TDP-43 regulates LC3ylation in neural tissue through ATG4B cryptic splicing inhibition.

Amyotrophic lateral sclerosis (ALS) is an adult-onset motor neuron disease with a mean survival time of three years. The 97% of the cases have TDP-43 nuclear depletion and cytoplasmic aggregation in motor neurons. TDP-43 prevents non-conserved cryptic exon splicing in certain genes, maintaining transcript stability, including ATG4B, which is crucial for autophagosome maturation and Microtubule-associated proteins 1A/1B light chain 3B (LC3B) homeostasis. In ALS mice (G93A), Atg4b depletion worsens survival rates and autophagy function. For the first time, we observed an elevation of LC3ylation

Neonatal neural responses to novelty related to behavioral inhibition at 1 year.

Behavioral inhibition (BI), an early-life temperament characterized by vigilant responses to novelty, is a risk factor for anxiety disorders. In this study, we investigated whether differences in neonatal brain responses to infrequent auditory stimuli relate to children's BI at 1 year of age. Using functional magnetic resonance imaging (fMRI), we collected blood-oxygen-level-dependent (BOLD) data from N = 45 full-term, sleeping neonates during an adapted auditory oddball paradigm and measured BI from n = 27 of these children 1 year later using an observational assessment. Whole-brain analyses

The neural correlates of tic inhibition in Gilles de la Tourette syndrome.

Tics in Gilles de la Tourette syndrome (GTS) resemble fragments of normal motor behaviour but appear in an intrusive, repetitive and context-inappropriate manner. Although tics can be voluntarily inhibited on demand, the neural correlates of this process remain unclear. 14 GTS adults without relevant comorbidities participated in this study. First, tic severity and voluntary tic inhibitory capacity were evaluated outside the scanner. Second, patients were examined with resting state functional magnetic resonance imaging (RS-fMRI) in two states, free ticcing and voluntary tic inhibition. Local

A neural network approach to the acoustic startle reflex and prepulse inhibition.

Prepulse inhibition (PPI) is the normal suppression of the startle reflex when an intense stimulus is preceded by a weak non-startling prestimulus. PPI is widely used as a model for sensorimotor gating processes and has been shown to be impaired in various neuropsychiatric disorders, including schizophrenia. We have reproduced startle-like behavior and basic PPI modifications with a neural network. The network design was constrained by the attempt (1) to use as few connections as possible and (2) to relate neuroanatomical structures to the simulated network. Performance of the network was eval

Towards a common neural substrate in the immediate and effective inhibition of stuttering.

Stuttering can be effectively inhibited via exogenous sensory signals (e.g., speaking in unison or using altered auditory feedback) or by using endogenous motoric strategies (e.g., singing or therapeutically implementing long vowel prolongations to reduce speech rates). We propose that these channels, which superficially appear to be diametrically opposite, centrally converge in the engagement of mirror neurons for fluent gestural productions. Sensory changes incurred via exogenous speech signals allow for direct engagement of mirror systems, while endogenous motor strategies appear to require

Adrenergic inhibition of neurally evoked secretion in ground squirrel intestine.

The role of norepinephrine in the mucosal response to neural stimulation was examined in the ground squirrel jejunum. Electrical field stimulation of jejunal sheets in flux chambers evoked a tetrodotoxin-sensitive increase in short-circuit current (42 +/- 6 microA/cm2) that declined during the stimulus period, followed by a second rise in current after the stimulus was terminated. The initial increase in current was inhibited by bumetanide and significantly reduced in chloride-free solutions, suggesting neurotransmitter stimulation of electrogenic chloride secretion during electrical stimulati

Similar Neural Correlates of Planning and Execution to Inhibit Continuing Actions.

Inhibition of action is involved in stopping a movement, as well as terminating unnecessary movement during performance of a behavior. The inhibition of single actions, known as response inhibition (Inhibition of the urge to respond before or after actions) has been widely investigated using the go/no-go task and stop signal task. However, few studies focused on phase and volition-related inhibition after an action has been initiated. Here, we used functional magnetic resonance imaging (fMRI) to investigate the neural correlates of planning and execution underlying the voluntary inhibition of

The gentle grip of beauty: Neural bases of motor inhibition during aesthetic appreciation.

A century-old tradition characterizes aesthetic experiences as intense contemplative moments in which we suspend practical concerns and devote our full attention to the object at hand. In line with this tradition, recent theories suggest that aesthetic experiences involve heightened sensory processing accompanied by a suppression of motor activity. Yet, while the link between aesthetic appreciation and perceptual enhancement has received considerable attention, the role of motor inhibition remains largely unexplored. The present study addressed this gap by investigating whether the aesthetic a

Excitation/inhibition balance subtypes in autism and their genetic, neural, and clinical profiles.

Excitation (E)/inhibition (I) imbalance is considered a key mechanism in Autism Spectrum Disorder (ASD). However, E/I imbalance can have different etiologies with increased E relative to I (E > I) and increased I relative to E (E < I). Both neural profiles can be associated with altered clinical phenotype, suggesting "bell"-shape brain-behavior relationships. We derived E/I balance measures from resting-state EEG in a large sex-balanced sample of youths with and without autism (N = 310; 164 youths with ASD and 146 typically developing (TD) youths) to address group discriminative power of neura

Effects of Youth Sport Participation on Neural Processing During Response Inhibition in Children.

PURPOSE: Youth sports offer many benefits to developing children, but collision sports introduce additional risks from exposure to repetitive head impacts. Past research has linked this exposure to reduced cognitive performance, including response inhibition. The current study aimed to probe how participation in collision sports affects inhibitory control in current youth collision sport athletes, compared with peers who participate in non-contact sports or non-sport activities. METHODS: We used functional magnetic resonance imaging data acquired in the context of a response inhibition task f

Agrin Inhibition in Enteric Neural Stem Cells Enhances Their Migration Following Colonic Transplantation.

Regenerative cell therapy to replenish the missing neurons and glia in the aganglionic segment of Hirschsprung disease represents a promising treatment option. However, the success of cell therapies for this condition are hindered by poor migration of the transplanted cells. This limitation is in part due to a markedly less permissive extracellular environment in the postnatal gut than that of the embryo. Coordinated interactions between enteric neural crest-derived cells (ENCDCs) and their local environment drive migration along the embryonic gut during development of the enteric nervous syst

Attentional salience and the neural substrates of response inhibition in borderline personality disorder.

BACKGROUND: Impulsivity is a central symptom of borderline personality disorder (BPD) and its neural basis may be instantiated in a frontoparietal network involved in response inhibition. However, research has yet to determine whether neural activation differences in BPD associated with response inhibition are attributed to attentional saliency, which is subserved by a partially overlapping network of brain regions. METHODS: Patients with BPD (n = 45) and 29 healthy controls (HCs; n = 29) underwent functional magnetic resonance imaging while completing a novel go/no-go task with infrequent od

Neural Correlates of Semantic Inhibition in Relation to Hypomanic Traits: An fMRI Study.

OBJECTIVES: Language modifications are a core feature of mania, but little is known about the semantic mechanisms behind these disturbances. The aim of the present study was thus to identify deficits in semantic inhibition and their respective neural activation patterns in a sample of individuals assessed for hypomanic personality traits. METHODS: Thirty-six young adults with no neurological or psychiatric diagnoses were assessed for hypomanic personality traits with the Hypomanic Personality Scale (HPS) and underwent an fMRI task of semantic ambiguity resolution. RESULTS: Regression analyse

Neural correlates of generation and inhibition of verbal association patterns in mood disorders.

OBJECTIVES: Thought disorders such as rumination or flight of ideas are frequent in patients with mood disorders, and not systematically linked to mood state. These symptoms point to anomalies in cognitive processes mediating the generation and control of thoughts; for example, associative thinking and inhibition. However, their neural substrates are not known. METHOD: To obtain an ecological measure of neural processes underlying the generation and suppression of spontaneous thoughts, we designed a free word association task during fMRI allowing us to explore verbal associative patterns in p

Neural imbalance between feedback sensitivity and motor inhibition in compulsivity and negative urgency.

Compulsivity and emotional impulsivity (negative urgency) are considered transdiagnostic risk factors for compulsive-impulsive psychopathology that is linked to a neural imbalance between executive and motivational-emotional systems. However, existing evidence does not derive from within-subjects designs, leaving it unclear whether imbalance occurs with high expressions in compulsivity and negative urgency. To address this gap, we conducted a preregistered analysis to examine how feedback sensitivity and motor inhibition interact as a function of compulsivity and negative urgency.Participants

Age-related neural inefficiency: fNIRS evidence of prefrontal hyperactivation during emotional response inhibition.

Older adults often exhibit reduced inhibitory control accompanied by altered recruitment of prefrontal networks. Whether the emotional context changes these age-related neural patterns during response inhibition remains unclear. In this study, 31 older adults and 19 young adults completed four blocks of a Go/No-Go paradigm while bilateral prefrontal cortex (PFC) activity was recorded using functional near-infrared spectroscopy (fNIRS). Blocks 1 and 2 comprised a neutral (non-emotional) Go/No-Go task using geometric shapes, while blocks 3 and 4 comprised an emotional Go/No-Go task using facial

Dissociating the neural substrates for inhibition and shifting in domain-general cognitive control.

Inhibition and shifting are two key components of domain-general cognitive control. Numerous studies have investigated the neural substrates of both components, but it is still unclear whether the relevant brain regions are specifically involved in one specific component or commonly engaged in both components. Here, we addressed this question by using functional magnetic resonance imaging and a modified saccade paradigm that was effective to disentangle inhibition and shifting in one experiment. The results showed that both the middle frontal gyrus and left parietal lobe were involved in both

Large-scale functional neural network correlates of response inhibition: an fMRI meta-analysis.

An influential hypothesis from the last decade proposed that regions within the right inferior frontal cortex of the human brain were dedicated to supporting response inhibition. There is growing evidence, however, to support an alternative model, which proposes that neural areas associated with specific inhibitory control tasks co-exist as common network mechanisms, supporting diverse cognitive processes. This meta-analysis of 225 studies comprising 323 experiments examined the common and distinct neural correlates of cognitive processes for response inhibition, namely interference resolution

Neural Stem Cells and Its Derivatives as a New Material for Melanin Inhibition.

The pigment molecule, melanin, is produced from melanosomes of melanocytes through melanogenesis, which is a complex process involving a combination of chemical and enzymatically catalyzed reactions. The synthesis of melanin is primarily influenced by tyrosinase (TYR), which has attracted interest as a target molecule for the regulation of pigmentation or depigmentation in skin. Thus, direct inhibitors of TYR activity have been sought from various natural and synthetic materials. However, due to issues with these inhibitors, such as weak or permanent ability for depigmentation, allergy, irrita

Neural Inhibition of Dopaminergic Signaling Enhances Immunity in a Cell-Non-autonomous Manner.

The innate immune system is the front line of host defense against microbial infections, but its rapid and uncontrolled activation elicits microbicidal mechanisms that have deleterious effects [1, 2]. Increasing evidence indicates that the metazoan nervous system, which responds to stimuli originating from both the internal and the external environment, functions as a modulatory apparatus that controls not only microbial killing pathways but also cellular homeostatic mechanisms [3-5]. Here we report that dopamine signaling controls innate immune responses through a D1-like dopamine receptor, D
