[Effects of mercury compounds on growth, cell composition and glucose uptake in Tetrahymena pyriformis].

The effects of the mercuric compounds methylmercuric chloride (MMC) and mercuric chloride (MC) on growth, cell composition, and glucose uptake were studied in the ciliate protozoan Tetrahymena pyriformis GL. MMC was the most effective growth inhibitor of Tetrahymena among mercuric compounds and sulfhydryl inhibitors; it completely inhibited growth of Tetrahymena at concentrations of 1/30-1/3000 of the others. The growth inhibition of Tetrahymena was inversely proportional to concentrations of chemicals in medium. Particularly in the case of MMC and MC the relationship between the concentration…

Analysis of the genes and mechanisms responsible for the cytotoxicity of the Legionella Ofk308 strain in the Tetrahymena host.

BACKGROUND: Legionella pneumophila, an intracellular pathogen responsible for the pneumonia-like Legionnaires' disease in humans, inhabits aquatic environments, including man-made water systems such as water fountains, foot spas, and tap water, and exists as part of biofilms or as a protozoan parasite. As a bacterivore, Tetrahymena thermophila provides a favorable environment for Legionella to establish a replicative niche (Legionella-containing vacuole; LCV) under environmental stress. Conversely, the L. pneumophila Ofk308 strain, isolated from an Ashiyu foot spa, has been found to be cytotox…

Organic matter excreted by the protozoan Tetrahymena thermophila and its effects on the bioaccumulation of nanoparticles.

Although organic matter (exudate) excreted by aquatic organisms is an important component of dissolved organic matter (DOM) in the natural environment, its potential effects on the bioaccumulation of nanoparticles (NPs) remain unclear. In the present study, we examined the effects of the exudates from the protozoan Tetrahymena thermophila on the bioaccumulation (including uptake and cell surface adsorption) of iron oxide (Fe2O3, polyacrylate coated) and silica (SiO2) NPs in T. thermophila. The exudates were mostly (93.6 %, in carbon) composed of < 1-kDa molecules (e.g., lipids). When the exuda…

Effects of six pyrimidine analogs on the growth of Tetrahymena thermophila and their implications in pyrimidine metabolism.

Tetrahymena are ciliated protists that have been used to study the effects of toxic chemicals, including anticancer drugs. In this study, we tested the inhibitory effects of six pyrimidine analogs (5-fluorouracil, floxuridine, 5'-deoxy-5-fluorouridine, 5-fluorouridine, gemcitabine, and cytarabine) on wild-type CU428 and conditional mutant NP1 Tetrahymena thermophila at room temperature and the restrictive temperature (37°C) where NP1 does not form the oral apparatus. We found that phagocytosis was not required for pyrimidine analog entry and that all tested pyrimidine analogs inhibited growth …

Anandamide amidohydrolase activity, released in the medium by Tetrahymena pyriformis. Identification and partial characterization.

Anandamide, an endogenous cannabinoid receptor ligand, was rapidly metabolized by Tetrahymena pyriformis in vivo. Metabolic products were mainly phospholipids as well as neutral lipids, including small amounts of free arachidonic acid. Anandamide amidohydrolase activity was detected in the culture medium by the release of [3H]arachidonic acid from [3H]anandamide, in a time- and concentration-dependent manner. Kinetic experiments demonstrated that the released enzyme had an apparent K(m) of 3.7 microM and V(max) 278 pmol/min/mg protein. Amidohydrolase activity was maximal at pH 9-10, was abolis…

Purification and characterization of a novel dimeric 20 alpha-hydroxysteroid dehydrogenase from Tetrahymena pyriformis.

Tetrahymena pyriformis was found to exhibit high NADPH-dependent 20-oxosteroid reductase activity that converted 17 alpha-hydroxyprogesterone into 17 alpha,20 alpha-dihydroxypregn-4-en-3-one. The enzyme was purified 400-fold from the cytosolic fraction. The purified enzyme with a specific activity of 6.4 mumol/min per mg of protein had an isoelectric point of 4.9 and M(r) of 68,000, and was composed of two subunits of equal size. The N-terminal sequence was determined to be LAKTVPLNDGTNFPIFGG. The enzyme reduced pregnanes and pregnanes possessing a 17 alpha-hydroxy group to a greater extent th…

Recombinant Haemagglutinin Derived From the Ciliated Protozoan Tetrahymena thermophila Is Protective Against Influenza Infection.

Current influenza vaccines manufactured using eggs have considerable limitations, both in terms of scale up production and the potential impact passaging through eggs can have on the antigenicity of the vaccine virus strains. Alternative methods of manufacture are required, particularly in the context of an emerging pandemic strain. Here we explore the production of recombinant influenza haemagglutinin using the ciliated protozoan Tetrahymena thermophila. For the first time we were able to produce haemagglutinin from both seasonal influenza A and B strains. This ciliate derived material was im…

Preparation and biological effect of nucleotide-capped CdSe/ZnS quantum dots on Tetrahymena thermophila.

In this paper, we described the preparation and characterization of different types of modified CdSe/ZnS quantum dots (QDs) and explored the biological effects of QDs with different surface modifications on the whole growth of unicellular protozoan Tetrahymena thermophila BF(5) using a thermal activity monitor air isothermal microcalorimeter. Our results demonstrated that adenosine 5'-monophosphate (AMP) showed stronger interaction with QDs than other types of nucleotide. AMP-QDs could stimulate the growth of T. thermophila while mercaptoacetic acid-capped CdSe/ZnS quantum dots inhibited it. I…

Machine learning-based models to predict modes of toxic action of phenols to Tetrahymena pyriformis.

The phenols are structurally heterogeneous pollutants and they present a variety of modes of toxic action (MOA), including polar narcotics, weak acid respiratory uncouplers, pro-electrophiles, and soft electrophiles. Because it is often difficult to determine correctly the mechanism of action of a compound, quantitative structure-activity relationship (QSAR) methods, which have proved their interest in toxicity prediction, can be used. In this work, several QSAR models for the prediction of MOA of 221 phenols to the ciliated protozoan Tetrahymena pyriformis, using Chemistry Development Kit des…

Distinct toxicological characteristics and mechanisms of Hg2+ and MeHg in Tetrahymena under low concentration exposure.

Inorganic divalent mercury complexes (Hg2+) and monomethylmercury complexes (MeHg) are the main mercury species in aquatic systems and their toxicity to aquatic organisms is of great concern. Tetrahymena is a type of unicellular eukaryotic protozoa located at the bottom of food chain that plays a fundamental role in the biomagnification of mercury. In this work, the dynamic accumulation properties, toxicological characteristics and mechanisms of Hg2+ and MeHg in five Tetrahymena species were evaluated in detail. The results showed that both Hg2+ and MeHg were ingested and exhibited inhibitory …

Complete sequence of the mitochondrial genome of Tetrahymena pyriformis and comparison with Paramecium aurelia mitochondrial DNA.

We report the complete nucleotide sequence of the Tetrahymena pyriformis mitochondrial genome and a comparison of its gene content and organization with that of Paramecium aurelia mtDNA. T. pyriformis mtDNA is a linear molecule of 47,172 bp (78.7 % A+T) excluding telomeric sequences (identical tandem repeats of 31 bp at each end of the genome). In addition to genes encoding the previously described bipartite small and large subunit rRNAs, the T. pyriformis mitochondrial genome contains 21 protein-coding genes that are clearly homologous to genes of defined function in other mtDNAs, including o…

Tetrahymena JanusA gene encodes a polo kinase whose loss triggers a dorsal/ventral intracellular homeosis.

Homeotic mutations result in developmental abnormalities in which one body part is replaced by another. In flowering plants, ABC mutations r … we identify a gene whose loss of function transforms the dorsal surface of the unicellular ciliate, Tetrahymena thermophila, normally devoid of cortical organelles, into a mirror-image of the ventral surface, complete with supernumerary contractile vacuole pores (CVPs) and a secondary oral apparatus (OA) exhibiting reversed local left-right asymmetry.3 Throug …

Development of a Tetrahymena thermophila-Based Vaccine Expressing Miamiensis avidus Ciliary Proteins to Combat Scuticociliatosis.

Miamiensis avidus is a parasitic pathogen responsible for scuticociliatosis, a lethal infection affecting marine fish worldwide, including Japanese flounder. Immunisation with formalin-killed M. avidus has shown promise in inducing protective immunity, positioning it as a potential vaccine candidate against scuticociliatosis. However, challenges such as the high cost of producing sufficient cells and inconsistent quality due to the lack of cryopreservation methods hinder its development. In this study, we expressed M. avidus ciliary proteins in Tetrahymena, a culturable ciliate, and used these…

Comparative transcriptome analysis uncovers roles of hydrogen sulfide for alleviating cadmium toxicity in Tetrahymena thermophila.

BACKGROUND: Cadmium (Cd) is a nonessential heavy metal with potentially deleterious effects on different organisms. The organisms have evolved sophisticated defense system to alleviate heavy metal toxicity. Hydrogen sulfide (H2S) effectively alleviates heavy metal toxicity in plants and reduces oxidative stress in mammals. However, the function of H2S for alleviating heavy metal toxicity in aquatic organisms remains less clear. Tetrahymena thermophila is an important model organism to evaluate toxic contaminants in an aquatic environment. In this study, the molecular roles of exogenously H2S a…

Screening Snake Venoms for Toxicity to Tetrahymena Pyriformis Revealed Anti-Protozoan Activity of Cobra Cytotoxins.

Snake venoms possess lethal activities against different organisms, ranging from bacteria to higher vertebrates. Several venoms were shown to be active against protozoa, however, data about the anti-protozoan activity of cobra and viper venoms are very scarce. We tested the effects of venoms from several snake species on the ciliate Tetrahymena pyriformis. The venoms tested induced T. pyriformis immobilization, followed by death, the most pronounced effect being observed for cobra Naja sumatrana venom. The active polypeptides were isolated from this venom by a combination of gel-filtration, io…

Glutathione Relevance in the Response of Ciliate Tetrahymena thermophila Against Metals/Metalloids and Other Environmental Stressors.

The tripeptide glutathione (GSH) is the primary and most abundant non-protein thiol compound present in all aerobic organisms. The main function of this molecule is cellular antioxidant defense. For the first time in ciliates, using the eukaryotic microorganism Tetrahymena thermophila as a model organism, several parameters (cell mortality, molecules containing -SH groups, and total glutathione) have been evaluated under the toxic effects of various chemical stressors (including several metals/metalloids), either in combination with or without the inhibitory effects of BSO (a GSH biosynthesis …

Estimation of the Toxicity of Different Substituted Aromatic Compounds to the Aquatic Ciliate Tetrahymena pyriformis by QSAR Approach.

Nowadays, quantitative structure⁻activity relationship (QSAR) methods have been widely performed to predict the toxicity of compounds to organisms due to their simplicity, ease of implementation, and low hazards. In this study, to estimate the toxicities of substituted aromatic compounds to Tetrahymena pyriformis, the QSAR models were established by the multiple linear regression (MLR) and radial basis function neural network (RBFNN). Unlike other QSAR studies, according to the difference of functional groups (−NO₂, −X), the whole dataset was divided into three groups and further modeled separ…

The H1 phosphorylation state regulates expression of CDC2 and other genes in response to starvation in Tetrahymena thermophila.

In Tetrahymena thermophila, highly phosphorylated histone H1 of growing cells becomes partially dephosphorylated when cells are starved in preparation for conjugation. To determine the effects of H1 phosphorylation on gene expression, PCR-based subtractive hybridization was used to clone cDNAs that were differentially expressed during starvation in two otherwise-isogenic strains differing only in their H1s. H1 in A5 mutant cells lacked phosphorylation, and H1 in E5 cells mimicked constitutive H1 phosphorylation. Sequences enriched in A5 cells included genes encoding proteases. Sequences enrich…
