Nature of inhibition of renal aspartate reabsorption in experimental Fanconi syndrome.

Administration of heavy metals or maleic acid induces a condition resembling the Fanconi syndrome. In the case of metals, an apparently uncompetitive inhibition of reabsorption of amino acids was observed 2 days or longer after injection into rabbits (E. C. Foulkes and S. Blanck, Toxicol. Appl. Pharmacol. 64, 103-107, 1982). Transport of aspartate is now shown to be similarly inhibited by maleic acid and cephaloridin. Because maleic acid, as well as p-chloromercuribenzoate (PCMB) and Cd (in presence of mercaptoethanol), exerts an apparent uncompetitive effect within minutes of injection, the i

Filtered bicarbonate and plasma pH as determinants of renal bicarbonate reabsorption.

To examine if bicarbonate reabsorption varies with filtered bicarbonate and plasma pH, we infused anesthetized dogs i.v. with sodium chloride and sodium bicarbonate to alter plasma bicarbonate concentration (PHCO3) without changing hematocrit. Examinations in five dogs over a wide range of glomerular filtration rates (GFR) during ethacrynic acid infusion showed that bicarbonate reabsorption at equal filtered load and equal plasma pH of 7.5 was not significantly changed by increasing PHCO3 from 30.2 +/- 0.4 to 55.2 +/- 0.6 mM and PCO2 from 33.8 +/- 0.7 to 74.1 +/- 2.1 mm Hg. A micropuncture study of renal lithium reabsorption: effects of amiloride and furosemide.

The validity of the lithium clearance technique as a measure of end-proximal fluid delivery was assessed using micropuncture in sodium-replete, Inactin-anesthetized Sprague-Dawley rats. Three groups of animals were used: controls, amiloride treated, and furosemide treated. Diuretic-induced salt and water losses were replaced. Fractional lithium excretion (FELi) was 0.23 +/- 0.01, 0.24 +/- 0.02, and 0.40 +/- 0.03 in the control, amiloride, and furosemide groups, respectively. In each group, the tubular fluid-to-plasma lithium concentration ratio at the end of the proximal convoluted tubule (PCT

Early adaptation of renal magnesium reabsorption in response to magnesium restriction.

Dietary magnesium restriction with hypomagnesia is normally associated with diminished urinary magnesium excretion. Young rats were pair fed control magnesium diets (0.05% MgSO4) and magnesium-restricted diets (less than 0.01% Mg), and tubular magnesium reabsorption was assessed to determine the importance of filtered load (plasma magnesium) and to establish the presence of cellular adaptation of magnesium transport. Urinary magnesium excretion decreased from 17.2 +/- 2.7 to 5.9 +/- 1.2% over 20 h on low-magnesium diets without a change in plasma concentration (0.61 +/- 0.02 mM). This cellular

The influence of extracellular volume expansion on renal phosphate reabsorption in the dog.

Extracellular volume expansion (ECVE) was produced, by normal saline infusion, in five normal and six thyroparathyroidectomized anesthetized dogs while glomerular filtration rate was reduced by the inflation of an intra-aortic balloon located above the renal arteries. The effect of ECVE on the maximum renal tubular reabsorptive capacity of phosphate (phosphate Tm) was also evaluated in five additional dogs. During ECVE, phosphate excretion increased both in normal and thyroparathyroidectomized dogs, and a direct and significant correlation was found between the fractional excretion of phosphat

Urine flow dependence of renal clearance and interrelation of renal reabsorption and physicochemical properties of drugs.

A physiologically based model is presented for the analysis of the reabsorption kinetics of drugs in the renal tubules. The two reabsorption parameters, AR(I) X Pe and sigma, expressing the permeability of a drug through lipoidal membranes and the reflection coefficient, respectively, are obtained on the basis of this model using the results of clearance experiments in rats. These two reabsorption parameters are compared among four drugs, sulfanilamide, sulfamethizole, theophylline, and ethanol. Urine flow dependence of the percentage of reabsorbed drugs is reasonably expressed by the model. M

Plasma cAMP: a hepatorenal link influencing proximal reabsorption and renal hemodynamics?

The existence of a hepatorenal link is suggested by several pathophysiological observations (indirect actions of glucagon on the kidney, hepatorenal syndrome), but the nature of this link remains unidentified. We propose that extracellular circulating cyclic AMP could be this link. Cyclic AMP (cAMP) is the intracellular second messenger of glucagon (G) action in the liver, and this organ is known to release cAMP in the blood in relatively large amounts after G administration. On the other hand, the proximal tubule (mainly the pars recta) is known to take up cAMP through the organic acid transp

The absence of renal bicarbonate reabsorption maxima during carbonic anhydrase inhibition.

Present studies were performed to examine the effect of carbonic anhydrase inhibition on bicarbonate reabsorption under experimental conditions where extracellular fluid volume expansion was minimized. The data showed no tubular maximum for bicarbonate reabsorption in the normal state or after carbonic anhydrase inhibition. The data support a model which assumes that the bicarbonate reabsorption is accomplished by two processes. One process is independent of protonation from H+ secretion into the tubule and probably involves reabsorption of bicarbonate ion as such. This process has an unlimite

Mineralocorticoid-resistant renal hyperkalemia without salt wasting (type II pseudohypoaldosteronism): role of increased renal chloride reabsorption.

A rare syndrome has been described in which mineralocorticoid-resistant hyperkalemia of renal origin occurs in the absence of glomerular insufficiency and renal sodium wasting and in which hyperchloremic acidosis, hypertension, and hyporeninemia coexist. The primary abnormality has been postulated to be a defect of the potassium secretory mechanism of the distal nephron. The present studies were carried out to investigate the mechanism of impaired renal potassium secretion in a patient with this syndrome. When dietary intake of sodium chloride was normal, renal clearance of potassium was subno

The relationship between the renal reabsorption of cysteine and the lowered urinary pH in diabetics.

BACKGROUND/AIMS: In diabetic patients, reduced urinary pH (UpH) is a predictive factor for cardiorenal-vascular disorders. Synthesis of glutathione, an anti-oxidative stress substance, is induced to counteract renal oxidative stress. UpH declines as glutamate is consumed, as does the synthesis of ammonia from glutamate. Glutathione is synthesized from glutamate and cysteine; however, in diabetes, the relationship between lowered UpH and the roles of renal amino acids is unknown. We, therefore, examined the relationship between amino-acid kinetics, UpH, and renal function. METHODS: This cross-

A micropuncture study of renal tubular lithium reabsorption in sodium-depleted rats.

1. The marked reduction in fractional lithium excretion (FELi) which accompanies chronic sodium depletion was investigated using free-flow micropuncture in anaesthetized rats which had been maintained in a sodium-depleted state for 8-10 days. 2. Compared with previous values in sodium-replete rats, sodium depletion was associated with small reductions in total and superficial nephron glomerular filtration rate and enhanced fractional reabsorption of water, sodium and lithium in the proximal convoluted tubule. 3. In untreated (sodium-depleted) rats, fractional deliveries of lithium (FDLi) to th

Effects of insulin and glucose on renal phosphate reabsorption: interactions with dietary phosphate.

Both insulin deficiency and glycosuria are known to inhibit the tubular reabsorption of phosphate. This inhibition has previously been evaluated either in the fasted state or on a normal phosphate diet. The goal of this study was to evaluate how dietary phosphate depletion affected the relative effects of insulin deficiency and glycosuria on the tubular reabsorption of phosphate. Rats were maintained on either a low- (0.03%) or normal (0.8%) phosphate diet. After 5 days, one half of the animals in each group received streptozotocin to induce short-term insulin deficiency, whereas the other hal
