KA-bridged transplantation of mesencephalic tissue and olfactory ensheathing cells in a Parkinsonian rat model.

The pathology of Parkinson's disease (PD) results mainly from nigrostriatal pathway damage. Unfortunately, commonly used PD therapies do not repair the disconnected circuitry. It has been reported that using kainic acid (KA, an excitatory amino acid) in bridging transplantation may be useful to generate an artificial tract and reconstruct the nigrostriatal pathway in 6-hydroxydopamine (6-OHDA) lesioned rats. In this study, we used KA bridging and a co-graft of rat olfactory ensheathing cells (OECs) and rat E14 embryonic ventral mesencephalic (VM) tissue to restore the nigrostriatal pathway of

Ultrastructural changes induced by experimental subarachnoid haemorrhage and 6-hydroxydopamine in cat cerebral arteries.

Transmission electron microscopy of the middle cerebral artery from cats exposed to subarachnoid injection of blood 3 and 7 days before, showed several damage of the vascular ultrastructure. The intima was thickened with swelling and vacuolization of endothelial cells, with a plump appearance and disruption of tight junctions. The cellular surface was corrugated and the subendothelial space contained proliferating and vacuolated smooth muscle cells capped by elastin and collagen fibres. The internal elastic lamina was also corrugated and disrupted. The adventitial changes were axonic cytoplasm

In vitro modelling of Parkinson's disease using 6-OHDA is associated with increased NQO2 activity.

The pathogenesis of Parkinson's disease (PD) involves abnormalities in the metabolism of catecholamines. The enzyme quinone reductase 2 (NQO2) reduces quinone derivatives of catecholamines, which promotes the formation of reactive oxygen species (ROS), suggesting a role for NQO2 in the development of cellular damage typical of PD. In the present study, we investigated the relationship between 6-hydroxydophamine (6-OHDA) induced cellular damage and NQO2 activity and its levels in SH-SY5Y cell culture to establish an experimental model to evaluate the pharmacological properties of NQO2 inhibitor

Transcranial direct current stimulation improves motor function in rats with 6-hydroxydopamine-induced Parkinsonism.

Transcranial direct current stimulation (tDCS) is increasingly being used for Parkinson's disease (PD); however, the evaluation of its clinical impact remains complex owing to the heterogeneity of patients and treatments. Therefore, we used a unilateral 6-hydroxydopamine-induced PD rat model to investigate whether anodal tDCS of the primary motor cortex (M1) alleviates PD motor deficits. Before tDCS treatment, unilateral PD rats preferentially used the forelimb ipsilateral to the lesion in the exploratory cylinder test and showed reduced locomotor activity in the open field test. In addition,

Inhibition of anandamide hydrolysis does not rescue respiratory abnormalities observed in an animal model of Parkinson's disease.

NEW FINDINGS: What is the central question of this study? The respiratory frequency to hypercapnia is attenuated in an animal model of Parkinson's disease (PD): what is the therapeutic potential of inhibition of anandamide hydrolysis for this respiratory deficit? What is the main finding and its importance? In an animal model of PD there is an increased variability in resting respiratory frequency and an impaired tachypnoeic response to hypercapnia, which is accompanied by diminished expression of Phox2b immunoreactivity in the retrotrapezoid nucleus (RTN). Inhibition of anandamide hydrolysis

[Modulation of norepinephrinergic control of hypothalamus preoptic areas on hibernation bouts in ground squirrels (Citellus dauricus)].

In our previous work, it was shown that hibernation could be enhanced by injection of 6-hydroxydopamine (6-OHDA, 200 micrograms) into the lateral cerebral ventricle of ground squirrels by destructing norepinephrine (NE) nerve terminals in brain areas including hypothalamus, cerebral cortex, hippocampus and periaqueductal gray matter of middle brain. In order to identify the specific area contributing for the modulation of NE system on hibernation, small doses of 6-OHDA (4 micrograms) were microinjected bilaterally into medial preoptic hypothalamus (MPA), lateral preoptic hypothalamus (LPA) and

Neonatal destruction of dopaminergic neurons.

Rats treated as neonates with 6-hydroxydopamine are proposed to model the dopamine deficiency associated with Lesch-Nyhan syndrome (LNS). To understand the neurobiological basis of specific behaviors in LNS, investigations were undertaken in these neonatally lesioned rats. Several new findings resulted from these studies. The first was that D1-dopamine receptors are essential for the action of D2-dopamine receptors, a phenomenon called "coupling" of receptor function. Another finding was that D1-dopamine receptors must be repeatedly stimulated before maximal behavioral sensitivity can be obser

Pharmacological evidence for rapid destruction of efferent renal nerves in rats by intrarenal infusion of 6-hydroxydopamine.

In a previous study we reported that intrarenal (i.r.) infusion of 1 mg 6-hydroxydopamine (6-OHDA) in rats resulted in selective efferent denervation of the infused kidney. Although the vasoconstrictor response to electrical stimulation of the posterior hypothalamus (PH) was already abolished 24 h after infusion, norepinephrine (NE) content of the kidney was reduced by only approximately 50% at that time. In the present study, the status of renal nerves 45 min after infusion of 6-OHDA by i.r. application of scorpion venom (SV), a NE releasing agent, was investigated. In saline pretreated rats,

Accelerometry is a valid method to distinguish between healthy and 6-OHDA-lesioned parkinsonian rats.

In Parkinson's disease (PD), continuous sensor-based evaluation of motor symptom severity, e.g., using accelerometry, has become an emerging field of interest in clinical research. Continuous symptom monitoring would also be of interest in preclinical disease models; however, such devices are far less established in animal models, most likely due to additional requirements in size, energy consumption, and impairment-free attachment. In contrast, accelerometers manufactured in micro-electro-mechanical systems (MEMS) technology are promising sensor devices, which allow for space-saving and energ

Protective effects of silk fibroin against 6-OHDA in SH-SY5Y human neuroblastoma cells and comparative study with its release from gelatin films.

In this study, we analyzed the neuroprotective action of silk fibroin (SF) regenerated with calcium chloride (CaCl2), distinguishing the effects of CaCl2 and SF, and subsequently fabricating a neuroprotective hybrid material based on SF gelatin film. Cytotoxicity induced by 6-hydroxydopamine (6-OHDA) on the human neuroblastoma SH-SY5Y cell line showed that SF had a significant shielding power against 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in SH-SY5Y neuroblastoma cells, as assessed by the CCK-8 assay, cell imaging and cell cycle using flow cytometer. Specifically, the concurrent trea

Apnea behavior in early- and late-stage mouse models of Parkinson's disease: Cineradiographic analysis of spontaneous breathing, acute stress, and swallowing.

This study aimed to evaluate the timing and frequency of spontaneous apneas during breathing and swallowing by using cineradiography on mouse models of early/initial or late/advanced Parkinson's disease (PD). C57BL/6 J mice received either 6-OHDA or vehicle injections into their right striatum, followed by respiratory movement recordings during spontaneous breathing and swallowing, and a stress challenge, two weeks later. Experimental group animals showed a significantly lower respiratory rate (158.66 ± 32.88 breaths/minute in late PD, 173.16 ± 25.19 in early PD versus 185.27 ± 25.36 in contro

Partial lesion of the dopaminergic innervation of the ventral striatum induces "depressive-like" behavior of rats.

Depression is a frequent comorbid disorder in Parkinson's disease (PD) which may precede appearance of its motor symptoms by several years. Pathomechanisms underlying PD have been suggested to be responsible for the PD-related depression. The aim of the study was to examine the influence of a partial lesion of striatal dopaminergic terminals on the "depressive-like" behavior of rats in the forced swimming test (FS). 6-Hydroxydopamine (6-OHDA) was injected bilaterally into the ventro-lateral region of the caudate-putamen (CP) (3.75 μg/2.5 μl/side). The locomotor activity and behavior of rats in

Chick eyes under cycloplegia compensate for spectacle lenses despite six-hydroxy dopamine treatment.

PURPOSE: To test whether eye growth changes produced by spectacle lens wear are mediated by changes in ciliary muscle tonus in chicks. METHODS: Because there is evidence that deprivation myopia is based on a local-retinal mechanism in the eye that probably remains functional after cycloplegia as well as after ciliary ganglion or Edinger-Westphal lesions, none of these treatments provides insight into whether accommodation tonus is also important in the control of axial eye growth. Because 6-hydroxy dopamine (6-OHDA) suppresses deprivation myopia, to isolate growth changes mediated by accommod

Central pressor activity of cimetidine in spontaneously hypertensive rats.

The systemic blood pressure effect of cimetidine given intracerebroventricularly (i.c.v.) in anaesthetized spontaneously hypertensive (SH) rats has been investigated. Cimetidine (250 micrograms i.c.v.) caused a gradual long lasting rise in mean arterial blood pressure with maximum of 31.6 +/- 4.5 mm Hg. Chemical degeneration of catecholaminergic neurons with 6-OHDA treatment, central administration of phentolamine and prazosin, and the bilateral adrenalectomy significantly inhibited the pressor response of cimetidine, while propranolol (i.c.v.) had no effect. From these results it appears that

Neuroprotective Effects of Thymol-Loaded Selenium Nanoparticles against 6-OHDA-Induced Apoptosis and Oxidative Stress in an In Vitro Parkinson's Disease Model.

INTRODUCTION: Parkinson's disease (PD) is characterized by the degeneration of dopaminergic neurons within the substantia nigra, leading to progressive motor dysfunction. There are still limited diseasemodifying options that counteract the process of disease progression. This study aimed to evaluate the neuroprotective effects of thymol, both in its free form and when loaded onto selenium nanoparticles (SeNPs), in a 6-hydroxydopamine (6-OHDA)-induced PD model using SH-SY5Y cells. METHODS: SeNPs were synthesized using a chemical reduction method with ascorbic acid, achieving a 68% entrapment e

Golexanolone affords sustained microglia and astrocytes activation improvement in a rat model of Parkinson's disease.

INTRODUCTION: Golexanolone improves motor and non-motor alterations in the unilateral 6-OHDA rat model of PD. We hypothesized that a key mechanism by which golexanolone induces these beneficial effects is by reducing microglia activation, thus reducing pro-inflammatory factors (TNFα, IL-1α, HMGB1) which activate astrocytes. This work aims were to assess if golexanolone affords sustained improvement of glial activation and pro-inflammatory factors at 3 and 9 weeks after 6-OHDA injection. RESULTS: 6-OHDA rats show pro-inflammatory microglia in SN and striatum, with reduced area and increased TN

No Effects of Decanoic Acid on Locomotor Activity and Antioxidant Defences in an Experimental Animal Model of Attention-Deficit/Hyperactivity Disorder.

BACKGROUND: Medium-chain triglycerides such as decanoic acid (C10), which is one of the fatty acids that constitute dietary fats, are of substantial interest for their potential therapeutic effects on neuropsychiatric disorders. However, the effects of C10 on attention-deficit/hyperactivity disorder (ADHD) remain to be studied. We explored the effects of C10 on behavioural activity and antioxidant defences in an experimental animal model of ADHD. METHODS: To establish an experimental animal model of ADHD, neonatal rats were subjected to unilateral striatal lesions using 6-hydroxydopamine (6-O

Intrapallidal injection of botulinum toxin A recovers gait deficits in a parkinsonian rodent model.

AIM: Modulation of electrical activity in the subthalamic nucleus has been therapeutically effective in Parkinson's disease. Pharmacological manipulation of glutamate release from subthalamic neurons could also favourably alter basal ganglia activity to improve motor symptoms. This study investigates the efficacy of selective suppression of hyperactive glutamatergic input from the subthalamic nucleus to the globus pallidus internal segment by botulinum toxin A (BoNT-A) in a parkinsonian model. METHODS: Unilateral 6-hydroxydopamine lesioned parkinsonian rodents and controls received microinfus

Changes in Rat 50-kHz Ultrasonic Vocalizations During Dopamine Denervation and Aging: Relevance to Neurodegeneration.

Vocal communication is negatively affected by neurodegenerative diseases, such as Parkinson disease, and by aging. The neurological and sensorimotor mechanisms underlying voice deficits in Parkinson disease and aging are not well-understood. Rat ultrasonic vocalizations provide a unique behavioral model for studying communication deficits and the mechanisms underlying these deficits in these conditions. The purpose of this review was to examine the existing literature for methods using rat ultrasonic vocalization with regard to the primary disease pathology of Parkinson disease, dopamine dener
