Burden of disease & molecular epidemiology of group A rotavirus infections in India.

Rotavirus is the major cause of severe dehydrating diarrhoea in young children worldwide. Considerable research has been carried out on rotavirus disease in India. This review collated data from 46 epidemiological studies to determine rotavirus positivity rates and genotypes of infecting rotavirus strains from various settings in India. Studies on diarrhoea presenting to hospitals, neonatal rotavirus infections, symptomatic and asymptomatic infections in the community and nosocomial enteric infections were included. Rotavirus positivity rates varied greatly between different settings - diarrho…

Quercetin, a flavonoid, combats rotavirus infection by deactivating rotavirus-induced pro-survival NF-κB pathway.

Rotavirus (RV) is the leading cause of acute gastroenteritis and watery diarrhea in children under 5 years accounting for high morbidity and mortality in countries with poor socioeconomic status. Although vaccination against RV has been implemented in more than 100 countries, the efficacy of vaccine has been challenged in low-income settings. The lack of any FDA-approved drug against RV is an additional concern regarding the treatment associated with rotavirus-induced infantile death. With the purpose for the discovery of anti-RV therapeutics, we assessed anti-rotaviral potential of quercetin,

Rotavirus infection induces glycan availability to promote ileum-specific changes in the microbiome aiding rotavirus virulence.

Multiple studies have identified changes within the gut microbiome in response to diarrheal-inducing bacterial pathogens. However, examination of the microbiome in response to viral pathogens remains understudied. Compounding this, many studies use fecal samples to assess microbiome composition; which may not accurately mirror changes within the small intestine, the primary site for most enteric virus infections. As a result, the functional significance of small intestinal microbiome shifts during infection is not well defined. To address these gaps, rotavirus-infected neonatal mice were exami

[Prospective study of rotavirus infection in a maternity unit. Demonstration of a nosocomial infection].

Eighty-eight children born at the maternity hospital in Saint-Germain-en-Laye between May 24 and June 7, 1983 were followed clinically, with a special supervision concerning stools, weight curves and the way of feeding. Stool samplings looking for Rotavirus were performed in all the children and their mothers, at the 3rd and 6th days of life. No mother was found with Rotavirus infection. In neonates, Rotavirus excretion was significantly related to a slow down in weight curves and the occurrence of diarrhea. All rotaviruses had the same electrophoretype. Breast-feeding had an undeniable protec

Rotavirus disrupts cytoplasmic P bodies during infection.

Cytoplasmic Processing bodies (P bodies), the RNA-protein aggregation foci of translationally stalled and potentially decaying mRNA, have been reported to be differentially modulated by viruses. Rotavirus, the causative agent of acute infantile gastroenteritis is a double stranded RNA virus which completes its entire life-cycle exclusively in host cell cytoplasm. In this study, the fate of P bodies was investigated upon rotavirus infection. It was found that P bodies get disrupted during rotavirus infection. The disruption occurred by more than one different mechanism where deadenylating P bod

Descriptive epidemiology of rotavirus infection in a community in North India.

In India, rotavirus infections cause the death of 98621 children each year. In urban neighbourhoods in Delhi, children were followed up for 1 year to estimate the incidence of rotavirus gastroenteritis and common genotypes. Infants aged f1 week were enrolled in cohort 1 and infants aged 12 months (up to +14 days) in cohort 2. Fourteen percent (30/210) gastroenteritis episodes were positive for rotavirus. Incidence rates of rotavirus gastroenteritis episodes in the first and second year were 0.18 [95% confidence interval (CI) 0.10–0.27] and 0.14 (95% CI 0.07–0.21) episodes/child-year, respectiv…

Clinical neonatal rotavirus infection: association with necrotising enterocolitis.

BACKGROUND: Rotavirus is the most important aetiological agent causing severe gastroenteritis in children <2 years of age in South Africa and worldwide. Most endemic neonatal nursery strains are thought to be asymptomatic. However, serious conditions have been reported to be associated with rotavirus infection, such as necrotising enterocolitis (NEC), diffuse intravascular coagulopathy, pneumonia, apnoea and seizures. METHODS: We studied newborns needing screening for sepsis in our Neonatal Unit. Rotavirus screening was included in the septic screen. The clinical signs and symptoms were studi

Rotavirus infection in four states in north-western Nigeria.

BACKGROUND: Rotaviruses are associated with approximately 611,000 deaths worldwide and with 33,000 deaths in Nigeria in children < 5 years of age annually However, limited data exit on rotavirus (RV) infection in North-western Nigeria. This study surveyed RV infection in four states in Northwestern Nigeria. METHODS: During July 2002 to July 2004, 1063 (869 diarrhoeic and 194 control) stool samples were collected from children <5 years of age presenting with diarrhoea in clinics/hospitals in Kaduna, Kebbi, Sokoto and Zamfara States. The stools were analysed for RV antigen and the RV positive s

Central nervous system involvement in neonatal rotavirus infection.

Rotavirus is an ubiquitous virus associated with severe gastro-enteritis in children under 2 years of age. However, serious conditions, including central nervous system involvement, have been associated with rotavirus infection. We investigated the clinical signs and symptoms of rotavirus infection in 156 neonates, specifically focusing on neurological signs. Neurological signs identified were apnoea, jitteriness, convulsions and poor sucking. Of the 156 babies, 66 were rotavirus-positive and 90 rotavirus-negative. In the rotavirus-positive group, 19.7% manifested apnoea compared with 22.2% in

Molecular epidemiology of human rotavirus infection in Coro, Venezuela.

From December 1984 to December 1987, rotaviruses were detected in 115 (24%) of 470 hospitalized children, by electrophoretic analysis of viral RNA. Three peaks of increased incidence were observed, coincident with the cooler months of each year. Rotavirus was found in 31% of the children with diarrhea and in 47% of those who were severely dehydrated. Vomiting was significantly associated with rotavirus shedding in diarrheic children. Eleven electropherotypes were identified, showing a sequential pattern of appearance. Strains exhibiting a "short" pattern were present in 10% of the positive cas

[Mixed coli-rotavirus and Cryptosporidium infection in newborn piglets].

Complex morphologic, parasitologic, virologic, and bacteriologic studies on pigs and fecal samples from pigs were carried out on a farm where an enteritis infection in newborn pigs was recorded. A mixed coli-rotavirus and cryptosporidium infection was established in 10--20-day-old pigs of the Camborough hybrid with a clinically manifested enteritis syndrome. Morphologically, the cryptosporidium component of the infection substantiated atrophic and dystrophic changes in the intestinal epithelium. Developmental forms of the parasite were also found in the border wall of the enterocytes.

Epidemiological aspects of rotavirus infection in young Gambian children.

Rotavirus gastro-enteritis in young Gambian children has its maximum impact on infants after the age of one month, in whom it produces short, well-defined annual winter epidemics with clinical dehydration in up to 18% of those infected. Sporadic infection was observed in neonates who were often asymptomatic, throughout one year but not in the subsequent year. In two consecutive years studied there was a major change from subgroup I, serotype 2 to subgroup II, serotypes 1 and 3. This could have contributed to the failure of children to develop protective immunity against sequential disease foll

Segmented Filamentous Bacteria Prevent and Cure Rotavirus Infection.

Rotavirus (RV) encounters intestinal epithelial cells amidst diverse microbiota, opening possibilities of microbes influencing RV infection. Although RV clearance typically requires adaptive immunity, we unintentionally generated RV-resistant immunodeficient mice, which, we hypothesized, reflected select microbes protecting against RV. Accordingly, such RV resistance was transferred by co-housing and fecal transplant. RV-protecting microbiota were interrogated by heat, filtration, and antimicrobial agents, followed by limiting dilution transplant to germ-free mice and microbiome analysis. This

[Rotavirus infection in Brazil: epidemiology and challenges for its control].

Worldwide, rotaviruses account for 600,000 to 870,000 deaths per year among infants and young children. In Brazil, rotaviruses were first seen in 1976 by scanning electron microscopy of stool samples from diarrheic infants in Belém, Pará. Hospital-based studies have shown that rotaviruses are associated with 12-42% of cases of acute diarrhea. In addition, community-based studies yielded an average of 0.25 rotavirus-related diarrheal episodes per child per year. G types 1 to 4 account for about two-thirds of circulating strains, but the (unusual) P[8],G5 genotype has been claimed to cause over

Persistent rotavirus infection in mice with severe combined immunodeficiency.

Rotaviruses are important pathogens of human infants and the infants of many animal species. The disease produced by these viruses can be described as an acute, self-limiting diarrheal disease, with virus replication localized to the differentiated epithelial enterocytes of the small intestine. Immunologically normal infants shed virus for approximately 5 to 12 days after the onset of infection. Recently, it has been shown that rotavirus can produce a chronic infection in severely immunocompromised children, with virus shedding and intermittent diarrhea lasting from 6 weeks to 2 years (G. A.
