Efficient asymmetric oxidation of sulfides and kinetic resolution of sulfoxides catalyzed by a vanadium-salan system.

The asymmetric oxidation of sulfides to chiral sulfoxides with hydrogen peroxide in good yield and high enantioselectivity has been catalyzed very effectively by chiral vanadium-salan [N,N'-alkyl bis(salicylamine)] complex. The salan ligand shows results superior in terms of reactivity and enantioselectivity to those of salen [N,N'-alkylene bis(salicylideneimine)] analogue, and provides the sulfoxide with opposite configuration. The high enantioselectivity of this reaction is the direct result of the asymmetric oxidation. The efficient kinetic resolution of racemic sulfoxides catalyzed by the …

Methionine sulfoxide reductases and methionine sulfoxide in the subterranean mole rat (Spalax): characterization of expression under various oxygen conditions.

The blind subterranean mole rat (Spalax ehrenbergi) exhibits a relatively long life span, which is attributed to an efficient antioxidant defense affording protection against accumulation of oxidative modifications of proteins. Methionine residues can be oxidized to methionine sulfoxide (MetO) and then enzymatically reduced by the methionine sulfoxide reductase (Msr) system. In the current study we have isolated the cDNA sequences of the Spalax Msr genes as well as 23 additional selenoproteins and monitored the activities of Msr enzymes in liver and brain of rat (Rattus norvegicus), Spalax gal

Copper-Catalyzed Radical Alkyl- and Aminosulfinylation of Styrenes Enabled by β-Sulfonyl Sulfoxides as Sulfenate Anion Precursors.

Sulfenate anions are powerful but labile intermediates for sulfoxide synthesis, and their generation under harsh conditions has limited their use in radical chemistry. Here, bench-stable β-sulfonyl sulfoxides are introduced as readily accessible sulfenate precursors that release sulfenates under mild conditions. This platform enables copper-catalyzed three-component 1,2-alkylsulfinylation and 1,2-aminosulfinylation of styrenes with a broad scope, late-stage applicability, and mechanistic evidence for radical-anion crossover.

Intertwined Processes in the Pummerer-Type Rearrangement Affording α-Trifluoromethoxylated Thioethers and the Corresponding Highly Enantioenriched Sulfoxides.

A method to access racemic or highly stereoenriched α-trifluoromethoxylated sulfoxides through the Pummerer rearrangement of aryl methyl sulfoxides followed by the oxidation of the resulting trifluoromethoxylated sulfides is described. Among the three reagents selected as both rearrangement activators and F3CO- anion sources, 2,4-dinitro-1-(trifluoromethoxy)benzene (DNTFB) was found to be the most effective. In each case, the OCF3-source was shown to play diverse key roles in the rearrangement. Enantioenriched trifluoromethoxy-sulfoxides (up to 95% enantiomeric excess) were obtained using an o…

Dimethyl Sulfoxide Induces Hemolysis and Pulmonary Hypertension.

Vascular and lung injury are well established complications associated with hemolytic disorders, and hemolysis associated pulmonary hypertension (PH) has emerged as the most serious complication of sickle cell disease. The causal relationship between intravascular hemolysis and the development of PH is still under investigation. Previously we have shown that repetitive administration of hemolyzed autologous blood causes PH in rats. Dimethyl sulfoxide (DMSO), a widely used solvent and anti-inflammatory agent, induces hemolysis in vivo. We hypothesized that repetitive administration of DMSO woul

Development of chiral sulfoxide ligands for asymmetric catalysis.

Nitrogen-, phosphorus-, and oxygen-based ligands with chiral backbones have been the historic workhorses of asymmetric transition-metal-catalyzed reactions. On the contrary, sulfoxides containing chirality at the sulfur atom have mainly been used as chiral auxiliaries for diastereoselective reactions. Despite several distinct advantages over traditional ligand scaffolds, such as the proximity of the chiral information to the metal center and the ability to switch between S and O coordination, these compounds have only recently emerged as a versatile class of chiral ligands. In this Review, we …

Molecular complexes of dimethyl sulfoxide with tri- and dichloromethane.

Crystals of molecular complexes of dimethyl sulfoxide with trichloromethane (chloroform), (CH(3))(2)SO·2CHCl(3), (I), and dichloromethane, (CH(3))(2)SO·CH(2)Cl(2), (II), have been grown in situ. In both compounds, the components are linked together by (Cl)C-H···O interactions. The dimethyl sulfoxide molecules in (I) are bound into chains by C-H···O interactions. In (II), pairs of the components form centrosymmetric rings, linked into a three-dimensional network by C-H···O contacts and dipole-dipole interactions between dimethyl sulfoxide molecules.

Solvent effects on isomerization in a ruthenium sulfoxide complex.

We report the structure, electrochemistry, and isomerization kinetics for [Ru(bpy)(biq)(OSO)](PF(6)), where bpy is 2,2'-bipyridine, biq is 2,2'-biquinoline, and OSO is 2-methylsulfinylbenzoate. UV-visible and infrared data are suggestive of intramolecular S-->O and O-->S isomerization of the sulfoxide. Cyclic voltammetry reveals evidence for isomerization triggered by oxidation and reduction. Of particular note is the variation of the S-->O isomerization rate constant in different solvents. The rates were found to be 3.2 (+/-0.4) s(-1) in propylene carbonate, 0.80 (+/-0.03) s(-1) in acetonitri

Pharmacology of dimethyl sulfoxide in cardiac and CNS damage.

The pharmacological effects of dimethyl sulfoxide (DMSO) administration include some desirable properties that may be useful in the treatment of medical disorders resulting in tissue injury and compromised organ systems. These properties include the reported effects of DMSO on impaired blood flow, suppression of cytotoxicity from excess glutamate release that may result in lethal NMDA-AMPA activation, restriction of cytotoxic Na(+) and Ca(2+) entry into damaged cells, blocking tissue factor (TF) from contributing to thrombosis, reduction of intracranial pressure, tissue edema, and inflammatory

Degradation and configurational changes of thioridazine 2-sulfoxide.

Thioridazine (THD) is a phenothiazine neuroleptic drug used for the treatment of psychiatric disorders. After oral administration THD is extensively biotransformed to thioridazine 2-sulfone (THD 2-SO(2)), thioridazine 5-sulfoxide (THD 5-SO) and thioridazine 2-sulfoxide (THD 2-SO). THD 2-SO and THD 5-SO have two chiral centres and therefore exist as two diastereoisomeric pairs. The degradation and epimerization of THD 2-SO in human plasma, buffer and methanolic solutions were studied using an enantioselective HPLC method. The samples were prepared by liquid-liquid extraction with diethyl ether

Dimethyl sulfoxide depresses skeletal muscle contractility.

Dimethyl sulfoxide (DMSO) is commonly used in studies of skeletal muscle as a selective antioxidant (DMSO preferentially scavenges hydroxyl radicals) or as a solvent for drugs. The present experiments tested DMSO for direct effects on diaphragm contractile properties. Fiber bundles were removed from anesthetized rats, mounted in vitro at optimal length (37 degrees C), curarized, and stimulated directly. Protocol 1 tested for contractile depression and dose dependence by comparing bundles treated with DMSO (0.6-640 mM) with time- and stimulus-matched controls. Protocol 2 tested reversibility of

Dimethyl sulfoxide inhibits NLRP3 inflammasome activation.

Dimethyl sulfoxide (DMSO) is an amphipathic molecule that is commonly/widely used as a solvent for biological compounds. In addition, DMSO has been studied as a medication for the treatment of inflammation, cystitis, and arthritis. Based on the anti-inflammatory characteristics of DMSO, we elucidated the effects of DMSO on activation of inflammasomes, which are cytoplasmic multi-protein complexes that mediate the maturation of interleukin (IL)-1β by activating caspase-1 (Casp1). In the present study, we prove that DMSO attenuated IL-1β maturation, Casp1 activity, and ASC pyroptosome formation

Characterization of the methionine sulfoxide reductases of Schistosoma mansoni.

Schistosomiasis, also known as Bilharzia, is an infectious disease caused by several species of Schistosoma. Twenty million individuals suffer severe symptoms and 200,000 people die annually from the disease. The host responds to the presence of S. mansoni by producing reactive oxygen species that cause oxidative stress. We hypothesized that schistosomes produce antioxidants in response to oxidative stress. A known antioxidant protein is methionine sulfoxide reductase (Msr). Methionine residues can be oxidized to methionine sulfoxide in the presence of oxidizing agents, and the process is read
