Role of FBXW2 in explant cultures of bovine periosteum-derived cells.

OBJECTIVE: Bone regeneration is a potential technique for treating osteoporosis. A previous study reported that F-box and WD-40 domain-containing protein 2 (FBXW2) localized with osteocalcin in bovine periosteum after 5 weeks of explant culture. However, the osteoblastic functions of FBXW2 remain unclear. In this study, double-fluorescent immunostaining was used to investigate the potential role of FBXW2 and its relationship with osteocalcin. RESULTS: At day 0, FBXW2 was expressed in the cambium layer between the bone and periosteum, while osteocalcin was expressed in bone. After explant cult

The role of the periosteum in healing a large structural defect following sequestrectomy.

An eight-year-old boy had a sequestrum involving the entire femoral diaphysis removed before significant involucrum had formed. This procedure resulted in a large structural defect. Ten weeks later, the periosteum had completely repaired the effect, and no infection remained. The benefit in this case of early, radical sequestrectomy is discussed.

Effect of estrogen on gene expression of fatty acid synthase in periosteum.

BACKGROUND: Estrogen deficiency contributes to postmenopausal osteoporosis. Periosteum might be a potential target of estrogen, but the underlying mechanism at gene level is far from being elucidated. The objective of this study was to investigate the correlation between estrogen and fatty acid synthase (FAS) expression in periosteum. METHODS: Human periosteum cells were cultured in vitro. Expressed genes in the substrated cDNA library were verified using semi-quantitative PCR and real-time PCR. The expression of FAS in periosteum of ovarectomized (OVX) SD rats was investigated. RESULTS: FAS

Measurement of mechanical characteristics of tibial periosteum and evaluation of local differences.

Stress-strain relationships of bovine tibial periosteum, harvested from anterior, medial, lateral, and posterior aspects of tibia, were successfully measured using a newly developed experimental system. Results showed a curvilinear stress-strain pattern having three regions, i.e., toe, almost linear, and rupture regions, which resembled those of biological soft tissues like ligaments, skin, etc. Tensile moduli in the toe region (Ee) and in the linear region (Ec) were obtained by linear regressional analyses. These values and the tensile strength (sigma t) showed clear local differences. The va

Pseudo-tumoral ossification of the muscles and/or periosteum. (A study of 57 cases).

The authors present a study of fifty-seven cases of pseudo-tumoral ossification of the muscles ("circumscribed ossifying myositis") and of the periosteum, supported by clinical, radiographic and histological evidence. This pathological condition is characterised by the formation of hyperplastic bone callus in muscle or periosteum, with a typical radiographic appearance, and with a relationship to trauma. We have excluded from our study pseudo-tumoral periosteal ossification in conditions of congenital origin (osteogenesis imperfecta, neurofibromatosis, Sane et al., 1971; Kullman et al., 1972),

Inveterate Section of Deep Flexor in Pediatric Hand, Periosteum Clues.

Diagnostic delay in flexor tendon injuries is common in pediatric patients, and age influences both recovery and postoperative management. We report the case of a three-year-old girl who presented with the inability to flex the distal interphalangeal joint of the left third finger, two months after the digit became entrapped in a chair. Ultrasonography revealed cortical ossification of the middle phalanx compressing the flexor tendon, without clear evidence of discontinuity. Surgical exploration was undertaken due to a strong clinical suspicion of flexor digitorum profundus rupture, revealing

Isolation and Culture of Periosteum-Derived Progenitor Cells from Mice.

This chapter describes the methods of isolation of mouse periosteal progenitor cells. There are three basic methods utilized. The bone grafting method was developed utilizing the fracture healing process to expand the progenitor populations. Bone capping methods requires enzymatic digestion and purification of cells from the native periosteum, while the Egression/Explant method requires the least manipulation with placement of cortical bone fragments with attached periosteum in a culture dish. Various cell surface antibodies have been employed over the years to characterize periosteum derived

Treatment of orbital floor fracture using a periosteum-polymer complex.

Various materials for the reconstruction of bone defects in orbital floor fractures have been developed and applied clinically. Recently, reconstruction using polymers, in place of autologous bone and artificial materials, has been actively introduced, but there are problems, such as the size of reconstructable bone defects and the decomposition rate of polymers. A basic study was performed on bone regeneration using a periosteum-polymer complex produced by attaching periosteum to a biodegradable polymer sheet. In this study, patients with orbital floor fractures were evaluated clinically who

Immunomodulatory cytoskeleton of bionic periosteum affects mitochondrial dynamics to promote osteogenesis.

PURPOSE: Periosteum is a key regulatory hub in the immune microenvironment of bone repair and the mitochondrial dynamics of bone marrow mesenchymal stem cells (BMSCs) directly determine the osteogenic efficiency; therefore, how to promote bone healing through the immunomodulation of mitochondrial dynamics is a key challenge. METHODS: Inspired by the natural periosteum, we designed a bionic periosteum that integrates topology and controlled Semaphorin 3 A (Sema3A) release. Hyaluronic acid-coated Sema3A granular microsols spontaneously formed a nucleus-sheath structure under a high-voltage elec

Optimizing Tissue-Engineered Periosteum Biochemical Cues to Hasten Bone Allograft Healing.

Although allografts remain the gold standard for treating critical-size bone defects, ~60% fail within 10 years of implantation. To emulate periosteum-mediated healing of live autografts, we have developed a tissue-engineered periosteum (TEP) to improve allograft healing. The TEP comprises cell-degradable poly(ethylene glycol) hydrogels encapsulating mouse mesenchymal stem cells and osteoprogenitor cells to mimic the periosteal cell population. Despite improvements in allograft healing, several limitations were observed using the TEP, specifically the modulation of host tissue infiltration and

Comparison of bone formation ingrafted periosteum harvested from tibia and calvaria.

Periosteum covers the bone surface and displays the potential to initiate bone formation, after injury to the bone. Numerous studies have demonstrated that the periosteum plays major roles in the healing process after bone fracture. Some reports have described that in the healing of long bone fractures, the periosteum forms new bone by intramembranous and endochondral ossification. Other researchers insist that healing of defects in membrane bone shows bone formation by intramembranous ossification. However, previous studies have not been able to clarify differences in bone formation patterns.

Evaluation of a Decellularisation Process for Developing an Osteogenic Periosteum Membrane In Vitro.

INTRODUCTION: The study addresses the need for a biocompatible, resorbable membrane with osteogenic properties, proposing a periosteum-based scaffold because of its inherent growth proteins and pliability. METHODS: The skin from the skull of a young sheep was removed to expose the connective tissue. A 2 cm x 2 cm periosteum sample was harvested and divided into 4 groups for different decellularisation treatments: sample A (control) was left untreated, sample B was treated with 100% alcohol, sample C was treated with 100% alcohol and 1% sodium hypochlorite, and sample D was treated with 100% a

Alternated activation with relaxation of periosteum stimulates bone modeling and remodeling.

Gradual elevation of the periosteum from the original bone surface, based on the principle of distraction osteogenesis, induces endogenous hard and soft tissue formation. This study aimed to assess the impact of alternating protocols of activation with relaxation (periosteal pumping) on bone modeling and remodeling. One hundred and sixty-two adult male Wistar rats were used in this study. Four test groups with different pumping protocols were created based on the relaxation applied. Two control groups underwent an activation period without relaxation or only a single activation. One group was

Skull Subsidence due to Periosteum Defect following Craniotomy in a Child.

We report a case of a 7-year-old child with a cranial deformity secondary to a craniotomy for an intracranial hematoma. He suffered from an acute epidural hematoma with a lineal fracture of the right temporal bone following a severe head injury. A large question mark-shaped skin flap with the periosteum and temporal muscle was created for a decompressive craniectomy; however, neither the acute epidural hematoma nor brain swelling was severe, and we performed a small craniotomy compared with the skin flap without a decompressive craniectomy. Eighteen months after the operation, he complained of

Uncovering the periosteum for skeletal regeneration: the stem cell that lies beneath.

The cartilage- and bone-forming properties of the periosteum have long since been recognized. As one of the major sources of skeletal progenitor cells, the periosteum plays a crucial role not only in bone development and growth, but also during bone fracture healing. Aided by the continuous expansion of tools and techniques, we are now starting to acquire more insight into the specific role and regulation of periosteal cells. From a therapeutic point of view, the periosteum has attracted much attention as a cell source for bone tissue engineering purposes. This interest derives not only from t

Penile torsion repair by suturing tunica albuginea to the pubic periosteum.

PURPOSE: The aim of the study was to evaluate the effectiveness of a new technique to repair penile torsion in children. METHODS: Eighteen boys with penile torsion were evaluated in our department from 1989 to 2003. Eleven had associated hypospadias, 4 had chordee without hypospadias, and 3 had torsion only. The direction of rotation was counterclockwise in 16 cases and clockwise in 2 cases. Torsion was repaired in 6 patients by degloving the penis and reattaching the dartos and the skin (comparative group). In 11 patients (1 with prior repair), the torsion was corrected by suturing the later

A bilayer hydrogel mimicking the periosteum-bone structure for innervated bone regeneration.

In bone tissue, nerves are primarily located in the periosteum and play an indispensable role in bone defect repair. However, most bone tissue engineering approaches ignored the reconstruction of the nerve network. Herein, we aimed to develop a bilayer hydrogel simulating periosteum-bone structure to induce innervated bone regeneration. The bottom "bone" layer consisted of gelatin methacryloyl (GelMA), poly(ethylene glycol) diacrylate (PEGDA), and nano-hydroxyapatite (nHA), whereas the upper "periosteum" layer consisted of GelMA, sodium alginate (SA) and MgCl2. The mechanical properties of the

Review of the Literature on the Current State of Periosteum-Mediated Craniofacial Bone Regeneration.

STUDY DESIGN: This is an article review on the current state of periosteum-mediated bone regeneration (PMBR). It is a known mandibular reconstruction option in children, and though poorly understood and unpredictable, the concerns of developmental changes to donor and recipient tissues shared by other treatment options are nonexistent. The definitive role of periosteum during bone regeneration remains largely unknown. OBJECTIVE: The objective is to review the literature on the clinical and molecular mechanism evidence of this event. METHODS: Our search methodology was modeled after the PRISM
