2-Thiouracil (C4H4N2OS), an antithyroid drug (ATD) used in the treatment of Graves' disease and thyroid storm, requires an effective drug delivery vehicle to successfully reach the thyroid gland. Using first-principles calculations, the adsorption of the 2-thiouracil molecule on pristine, Si-doped, and Al-doped BC3 monolayers was studied based on density functional theory (DFT). Two established antithyroid drugs, 6-propyl-2-thiouracil and 6-methyl-2-thiouracil, as well as S-methylthiouracil, are shown to be competitive inhibitors of nitric oxide synthase (NOS) (K(I) values ranging from 14 to 60 microM), with moderate selectivity for the neuronal isoform. Other thioureylene and thioamide-containing heterocyclic systems proved virtually ineffective as NOS inhibitors. The effect of hypothyroidism on adrenals and gonads in adult female rats was investigated throughout the estrous cycle. Hypothyroidism was induced by administration of 4-Methyl-2-Thiouracil (Thiouracil) in the drinking water. The weight of ovaries and adrenals, and the plasma levels of corticosterone decreased in hypothyroid rats as compared with euthyroid rats throughout the estrous cycle.
