Pelvic Ewing Sarcoma: The Great Mimicker.

Ewing sarcoma is the most common malignant bone tumor of the pelvis in children and young adults. Even with aggressive treatment, its survival rate is amongst the poorest. Classical presentation may not be the rule. It may simulate clinically, imagiologically and histopathologically other nonmalignant entities. Therefore, its suspicion should not be overlooked. We report two cases of pelvic Ewing sarcoma: the first mimicking eosinophilic granuloma, and the second mimicking osteomyelitis. In the latter, we also report an atypical finding of its natural history: an initial response to antibiotic

Ewing Sarcoma in the Right Ventricle.

Ewing sarcoma is the second most prevalent malignant primary bone tumor but constitutes only a small proportion of cardiac metastases. We present a case of asymptomatic Ewing sarcoma metastatic to the right ventricle. A 36-year-old man presented for evaluation and resection of a pedunculated right ventricular cardiac tumor. Three years before, he had been diagnosed with translocation-negative Ewing sarcoma, for which he had undergone chemotherapy and amputation of the left leg below the knee. We resected the right ventricular tumor. Analysis of the resected mass supported the diagnosis of meta

Primary cutaneous and subcutaneous Ewing sarcoma.

BACKGROUND: Primary cutaneous/subcutaneous Ewing sarcoma (scEWS) is extremely rare. We describe clinical features, treatment, and outcome of this Ewing localization. PROCEDURE: Retrospective study (1996-2012) on 56 patients. RESULTS: Most primary scEWS occurred in late adolescent/young adult females (F/M = 1.9; median age 21.5 years), with primary tumor in the extremity/trunk (48.5%/39%). Only 35/56 samples had Real-Time-Polymerase-Chain-Reaction/Fluorescent-In-Situ-Hybridization analysis, 32/35 had EWS-translocation. Most of them exhibited known favorable prognostic factors: localized disea

Anti-Epileptic Drug Targets Ewing Sarcoma.

Ewing Sarcoma (ES) is a rare form of bone cancer that most commonly affects children and adolescents. Chromosomal translocations are fundamental to the development of Ewing Sarcoma, linked to the changes in gene expression affecting transcription factors. Histone acetyl transferases (HATs) and histone deacetylases (HDACs) regulate transcription by modifying acetylation of both histones and transcription factors. Despite the use of multimodal therapeutic approaches current therapies are associated with significant short and long-term side effects. Hence, new therapeutic approaches are needed. I

[Rare localization of sarcoma in an adolescent: thoracic Ewing sarcoma].

BACKGROUND: Ewing Sarcoma is considered as primitive neuro ectodermic tumor. It's the most frequent osseous tumor in children and adolescent. It was localised frequently at long osseous and pelvis, however, it can be arising from the rib. AIM: this article aimed to show that Ewing sarcome could arise twely from thorax. CASE REPORT: We report the case of 15-year-old girl, admitted in our hospital because of left scapular pain with important weight loss. Chest X ray showed dense left latero tracheal opacity with mediastinal limits. Bronchofiberoscopy was performed and it showed no abnormalitie

Cytology of Undifferentiated Round-Cell Sarcomas of Bone and Soft Tissue: Ewing Sarcoma or Not Ewing Sarcoma, That Is the Question.

BACKGROUND: Undifferentiated round-cell sarcomas (URCSs) of soft tissue and bone are a group of clinically heterogeneous tumors. Diagnosis of these malignancies is based mainly on recurrent genetic alterations. The most common and the best known representative of this group is Ewing sarcoma (ES) which is characterized by gene fusions including EWSR1 or FUS and ETS transcription factors family. Other newly described entities are CIC-rearranged sarcoma, sarcoma with BCOR genetic alterations, and round-cell sarcoma with EWSR1-non-ETS fusions. All these novel tumors are known as Ewing-like sarcoma

Suicide gene therapy targeting ewing sarcoma via an ewing-specific GGAA promoter.

We recently demonstrated that GGAA promoters (GGAAprom) can be used to drive the expression of therapeutic genes (i.e., Cas9) in vitro and in vivo in a highly specific way in Ewing sarcoma cells. In this study, we take advantage of this strategy to express the suicide gene thymidine kinase from herpes simplex virus (HSV-TK), a suicide gene frequently used as an antitumoral therapy in clinical settings. The expression of HSV-TK in Ewing sarcoma cells under the control of the GGAA promoter resulted in cell death in the presence of ganciclovir both in vitro and in vivo. Interestingly, in vitro st

Ewing Sarcoma With Emphasis on Extra-skeletal Ewing Sarcoma: A Decade's Experience From a Single Centre in India.

INTRODUCTION: The diagnosis of Ewing sarcoma family of tumours (ESFT) is challenging, especially in adults and in extra-skeletal or visceral location. Several morphologic mimics with varied treatment options and prognosis confer diagnostic dilemmas. Application of ancillary diagnostic modalities in surgical pathology in clinical routine has enabled accurate diagnosis of ESFT in bone, soft tissues, and viscera. AIM: The study aims to assess the clinicopathological features including molecular test results of ESFT with emphasis on sex, age, and location, especially extra-skeletal soft tissue an

Ewing's sarcoma metastatic at diagnosis. Results and comparisons of two intergroup Ewing's sarcoma studies.

Two Pediatric Intergroup Ewing's Sarcoma studies of patients with metastatic disease (IESS-MD) have used multimodal therapy consisting of intensive combination chemotherapy and radiation therapy (XRT) to areas of gross disease detected at the time of diagnosis. In IESS-MD-I, conducted from 1975 to 1977, 53 eligible patients were entered and received the chemotherapeutic agents vincristine, Adriamycin (doxorubicin; Adria Laboratories, Columbus, OH), cyclophosphamide, and dactinomycin with concomitant XRT (VACA + XRT). In IESS-MD-II, conducted from 1980 to 1983, 69 eligible patients were entered

Genetically Engineered Mouse Model in Ewing Sarcoma.

Modeling Ewing sarcoma is challenging, since overexpression of EWSR1::FLI1 induces apoptosis and is not sufficient for tumor induction. It is therefore important to obtain the cell-of-origin of Ewing sarcoma that is tolerant of EWSR1::FLI1 expression. Here we describe the generation of the EWSR1::FLI1-expressing mouse model for Ewing sarcoma by selecting embryonic chondrogenic progenitor, eSZ cells that contain Ewing sarcoma precursors.

3D Ewing Sarcoma Modeling for Metabolic Studies.

Three-dimensional (3D) in vitro models provide a physiologically relevant platform that more accurately recapitulates tissue architecture, cell-cell and cell-extracellular matrix interactions, and the physical microenvironment than traditional two-dimensional (2D) cultures. In particular, these models reproduce metabolic gradients, including oxygen and nutrient availability, enabling more precise investigations of cellular metabolism. This chapter describes two methods to grow Ewing sarcoma cell lines as 3D spheroids using physiological media, as well as the collection of spheroids in bulk or

Adamantinoma-like Ewing sarcoma of the bladder.

Adamantinoma-like Ewing sarcoma (ALES) is an exceedingly rare neoplasm, typically arising in the head and neck. We present a case of bladder tumour initially diagnosed as urothelial carcinoma on transurethral resection, which after neoadjuvant dose-dense methotrexate, vinblastine, doxorubicin and cisplatin and subsequent radical cystoprostatectomy was reclassified as ALES with no residual urothelial carcinoma.

Pulmonary Ewing Sarcoma Presenting As Chronic Cough.

Ewing sarcoma is an aggressive malignant small round cell tumour primarily affecting bone and soft tissue in young individuals. Pulmonary involvement is commonly seen in metastatic disease, while late recurrence after a prolonged disease-free interval is uncommon and may pose a diagnostic challenge. We report a 26-year-old male with previously treated non-metastatic Ewing sarcoma of the left fibula, who presented four years later with persistent chronic cough. Imaging revealed a large heterogeneously enhancing mass in the left lower lobe with bronchial cutoff, raising suspicion for primary lun

Ewing sarcoma - current diagnostic and therapeutic approaches.

BACKGROUND: Ewing sarcoma belongs to the family of undifferentiated small round cell sarcomas of bone and soft tissue. It is characterized by a gene fusion involving EWSR1 and an ETS-family transcription factor gene. In 85-96% of cases, a specific chromosomal translocation results in the EWSR1-FLI1 fusion gene, whose product functions as an oncogene essential for tumorigenesis. Ewing sarcoma is most common in adolescents and young adults. It primarily affects the diaphyses of long bones, the pelvis, and the axial skeleton, although extraosseous involvement is not uncommon. This is a highly mal

Extraosseous Ewing Sarcoma With Upper Gastrointestinal Bleeding.

Ewing sarcoma/primitive neuroectodermal tumor (ES/PNET) is a type of malignancy that usually appears as a tumor in the bone. However, in a few patients with ES/PNET, it can occur outside of the bone. Although extraosseous ES/PNET can appear in various parts of the body, involvement of small bowel is rare. If it does, it can present with vague abdominal pain and gastrointestinal bleeding. This case report presents a 28-year-old gentleman with extraosseous ES/PNET in the duodenum who experienced gastrointestinal bleeding as a presenting symptom.

Ewing Sarcoma: Rare Metastasis to the Pancreas.

Ewing sarcoma is a highly aggressive malignancy of bone or soft tissue, which may present with metastasis for 20%-25% of patients. The most common sites of metastatic lesions are the bone, bone marrow, and lungs. When metastatic lesions present within rare visceral sites, such as the pancreas, it may lead to an incorrect diagnosis of small cell neuroendocrine carcinoma. We report a case of a 37-year-old man with metastatic Ewing sarcoma involving the pancreas, confirmed by imaging, sequencing, fluorescence in situ hybridization, and histology, which was initially mistaken for small cell neuroe

Adamantinoma-like Ewing sarcoma of the thyroid.

Adamantinoma-like Ewing sarcoma (ALES) is a rare variant of Ewing sarcoma family of tumors (ESFTs). ALES is characterized by translocations of the EWSR1 (Ewing sarcoma breakpoint region 1) gene on chromosome 22, morphologic features of small round blue cell tumors with focal squamous differentiation, and a unique immunoprofile. Herein, we report a 21-year-old patient who presented with severe, acute onset back pain. Imaging revealed a large, exophytic, heterogeneously enhancing mass in the left thyroid and numerous lytic bone lesions. Fine-needle aspiration of the thyroid, revealed a cellular

Extraskeletal Ewing sarcoma of the buccal space.

Extraskeletal Ewing sarcoma (EES) is a rare soft tissue tumor, and EES of the head and neck is particularly rare. Radiographic imaging of these lesions is crucial given their anatomical complexity and infrequent incidence. Conventional EES imaging features include a large, hyperintense, heterogeneously enhancing lesion, with frequent invasion of local structures. In this case report, a 19-year old male presented with left facial swelling and pain. He underwent sclerotherapy and bleomycin treatment for a presumed lymphatic malformation. Initial imaging demonstrated a rim-enhancing lesion within

Spinal intradural metastasis from scapular Ewing sarcoma.

BACKGROUND: Ewing sarcoma is a primary bone neoplasm, which is a high grade aggressive small round blue cell tumour, and is currently recognized as a part of the Ewing family of tumours. It is the most lethal bone tumor, and is a rare malignant bone tumor accounting for 10% of all primary bone tumors, and 6% of malignant bone tumors. It has an average annual incidence of 3 per 1 million, found almost exclusively in Caucasians. It commonly occurs in long bones and pelvis but rarely involves the scapula. 85% of cases have metastasis within 2 years of diagnosis, rarely involving the meninges. CA
