transtibial amputees. These techniques protect the bone-implant interface, prevent skin-implant friction, and reduce neuroma form

Sympathetic nerve tissue in milky spots of the human greater omentum.

Omental milky spots (OMSs), small lymphoid structures positioned in the greater omentum, are involved in peritoneal immune homeostasis and the formation of omental metastases. Sympathetic nerve activity is known to regulate immune function in other lymphoid organs (e.g. spleen and lymph nodes) and to create a favourable microenvironment for various tumour types. However, it is still unknown whether OMSs receive sympathetic innervation. Therefore, the aim of this study was to establish whether OMSs of the adult human greater omentum receive sympathetic innervation. A total of 18 OMSs were isola

Preparation and characterisation of zein/polyphenol nanofibres for nerve tissue regeneration.

Bridging strategies are required to repair peripheral nerve injuries that result in gaps >5-8 mm. Limitations such as donor-site morbidity and size mismatches with receptor sites for autografts, together with immunological problems associated with allografts and xenografts, have created an increased interest in the field of manufactured nerve guide conduits. In this study, zein, a plant protein-based polymer, was electrospun to prepare nanofibrous mats. An important challenge with zein mats is the rapid change from fibre to film under aqueous conditions. Tannic acid (TA), which is a polyphenol

Ectopic thyroid tissue surrounding the right laryngeal nerve: a case report.

Ectopic thyroid tissue (ETT) is a rare developmental anomaly of the thyroid tissue which is defined as the presence of thyroid tissue in locations other than the pretracheal area. However, ectopic thyroid tissue in the lateral neck surrounding the recurrent laryngeal nerve is unusually found. Here we describe a case of a 64-year-old woman who was found bilateral thyroid goiter by the ultrasound examination. The total thyroidectomy plus a modified radical neck dissection was performed. Surprisingly we also found a nodule surrounding the right recurrent laryngeal nerve at the same time. Neverthe
