A natural brake tunes early calcium signaling and salt tolerance in rice.

In this issue of Developmental Cell, Feng et al.1 identify TOLS1, a non-canonical GAP-like protein, as a naturally variable negative regulator of rice salt tolerance. By linking TOLS1 to OsRacB and cytosolic Ca2⁺ signaling, the study reveals how crop alleles can quantitatively tune an early cellular stress response.

The Rice Protein Kinase OsCIPK7 Enhances Salt Tolerance by Regulating Potassium Homeostasis.

Salt stress disrupts ion homeostasis, leading to toxic sodium accumulation and reduced potassium uptake. The Calcineurin B-like protein (CBL) and CBL-interacting protein kinase (CIPK) network plays pivotal roles in response to abiotic stresses. Here, the root preferentially expressed OsCIPK7 with significantly salt-induced transcription was identified and functional characterized. Knockout of OsCIPK7 increased sensitivity to salt stress and a pronounced reduction of potassium content in their tissues under salt stress was observed compared to wild type. Decreased allocation of potassium was ob

The COBRA-Like9 from Glycine tabacina enhanced salt tolerance in cultivated soybean.

Soybean is one of the major sources of high-quality protein and oil and plays a crucial role in ensuring nutritional and economic security. The rapidly increasing population, soybean supply-demand imbalance, and yield losses due to soil salinity collectively necessitate the development of salt-tolerant cultivars. Perennial wild Glycine species harbor-rich genetic diversity and constitute a reservoir of stress-tolerant genes. This study systematically characterized the COBL gene family in the salt-resilient Glycine tabacina (Labill.) Benth., a wild soybean that tolerates 500 mM NaCl with 100% s

Bacillus subtilis B21-Mediated Salt Tolerance in Cucumber via Auxin Signalling Activation.

Soil salinization is a major constraint on global crop productivity. While certain plant growth-promoting rhizobacteria (PGPR) improve plant survival under salt stress, their underlying molecular mechanisms remain largely uncharacterised. In this study, a highly halotolerant PGPR strain, Bacillus subtilis B21, was isolated from saline - alkali soils in Ningxia, China, and investigated its mechanism of action in cucumber (Cucumis sativus). Inoculation with B21 significantly promoted root growth, re-established ion and reactive oxygen species (ROS) homoeostasis, and alleviated osmotic stress und

Serum- and glucocorticoid-inducible kinase 1 mediates salt sensitivity of glucose tolerance.

Excess salt intake decreases peripheral glucose uptake, thus impairing glucose tolerance. Stimulation of cellular glucose uptake involves phosphatidylinositide-3-kinase (PI-3K)-dependent activation of protein kinase B/Akt. A further kinase downstream of PI-3K is serum- and glucocorticoid-inducible kinase (SGK)1, which is upregulated by mineralocorticoids and, thus, downregulated by salt intake. To explore the role of SGK1 in salt-dependent glucose uptake, SGK1 knockout mice (sgk1(-/-)) and their wild-type littermates (sgk1(+/+)) were allowed free access to either tap water (control) or 1% sali

Molecular cloning and chromosomal localization of human salt-tolerant protein.

We have isolated a novel human cDNA coding for human salt-tolerant protein (HSTP), that is a homologue of the rat salt-tolerant protein (STP) and may contribute to salt-induced hypertension by modulating renal cation transport. The nucleotide sequence (1988bp) of the HSTP cDNA contains an open reading frame encoding a polypeptide comprising 545 amino acids, two residues fewer than the rat STP cDNA. The predicted amino acid sequence exhibits 92% identity to that of the rat protein. HSTP contains predicted coiled-coil domains and Src Homology 3 domain, and shows a high degree of identity to CIP4

TaSTG4B enhances wheat salt tolerance via regulation of antioxidant and photosynthetic protection pathways.

Soil salinization represents a major abiotic stress constraining wheat yield. Identifying key salt tolerance-related genes and unraveling their regulatory mechanisms is critical for breeding salt-tolerant wheat varieties. In this study, TaSTG4B (Triticum aestivum Salt-Tolerant Gene on 4B, gene ID: TraesCS4B02G042300), a key regulatory gene encoding an oxysterol-binding protein homolog, was jointly identified by ethyl methanesulfonate (EMS)-induced mutagenesis screening and genome-wide association study (GWAS), and systematically elucidated its salt tolerance mechanisms. TaSTG4B exhibits relati

A NAC-MYB-cysteine regulatory module enhances drought and salt tolerance in soybean.

Drought and salinity are two major factors limiting soybean productivity worldwide, and both stresses can negatively affect plant growth in similar manners by disrupting cellular redox homeostasis and metabolism. However, the regulatory networks coordinating transcriptional control and metabolic adaptation under both types of stress remain poorly understood. Here, we demonstrate that the R2R3-MYB transcription factors GmMYB60a and GmMYB60b serve as core negative regulators of drought and salt tolerance in soybean. Loss-of-function mutations in GmMYB60a/b clearly increased stress tolerance, whi

PtoeIF5A1: A Pleiotropic Regulator of Development, PCD, and Salt Tolerance in Populus tomentosa.

Eukaryotic translation initiation factor 5A (eIF5A) is a highly conserved protein family unique to eukaryotes, yet its functional characterization in woody plants remains limited. In this study, we identified four eIF5A genes (PtoeIF5A1-PtoeIF5A4) from the genome of Populus tomentosa, a fast-growing tree species indigenous to China, and characterized their expression patterns and functional roles through bioinformatics analysis, quantitative real-time PCR, stable overexpression in Arabidopsis thaliana, and transient expression in Nicotiana benthamiana leaves. Our results demonstrated that all

Mechanistic insights and breeding prospects of secondary metabolites in wheat salt stress tolerance.

Secondary metabolites play important roles in osmotic adjustment, ion homeostasis, and redox signaling in wheat under salinity stress. Together, these functions support plant acclimation to saline conditions. Integrative omics approaches can clarify the regulation of their biosynthetic pathways. Applying this knowledge in targeted breeding may accelerate the development of saltresilient wheat cultivars. Salt stress is a major environmental challenge that adversely affects wheat growth, developmental cascades, and grain yield and quality. As a major staple crop, it is imperative to improve whea

REM Gene Family in Sugar Beet and Functional BvREM12 in Salt Stress Tolerance.

Soil salinization acts as a major abiotic stress and severely hampers global agricultural productivity. Elucidating the molecular mechanisms underlying plant salt tolerance is of great significance for crop improvement. Remorin (REM) proteins are plant-specific membrane marker proteins that play a key role in stress signaling; however, their function in the salt tolerance response of sugar beet remains unclear. In this study, 14 BvREM gene family members were identified. Phylogenetic analysis revealed that this family can be divided into five subgroups and exhibits significant interspecies col

Melatonin enhances salt tolerance by promoting CcCAD10-mediated lignin biosynthesis in pigeon pea.

Melatonin plays a crucial role in enhancing plant resistance to salt stress by regulating biosynthesis of specialized metabolites. Phenylpropanoids, especially lignin, contribute to all aspects of plant responses toward biotic and abiotic stresses. However, the crosstalk between melatonin and lignin is largely unknown in pigeon pea under salt stress. In this study, the cinnamyl alcohol dehydrogenase CcCAD10 was identified to be involved in melatonin treatment and salt stress. The content of lignin was significantly increased in CcCAD10 over-expression (OE) lines, the enhanced antioxidant enzym

Distinct vacuolar responses to salinity stress in the hyphal cell of salt-tolerant and salt-sensitive strains of Rhizopogon roseolus.

The vacuole is a highly dynamic organelle that can change its morphology in response to environmental conditions. In this study, we investigated the vacuolar dynamics in the terminal and subterminal hyphal cells of ectomycorrhizal fungi Rhizopogon roseolus under saline stress using a microscopic approach with artificial seawater. Terminal cells responded to seawater exposure by increasing spherical vacuole formation. Artificial seawater induced progressive vacuolar fragmentation in subterminal cells of salt-sensitive strains. In contrast, the salt-tolerant strain exhibited a distinct adaptive

EsPIP5K4, a target of Es-miR408, positively regulates salt tolerance in Siberian wildrye.

Siberian wildrye (Elymus sibiricus L.) is a native grass of significant ecological and forage value for ecological restoration and livestock production in western China. However, the miRNA-mediated regulatory mechanisms underlying its salt tolerance remain largely unexplored. In this study, through integrative miRNA-mRNA profiling under salt stress, we identified EsPIP5K4 as a candidate target of Es-miR408. The inverse expression patterns of Es-miR408 (downregulated) and EsPIP5K4 (upregulated) under salt stress, together with dual-luciferase reporter assays, indicated that Es-miR408 negatively

A domestication-selected CIPK-MYB-HAK module improves salt tolerance in modern maize.

Domestication has profoundly reshaped the maize genome, yet its impact on abiotic stress resilience remains largely unresolved. Here we show that modern maize exhibits markedly greater salt tolerance than its wild progenitor, teosinte. Using a maize-teosinte (W22-T8759) recombinant inbred line population, we identify the transcriptional repressor MYB28 as a major genetic determinant underlying this divergence. MYB28 negatively regulates salt tolerance by impairing shoot Na+ exclusion. In modern maize (W22), however, salt stress activates CIPK20, which phosphorylates MYB28 to relieve its repres

Transcriptomics-guided identification of engineering targets for improving salt tolerance in Saccharomyces cerevisiae.

BACKGROUND: High-salinity conditions frequently impair the fermentation performance of Saccharomyces cerevisiae in industrial processes involving high-osmolarity substrates. Identifying genetic determinants that enhance salt tolerance is therefore essential for the development of robust yeast cell factories. In this study, a comparative transcriptomic analysis was performed to investigate the transcriptional responses of a salt-tolerant strain, E-158, and its parental strain, KF-7, under 1.25 M NaCl stress, with the aim of identifying potential targets for strain engineering. RESULTS: Compara
