Online Monitoring and Control of Methanol Concentration in Pichia Pastoris Fermentations.

Culture broth methanol concentration is the key parameter in the production of recombinant proteins by using Pichia pastoris (Komagataella phaffii) as a cell factory under the regulation of the methanol-inducible Alcohol Oxidase 1 (AOX1) promoter (PAOX1) or other alternative promoters induced directly or indirectly by methanol. Accordingly, the implementation of an efficient methanol monitoring and control system is critical to guarantee the proper methanol concentration toward achieving an efficient and robust production while assuring reproducible bioprocessing conditions.

Expression of functional human sialyltransferases ST6GalNAc5 and ST6GalNAc6 in Pichia pastoris.

The two sialyltransferases in the ST6GALNAC subfamily (EC 2.4.99.-; CAZy family GT29), ST6GalNAc5 and ST6GalNAc6, catalyze the formation of the linkage from the sialic acid moiety to the C6 position of N-acetylgalactosamine (GalNAc) as well as to N-acetylglucosamine (GlcNAc), and are known as α-2,6-sialyltransferases. This activity is interesting for the synthesis of the disialylated oligosaccharide disialyllacto-N-tetraose (DSLNT). Human sialyltransferases ST6GalNAc5 and ST6GalNAc6 produced in HEK293 cells are commercially available at a smaller scale. In this study, we demonstrated that ST6G

Recent advances in the production of recombinant subunit vaccines in Pichia pastoris.

Recombinant protein subunit vaccines are formulated using defined protein antigens that can be produced in heterologous expression systems. The methylotrophic yeast Pichia pastoris has become an important host system for the production of recombinant subunit vaccines. Although many basic elements of P. pastoris expression system are now well developed, there is still room for further optimization of protein production. Codon bias, gene dosage, endoplasmic reticulum protein folding and culture condition are important considerations for improved production of recombinant vaccine antigens. Here w

Expression and characterization of a synthetic protein C activator in Pichia pastoris.

Protein C activators are proteases that activate protein C in the mammalian coagulation system. A reptilian protein C activator is a critical component in current functional assays for protein C, its cofactor protein S, as well as for the overall status of the protein C pathway. We have constructed a synthetic gene for a protein C activator, based on a published snake-venom polypeptide sequence. This recombinant protein C activator was expressed in Pichia pastoris as a secreted glycoprotein (ILPCA) using the AOX1 promoter and the alpha-factor signal sequence. A fermentation protocol was develo

Methanol-based biosynthesis of p-coumaric acid by engineered Pichia pastoris.

p-Coumaric acid (p-CA) is a key aromatic precursor for the biosynthesis of flavonoids, stilbenoids, and other high-value phenylpropanoids. While microbial production of p-CA typically relies on sugar-based substrates, methanol offers a sustainable and cost-effective alternative, though its use for aromatic biosynthesis remains unexplored. Here, we report the first de novo production of p-CA from methanol using engineered methylotrophic yeast Pichia pastoris. Through heterologous expression of a tyrosine ammonia-lyase and implementing a balanced push-pull strategy in the shikimate pathway using

Random mutagenesis platform for accelerated genome evolution (RAMPAGE) of Pichia pastoris.

The biological conversion of methanol into valuable chemicals represents a promising route for achieving global carbon neutrality. However, the rational engineering of complex industrial traits in microbial cell factories is hindered by an incomplete understanding of cellular metabolism and regulation. In this study, we developed a Random Mutagenesis Platform for Accelerated Genome Evolution (RAMPAGE) for the yeast Pichia pastoris (Komagataella phaffii). This platform enables continuous genome-wide diversification and facilitates the rapid evolution of superior phenotypes under defined selecti

Promoter engineering for enhanced 3-hydroxypropionic acid production in Pichia pastoris.

Enabling tools are essential for facilitating the methanol bioconversion in Pichia pastoris. However, there is still a relative lack of promoters that can stably express high levels without being affected by the carbon source, which hinders the construction and modification of cell factories containing long metabolic pathways. This study mapped a gene expression intensity library of central metabolic pathways in P. pastoris under methanol and glucose conditions. Through modification of the upstream sequences of promoters, an artificial promoter P S2 was developed with a strong intensity up to

Multiplex Marker-Less Genome Integration in Pichia pastoris Using CRISPR/Cas9.

Pichia pastoris is known for its excellent protein expression ability. As an industrial methyl nutritional yeast, it can effectively utilize methanol as the sole carbon source, serving as a potential platform for C1 biotransformation. Unfortunately, the lack of synthetic biology tools in P. pastoris limits its broad applications, particularly when multigene pathways should be manipulated. Here, the CRISPR/Cas9 system is established to efficiently integrate multiple heterologous genes to construct P. pastoris cell factories. In this protocol, with the 2,3-butanediol (BDO) biosynthetic pathway a

Characterization of Ceriporiopsis subvermispora bicupin oxalate oxidase expressed in Pichia pastoris.

Oxalate oxidase (E.C. 1.2.3.4) catalyzes the oxygen-dependent oxidation of oxalate to carbon dioxide in a reaction that is coupled with the formation of hydrogen peroxide. Although there is currently no structural information available for oxalate oxidase from Ceriporiopsis subvermispora (CsOxOx), sequence data and homology modeling indicate that it is the first manganese-containing bicupin enzyme identified that catalyzes this reaction. Interestingly, CsOxOx shares greatest sequence homology with bicupin microbial oxalate decarboxylases (OxDC). We show that CsOxOx activity directly correlates

Proteomic and Functional Profiling of Trichoderma reesei, Pichia guilliermondii, and Hericium erinaceus.

Fungi relevant to food systems and biotechnology differ in physiology, yet their condition-specific intracellular proteomes have rarely been compared using a common workflow. We hypothesized that harmonized extraction and data-independent acquisition mass spectrometry would reveal distinct functional signatures across Trichoderma reesei, Pichia guilliermondii, and Hericium erinaceus. Overall, 199, 889, and 1456 proteins were identified, respectively, revealing species-associated differences in protein biosynthesis, bioenergetics, redox metabolism, carbohydrate catabolism, proteostasis, and RNA

Expanding the Genetic Toolkit of Pichia kudriavzevii E1 for Enhanced Mycoprotein Production.

Pichia kudriavzevii is gaining attention as a non-conventional yeast with significant industrial potential, owing to its robust tolerance to low pH and diverse process-related stresses. However, the lack of a reliable episomal expression system and the scarcity of well-characterized promoters currently impede predictable gene expression and modular metabolic engineering in this workhorse. Here, we report development of the stable episomal expression vector for P. kudriavzevii E1. This was achieved through identification of a functional autonomously replicating sequence from the endogenous geno
