This study investigates the use of natural deep eutectic solvents (NADES) for environmentally friendly extraction of pectic polysaccharides from carrot pomace, a low-cost agro-industrial byproduct. Two response surface methodologies, Box-Behnken design (BBD) and I-optimal design (IOD), were compared to optimize the extraction using a ternary NADES system composed of choline chloride, glucose, and citric acid (ChCl/Glc/CA). The IOD model showed superior predictive performance (R2 = 0.95 for yield; R2 = 0.81 for uronic acid content) and greater validity across a broader parameter range. Under op

Colorectal cancer (CRC) is the third most common cancer worldwide. 5-fluorouracil (5-FU) is a first-line chemotherapeutic drug for the treatment of CRC; however, its selectivity is poor, it has serious side effects, and its long-term use is prone to drug resistance. In this study, the ultrasonic-assisted cellulase method was used to extract polysaccharides from Cibotium barometz. Under the optimum extraction conditions, the highest polysaccharide extraction rate reached 14.34 ± 0.41 %, and the crude polysaccharide of C. barometz (CBP) was extracted under these conditions. Furthermore, an acidi

Natural polysaccharides from Siegesbeckia orientalis L. have been reported to exhibit promising bioactivities. To enhance extraction efficiency, low-temperature plasma-assisted extraction was optimized for S. orientalis L. polysaccharides using single-factor experiments and response surface methodology. Column chromatography purified a homogeneous SIE-III fraction, followed by structural characterization. Optimal parameters were 80 kV discharge voltage, 153 Hz frequency, and 109 s treatment time, under which the polysaccharide yield reached 15.68%, significantly higher than that of the convent

Pectic galactan is a rhamnogalacturonan-I side chain, composed of β-(1→4)-linked D-galactose residues. Its multifunctional role in cell wall assembly and properties has been proposed in various developmental contexts. In this study, we investigated the role of galactan in pea (Pisum sativum syn. Lathyrus oleraceus). First, we used glycan profiling to examine stems with different auxin-induced elongation rates for changes in cell wall composition. The galactan epitopes, recognised with the LM5 monoclonal antibody, were more abundant in the convex, more elongated part of the pea stem. This findi

Pectic polysaccharides in A. marina fruit exhibit excellent prebiotic effects and may be the key to treating constipation and dysentery; however, their detailed structures still need to be clarified. In this study, we obtained two pectic polysaccharide fractions (AMFPs-3-2 and AMFPs-3-4) from this fruit for the first time and comprehensively investigated their fine structures using chemical methods, GC-MS, and 1D and 2D-NMR. AMFPs-3-2 was identified as a type I rhamnogalacturonan (RG-I) with a backbone composed of →2)-α-Rhap-(1 → 4)-α-GalpA-(1→, and the side chains mainly consisted of arabinan

Previous work has reported the results of a multidisciplinary effort producing a proof-of-concept on the use of pectic polysaccharides in the surface modification of medical devices. This study was designed to learn more about the capability of engineered rhamnogalacturonan-I (RG-I) fractions of apple pectin to control bone cell and macrophage behavior. Thermanox or polystyrene Petri dishes were surface modified with two different modified hairy regions (MHRs) obtained by different enzymatic liquefaction processes of apples differing in relative amounts and lengths of their neutral side chains

Peach pectin is a promising functional polysaccharide, yet the structure-activity associations among its fractions remain poorly defined. Here, four structurally distinct peach pectin fractions, water-soluble (WSP), chelator-soluble (CSP), sodium carbonate-soluble (SSP), and an enzymatically modified fraction (ES) with targeted homogalacturonan fragmentation, were evaluated in cyclophosphamide-induced immunosuppressed female mice. Among them, SSP exerted the most comprehensive benefits. Specifically, SSP treatment restored cytokine balance; increased red blood cell count and hemoglobin levels

Inflammatory bowel disease (IBD) induces irreversible intestinal damage and is a pivotal risk factor for colorectal cancer. Sanguisorba officinalis extracts have been shown to alleviate intestinal inflammation, yet their underlying structural basis is still elusive. Hence, we proposed that the polysaccharide from Sanguisorba officinalis may exert a potential anti-inflammation activity for IBD. Herein, the structure of an active pectin 147-02-S2 (88.37 kDa), featuring a backbone of rhamnogalacturonan-I and xylogalacturonan, isolated from Sanguisorba officinalis roots was elucidated. Its branche

Bacterial cellulose (BC) is a subject of interest for researchers due to its advantageous characteristics, including a straightforward manufacturing process, biocompatibility, and extensive modification potential. The hydrophilic nature of the material is beneficial in some applications, yet a limiting factor in others. This study aimed to develop BC-based materials with goFogureod moisture resistance. The modification of bacterial cellulose (BC) using apple powder, stearic acid, or a combination of these modifiers resulted in the formation of a range of materials, some of which had their surf

Acute lung injury (ALI) is a critical respiratory disorder that severely endangers public health. As natural macromolecular active compounds with outstanding biosafety and negligible toxic side effects, natural polysaccharides exhibit prominent development potential in the field of anti-ALI pharmacological research. This review finds that natural polysaccharides with ALI activity mostly possess a molecular weight below 20 kDa, and core monosaccharide residues mainly include glucose, galactose, mannose, rhamnose and galacturonic acid, and the predominant polysaccharide types are rhamnogalacturo

RGI (rhamnogalacturonan-I) pectin, a polyhydroxy-carboxylic acid polymer abundant in neutral sugar side chains, exhibits thermal aggregation during heat drying which leads to its poor solubility. It is hypothesized that insufficient electronic repulsion among the neutral sugar side chains causes their entanglements. Therefore, our purpose is to offer other polysaccharides to the system as steric scaffolds to inhibit aggregation potentially. Various functional polysaccharides with distinct charge characteristics, including chitosan, glucan, commercial pectin, κ-Carrageenan, and ι-Carrageenan, w

Gardenia jasminoides Ellis is an important medicinal and edible resource. However, the structure-activity relationships underlying the immunomodulatory activity of its polysaccharides remain incompletely understood. Herein, we aimed to characterize the structure of a novel pectin from Gardenia fruit and clarify its immunomodulatory mechanism. A homogeneous polysaccharide, GJP70-0.2 (Mw = 54.5 kDa), was isolated and purified from gardenia fruit. Structural analysis confirmed that it was a pectic polysaccharide characterized by a notably homogalacturonan (HG)-dominant architecture with minor rha
