Decoding the role of RNA sequences and their interactions in influenza A virus infection and adaptation.

Influenza viruses (types A, B, C, and D) belong to the family orthomyxoviridae. Out of all the influenza types, influenza A virus (IAV) causes human pandemic outbreaks. Its pandemic potential is predominantly attributed to the genetic reassortment favored by a broad spectrum of host species that could lead to an antigenic shift along with a high rate of mutations in its genome, presenting a possibility of subtypes with heightened pathogenesis and virulence in humans (antigenic drift). In addition to antigenic shift and drift, there are several other inherent properties of its viral RNA species

Long non-coding RNAs are associated with Seneca Valley virus infection.

Sporadic outbreaks of Seneca Valley virus (SVV) have been detected in recent years causing huge economic losses to the pig industry. SVV infection can lead to redness and fever of the mouth, nose or hoof wall, ulcerative injury and inflammation in pigs. Although long non-coding RNAs (lncRNAs) have been shown to play an important role in antiviral and inflammatory regulation, how lncRNAs regulate and induce SVV infection inflammation remains unclear. Here, we found the differential expression of 1332 lncRNAs and 3299 mRNAs in SVV-infected ST cells using RNA-seq. Functional annotation analysis r

Performance of two commercial assays for detecting hepatitis E virus RNA in acute or chronic infections.

We assessed the performance of the Ceeram and Altona assays, the first two commercially available hepatitis E virus (HEV) RNA assays, using serial dilutions of 4 HEV-positive reference samples (genotypes 3a, 3c, 3e, and 3f). Both assays provided good analytical sensitivity and high reproducibility for detecting genotype 3 HEV RNA.

Analysis of expression profiles of long noncoding RNAs and mRNAs in brains of mice infected by rabies virus by RNA sequencing.

Rabies, caused by rabies virus (RABV), is still the deadliest infectious disease. Mechanism of host immune response upon RABV infection is not yet fully understood. Accumulating evidences suggest that long noncoding RNAs (lncRNAs) plays key roles in host antiviral responses. However, expression profile and function of lncRNAs in RABV infection remain unclear. In the present study, expression profile of lncRNAs and mRNAs profiles were investigated in RABV-infected brain tissues of mice by RNA sequencing. A total of 140 lncRNAs and 3,807 mRNAs were differentially expressed in RABV-infected anima

[Activity and properties of RNA-dependent RNA polymerase induced in cell cultures infected with Venezuelan equine encephalomyelitis virus].

The activity and properties of RNA-dependent RNA-polymerase in cells infected with Venezuelan equine encephalomyelitis virus were studied. The maximum activity of the enzyme was found 4-5 hours after infection. The effect of two synthetic drugs (rimantadine and diacetylaminocyclohexenon) with antiviral activity against some viruses on RNA-polymerase was studied. Rimantadine was shown to have no effect on the synthesis and activity of the enzyme. Diacetylaminocyclohexenon added into the growth medium was found to inhibit production of RNA-polymerase by 35-40% without reducing the activity of th

Hepatitis C virus RNA and antibody response in the clinical course of acute hepatitis C virus infection.

Hepatitis C virus RNA, anti-hepatitis C virus immune response and biochemical markers of liver injury were investigated in 17 patients with acute non-A, non-B hepatitis. At the first observation, 1 to 3 wk from the clinical onset, all patients had hepatitis C virus RNA in their serum, and most (15 of 17) were positive for second-generation anti-hepatitis C virus enzyme immunoassay. Follow-up serum samples were available for 10 patients. The rate of recombinant immunoblot assay-confirmed anti-hepatitis C virus enzyme immunoassay reactivities increased from 67% in the first 3 wk to 86% after 21

Serum Hepatitis B Virus RNA: A New Potential Biomarker for Chronic Hepatitis B Virus Infection.

Chronic hepatitis B infection is one of the major etiological causes of liver failure, cirrhosis, and hepatocellular carcinoma (HCC) worldwide. This condition cannot be completely cured by currently available drugs due to the persistent existence of hepatitis B virus (HBV) covalently closed circular DNA (cccDNA), the bona fide transcription template for HBV RNAs, in infected hepatocytes. Because quantifying cccDNA per se requires an invasive procedure, serum biomarkers reflecting intrahepatic cccDNA activity are warranted. Recently, a growing body of research suggests that the circulating HBV

[Virus-specific RNA synthesis in a cell culture in acute and chronic infection with the tick-borne encephalitis virus].

Virus particles produced in acute and chronic infection of cell cultures with tick-borne encephalitis virus (TBE) and examined by centrifugation in sucrose density gradient had the buoyant density of 1.19 g/ml and sedimentation constant 290 S. Studies of the synthesis of virus-specific RNAs in the cytoplasm of TBE virus chronically infected cells revealed synthesis of all RNA classes typical of TBE virus reproduction in acutely infected cells. The difference was in the high percentage of polyadenylation of intracellular virion RNA in chronic infection (30%) as compared with polyadenylation of

Characterization of virus-specific RNAs from subacute sclerosing panencephalitis virus-infected CV-1 cells.

CV-1 cells infected with subacute sclerosing panencephalitis (SSPE) virus incorporated uridine-(3)H into at least four virus-specific RNA components in the presence of actinomycin D. The component sedimenting fastest had a sedimentation coefficient of 50s corresponding to a molecular weight of 6 x 10(6). The other three RNA components have sedimentation constants of 35s, 22s, and 18s corresponding to molecular weights of 2.5 x 10(6), 1.0 x 10(6), and 0.75 x 10(6), respectively. The base composition of the 50s RNA is distinct from that of cellular RNA and comparable with base compositions of vi

A comparative analysis of cell death signalling in RNA and DNA virus infections and potential inhibitory strategies.

The recent COVID-19 pandemic and the epidemic outbreak of Mpox have demonstrated that viruses, despite their apparently simple structure, are sophisticated microorganisms that have evolved complicated pro-survival mechanisms. Their potential to manipulate cell death pathways can affect and determine the outcome of the infection cycle and the host's immune response. RNA viruses typically display a more acute and aggressive nature, as they often promote cell death. In this way, they proliferate and release their virions to infect adjacent cells and perpetuate their life cycle. Common consequence

Stress granules formed during infections with RNA viruses have significant virus-specific differences in generation and composition.

Eukaryotic cells evolved a cellular stress response to cope with extrinsic and intrinsic stress stimuli including virus infections. The major result of this response is the shutdown of bulk translation to prevent damage and allow the reprogramming of translation towards stress-resolving pathways. The resulting translationally stalled mRNA and associated proteins are accumulated in membrane-less cytosolic condensates called stress granules (SG). While the inhibitory effect of translation arrest on virus growth is well established, the role of SGs in the cellular defense against viruses is still
