The aim of this study was to investigate the protective effects of olive leaf extract (OLE) on genotoxicity and oxidative damage in cultured human blood cells treated with permethrin (PM) in the presence of a rat liver S9 mix containing cytochrome P 450 enzymes. Anti-genotoxic activities of OLE were studied using sister chromatid exchange (SCE) and chromosome aberration (CA) tests and furthermore total antioxidant capacity (TAC) and total oxidative status (TOS) were examined to determine the oxidative damage. Our results clearly revealed that treatment with PM (200 mg/l) alone increased SCE an

Olea Europaea is the most important allergenic tree in Southern Italy and in the Naples area. Its pollination period lasts from the middle of April to the end of June. In our pollinosis patients we observed frequency of sensitization of 13.05% to Olea pollen. That means that this pollen follows Parietaria (47.80%) and Gramineae (34.70%) regarding the frequency of allergic sensitization. In an attempt to evaluate the degree of allergic sensitization to Olea pollen and the concordance of various tests, we studied 24 asthmatic patients with skin positivity to only this pollen using commercially a…

Olea europaea L. 'Lavagnina' is cultivated in the Eastern Ligurian coast (Italy), and during the pruning process a huge amount of pruning residues is produced. This by-product is generally disposed of by burning, despite still containing bioactive compounds. In particular, olive leaves are indeed rich in secondary metabolites, which can vary both in quality and quantity in relation to the cultivar considered and the area of cultivation. For this reason, we aimed to carry out a pharmacognostic study of the pruned leaves of the unexplored local cultivar 'Lavagnina', evaluating the possibility of…

BACKGROUND: Several cross-reacting proteins have been identified as responsible of the co-occurrence of pollinosis and plant-derived food allergy. This association has been mainly described in the birch-apple syndrome but other pollens such as Olea europaea and other fruits may also contain homologous proteins.
OBJECTIVE: To evaluate the associations between sensitization to allergens of Olea europaea pollen and confirmed plant-derived food allergy, in addition to investigate if any pattern of clinical hypersensitivity of food allergy reaction (oral allergy syndrome (OAS) or anaphylaxis) and/…

BACKGROUND: The olive fruit fly Bactrocera oleae (Rossi) (OLF) is a major agricultural pest, whose control primarily relies on the use of chemical insecticides. Therefore, development of sustainable control strategies is highly desirable. The primary endosymbiotic bacterium of OLF, 'Candidatus Erwinia dacicola', is essential for successful larval development in unripe olive fruits. Therefore, targeting this endosymbiont with antimicrobial compounds may result in OLF fitness reduction and may exert control on natural populations of OLF.
RESULTS: Here, we evaluate the impact of compounds with a…

In the last few years interest in the clinical aspects of Olea europaea (O.e.) pollen allergy has increased. For many years we have observed in our geographical area a perennial pattern of clinical symptoms in subjects with monosensitization to O.e. allergens without any worsening during the olive pollen season. We tried to demonstrate the clinical relevance of O.e. sensitization in our patients and, moreover, to determine why this pattern is elicited. We selected a group of 26 patients with rhinitis and/or bronchial asthma and an immediate positive skin reaction only to O.e. pollen extract. U…

Ethnopharmacological use of plant natural extracts has been known since ancient times. The optimization of plant molecule extraction is fundamental in obtaining relevant extraction yields. The main purpose of this study was to understand the role of different extraction techniques (solid-liquid, ultrasound, Soxhlet, and microwave) and solvents (water, methanol, ethanol, acetone, dichloromethane, and hexane) on the antimicrobial and antioxidant activities of extracts from Olea europaea (olive) and Acacia dealbata (mimosa). Crude plant extracts were evaluated for their antimicrobial activity aga…

With the increasing popularity of olive oil, the cultivation area of Olea europaea L. (olive) trees has been expanding, and the processing and utilization of olive leaves are gaining more attention. Besides oleuropein, the main active compounds in olive leaves include luteolin and its glycosides. Through fractionation, a component rich in key active substances, designated OLE, was prepared from olive leaves. Our activity analysis showed that luteolin exhibits excellent DPPH radical scavenging and higher anti-inflammatory activities compared to its glycosides. To enhance the activity of OLE, a …

The wild olive (Olea europaea L.) is one of the most iconic Mediterranean forest tree species and the ancestor of many cultivated olive varieties. In this study, twelve wild olive genotypes from three distinct subspecies (subsp. europaea, subsp. guanchica, and subsp. maroccana) were grown under ambient ([CO2]amb) and enriched CO2 atmospheres ([CO2]enr), subject to two watering regimes (WW - Well-watered/WS - Water-stressed). We measured water use at leaf and whole-plant levels, along with growth traits, leaf gas exchange, and leaf isotopic carbon (δ13C) and nitrogen composition (δ15N), to diff…

The main phenolics compounds in Olea europaea L. leaves are oleuropein, hydroxytyrosol, hydroxytyrosol hexoside, verbascoside and apingenin-7-glucoside, being oleuropein the most abundant. In the literature, there are different methods to determine some of these compounds, which imply an analytical preparation for their determination. In this line, the use of techniques based on infrared spectroscopy is presented as a promising alternative. Therefore, a near-infrared spectroscopy (NIR) calibration, validation and prediction to determine the main metabolites in olive leaves has been developed. …

Seasonal flu is caused by influenza infection, a virus that spreads easily in human population with periodical epidemic outbreaks. The high mutational rate of influenza viruses leads to the emergence of strains resistant to the current treatments. Due to that, scientific research is focusing on the development of new anti-influenza agents as alternative or complementary treatments. Olive tree (Olea europaea L.) has been a source of ancestral remedies due to its antimicrobial activity. Thus, the aim of this study was to test the anti-influenza activity of a standardized olive leaf extract rich …

Olive pruning residues represent a promising source of bioactive compounds. This study presents the first single-step supercritical fluid extraction (SFE) approach for the simultaneous recovery of phenols, carotenoids, and tocopherols from Olea europaea leaves and branches. Using bio-ethanol-modified CO2, the optimized method achieved recoveries of 76% for oleuropein, 84% for α-tocopherol, 88% for lutein, and 90% for β-carotene. Compared to ultrasound-assisted extraction (UAE), SFE yielded lower absolute amounts than the exhaustive UAE reference methods, while maintaining high recovery efficie…

Olive (Olea europea L.) is one of the oldest and most important fruit tree species cultivated in the Mediterranean region. Various plant tissues, drupes, and olive oil contain several phenolics (including verbascoside, although it is present in the plant at a low level) that are well-known for their highly beneficial effects on human health. An in vitro olive cell suspension culture (cultivar Cellina di Nardò, "CdN") was established, characterized for its growth and morphological features. Furthermore, a vital and relatively uniform population of protoplasts was generated from the olive suspen…

Doxorubicin is an anthracycline that is commonly used as a chemotherapy drug due to its cytotoxic effects. The clinical use of doxorubicin is limited due to its known cardiotoxic effects. Treatment with anthracyclines causes heart failure in 15-17% of patients, resulting in mitochondrial dysfunction, the accumulation of reactive oxygen species, intracellular calcium dysregulation, the deterioration of the cardiomyocyte structure, and apoptotic cell death. Polyphenols have a wide range of beneficial properties, and particular importance is given to Bergamot Polyphenolic Fraction; Oleuropein, on
