The effect of starvation and re-feeding on vasotocinergic and isotocinergic pathways in immature gilthead sea bream (Sparus aurata).

This study describes the responses of the vasotocinergic and isotocinergic systems to food deprivation and re-feeding processes in immature gilthead sea bream (Sparus aurata). The animals were subjected to the following experimental treatments: (1) normal feeding (control), (2) food deprivation for 21 days; and (3) re-feeding for 7 days, beginning 14 days after starvation. The animals were sampled at 0, 7, 14 and 21 days from the beginning of the trial. The pituitary and plasma arginine vasotocin (AVT) and isotocin (IT) levels and the hypothalamic pro-vasotocin and pro-isotocin mRNA expression…

Effects of Enteromyxum spp. (Myxozoa) infection in the regulation of intestinal E-cadherin: Turbot against gilthead sea bream.

Enteromyxoses are relevant diseases for turbot and gilthead sea bream aquaculture. The myxozoan parasites invade the intestinal mucosa, causing a cachectic syndrome associated with intestinal barrier alteration; nonetheless, their pathological impact is different. Turbot infected by Enteromyxum scophthalmi develop more severe intestinal lesions, reaching mortality rates of 100%, whereas in E. leei-infected gilthead sea bream, the disease progresses slowly, and mortality rates are lower. The mechanisms underlying the different pathogenesis are still unclear. We studied the distribution and expr…

Isolation of mast cells from the peritoneal exudate of the teleost fish gilthead sea bream (Sparus aurata L.).

Inflammation is the first response of animals to infection or tissue damage. Sparus aurata (Perciformes) was the first fish species shown to possess histamine-containing mast cells at mucosal tissues. We report a separation protocol for obtaining highly enriched (over 95% purity) preparations of fish mast cells in high numbers (5-20 million mast cells per fish). The peritoneal exudate of S. aurata is composed of lymphocytes, acidophilic granulocytes, macrophages and mast cells. We separated the lymphocyte fraction through discontinuous density gradient centrifugation. The remaining cells were

Modulation of leukocytic populations of gilthead sea bream (Sparus aurata) by the intestinal parasite Enteromyxum leei (Myxozoa: Myxosporea).

SUMMARY The cellular mucosal and systemic effectors of gilthead sea bream (GSB) (Sparus aurata) involved in the acute immune response to the intestinal parasite Enteromyxum leei were studied in fish experimentally infected by the anal route. In the intestinal inflammatory infiltrates and in lymphohaematopoietic organs (head kidney and spleen) of parasitized fish, the number of plasma cells, B cells (IgM immunoreactive) and mast cells (histamine immunoreactive) were significantly higher, whereas the number of acidophilic granulocytes (G7 immunoreactive) decreased, compared with non-parasitized …

Susceptibility of Sea Bream (Sparus aurata) to AIP56, an AB-Type Toxin Secreted by Photobacterium damselae subsp. piscicida.

Photobacterium damselae subsp. piscicida (Phdp) is a Gram-negative bacterium that infects a large number of marine fish species in Europe, Asia, and America, both in aquacultures and in the natural environment. Among the affected hosts are economically important cultured fish, such as sea bream (Sparus aurata), sea bass (Dicentrarchus labrax), yellowtail (Seriola quinqueradiata), and cobia (Rachycentron canadum). The best characterized virulence factor of Phdp is the Apoptosis-Inducing Protein of 56 kDa (AIP56), a secreted AB-type toxin that has been shown to induce apoptosis of sea bass phago…

Unraveling vasotocinergic, isotocinergic and stress pathways after food deprivation and high stocking density in the gilthead sea bream.

The influence of chronic stress, induced by food deprivation (FD) and/or high stocking density (HSD), was assessed on stress, vasotocinergic and isotocinergic pathways of the gilthead sea bream (Sparus aurata). Fish were randomly assigned to one of the following treatments: (1) fed at low stocking density (LSD-F; 5kg·m-3); (2) fed at high stocking density (HSD-F, 40kg·m-3); (3) food-deprived at LSD (LSD-FD); and (4) food-deprived at HSD (HSD-FD). After 21days, samples from plasma, liver, hypothalamus, pituitary and head-kidney were collected. Both stressors (FD and HSD) induced a chronic stres…

Characterization of a sea bream (Sparus aurata) thyroid hormone receptor-beta clone expressed during embryonic and larval development.

A clone encoding thyroid hormone receptor-beta (TR-beta) was isolated from a sea bream (Sparus aurata) ovary cDNA library. Sea bream (sb)TR-beta is closely related to its counterparts from other vertebrates and, like them, preferentially binds T3 rather than T4. However, the putative sbTR-beta protein contains a nine-amino-acid insert that is also present in the corresponding proteins from flounder and salmon but absent in TR-betas from zebrafish and terrestrial vertebrates. Semiquantitative RT-PCR analysis showed that sbTR-beta transcripts begin to accumulate during gastrulation and increase …

Development of a High-Hydrostatic-Pressure-Treated Recombinant Vaccine Targeting the Major Capsid Protein of Red Sea Bream Iridovirus.

Red sea bream (Pagrus major) aquaculture represents one of the most economically important marine aquaculture industries in Japan and East Asia. However, viral diseases, particularly those caused by red sea bream iridovirus (RSIV), pose a serious threat to aquaculture production in this region. In this study, we applied high-hydrostatic-pressure (HHP) refolding technology to develop a recombinant vaccine targeting the RSIV major capsid protein (MCP). The recombinant MCP (RSIV-rMCP) expressed in Escherichia coli was insoluble; however, HHP treatment under alkaline (pH 10) conditions in the pres…

Insulin during the early postembryonic development of the gilt-head sea bream, Sparus aurata: ultrastructural, immunohistochemical, and biochemical studies.

Insulin was sought during the early postembryonic development of gilt-head sea bream, Sparus aurata, using ultrastructural, immunohistochemical and biochemical techniques. The endocrine pancreas appeared at hatching (Day 0) as a single cluster of morphologically similar cells. Secretory granules formed from Day 1 onwards but the cells could only be identified as insulin-producing B cells at the end of the endotrophic period (Day 3-Day 4). Insulin-immunoreactive cells were detected in the pancreatic primordium from hatching onwards and their number increased after the end of the endo-exotrophic…

Insights into red sea bream iridovirus pathogenesis: Unveiling host-pathogen interactions using membrane yeast two-hybrid and molecular dynamics.

Red sea bream iridovirus (RSIV), a member of the genus Megalocytivirus, poses a substantial threat to global aquaculture through its infection of marine fish species. The major capsid protein (MCP) represents the principal immunogenic component of RSIV, yet the mechanisms governing its pathogenicity is unclear. Here, we employed a comprehensive approach combining protein interaction screening with artificial intelligence-based structural prediction to elucidate the interface between RSIV and its host. In brief, by constructing a yeast cDNA library of membrane protein in red snapper and using a…

Data on CXC chemokine ligand 10(CXCL10) expression and activation in red sea bream during bacterial and viral infection.

CXCL10 plays an important role in angiogenesis and inhibits the differentiation of endothelial cells into capillaries. It also plays an important role in the generation and transmission of effector T cell responses and the recruitment of T cells to inflammatory sites. In this article, we constructed cDNAs to identify and analyse the CXCL10 domain, and performed multiple alignments and a phylogenetic analysis to determine homology with other animals. Real-time PCR was performed to confirm construction and expression after bacterial and viral infection.

Effects of Enteromyxum leei (Myxozoa) infection on gilthead sea bream (Sparus aurata) (Teleostei) intestinal mucus: glycoprotein profile and bacterial adhesion.

The intestinal myxosporean parasite Enteromyxum leei causes severe desquamative enteritis in gilthead sea bream (Sparus aurata) (Teleostei) that impairs nutrient absorption causing anorexia and cachexia. In fish, as in terrestrial vertebrates, intestinal goblet cells are responsible for the adherent mucus secretion overlying epithelial cells, which constitutes a first line of innate immune defense against offending microorganisms but serves also as substrate and nutrient source for the commensal microflora. The secreted intestinal mucus of parasitized (n = 6) and unexposed (n = 8) gilthead sea…

Regulation of troponin T expression during muscle development in sea bream Sparus auratus Linnaeus: the potential role of thyroid hormones.

In the sea bream Sparus auratus three stage-specific fast troponin T (fTnT) isoforms have been cloned and correspond to embryonic-, larval- and adult-specific isoforms. Characterisation, using database searches, of the putative genomic organisation of Fugu rubripes and Tetraodon nigroviridis fTnT indicates that alternative exon splicing in the 5 region of the gene generates the different isoforms. Moreover, comparison of teleost fTnTs suggests that alternative splicing of fTnT appears to be common in teleosts. A different temporal expression pattern for each fTnT splice varotnt is found during…

Perinotochordal connective sheet of gilthead sea bream larvae (Sparus aurata, L.) affected by axial malformations: an histochemical and immunocytochemical study.

BACKGROUND: Spinal malformations in adult teleosts occur under natural conditions and, more frequently, in culture exploitations. Skeletal deformities are linked with dysfunctions in collagen metabolism. We studied axial deviations appearing in early larval stages of cultured sea bream (Sparus aurata, L.). METHODS: To evaluate connective tissue components of normal and lordotic fish we used histochemistry (alcian blue, picrosirius-polarization, clorhydric orcein, fuchsin resorcin), immunohistochemistry (anti-collagen I, II, III, and IV), and specific enzymatic digestions. The results were eva…

Comparative parasitology and host pathophysiology: integrated assessment of ectoparasite dynamics, seasonal prevalence, and physiological impacts in co-cultured European sea bass (Dicentrarchus labrax) and gilthead sea bream (Sparus aurata).

Ectoparasitic infestations pose significant threats to Mediterranean aquaculture, particularly for economically important species, European sea bass (Dicentrarchus labrax) and gilthead sea bream (Sparus aurata). Intensified aquaculture practices exacerbate these issues, impairing fish immunity, reducing growth rates, and incurring substantial economic losses. This study examined the prevalence, seasonal dynamics, organ tropism, and pathophysiological impacts of key ectoparasites in 1200 fingerlings (600 per species) collected seasonally (winter: December-February; summer: June-August) from pri…

AVT and IT regulate ion transport across the opercular epithelium of killifish (Fundulus heteroclitus) and gilthead sea bream (Sparus aurata).

The regulatory role of arginine vasotocin (AVT) and isotocin (IT) in Cl(-) secretion was investigated with the short circuit current (Isc) technique in opercular epithelia of killifish (Fundulus heteroclitus) and gilthead sea bream (Sparus aurata). Sea bream operculum showed ~4-fold lower number of Na/K-ATPase immunoreactive cells and ~12-fold lower secretory current than the killifish. In sea bream opercular membranes, the basolateral addition of AVT (10(-6) M) significantly stimulated Cl(-) secretion, while IT (10(-6) M) was without effect. In killifish, IT produced an immediate dose-depende…
