Identification and expression analysis of microRNAs in tibial growth plate of chicken through thiram toxicity.

Thiram is a widely known tibial dyschondroplasia (TD) inducer. TD, a common metabolic cartilage disease, presents in rapidly growing poultry birds. There are evidences that miRNAs are involved in diverse aspects of normal skeletal development, but very less is known about the role of miRNAs in TD. Therefore, this study aimed to determine which genes and pathways show differential expression between TD suffered chickens and normal chickens. We collected growth plates from ten-days-old TD chickens and control chickens and performed high-throughput RNA sequencing (RNA-Seq). Afterwards, target pre…

Residue of thiram in food, suppresses immune system stress signals and disturbs sphingolipid metabolism in chickens.

Thiram, a well-known sulfur containing organic compound is frequently and extensively used in agriculture because of high biological activity to control different pests. In certain cases, due to long persistence in the environment pesticides and other environmental contaminants induce undesirable toxic impacts to public health and environment. To ascertain the potential mechanisms of toxicity of thiram on different immune organs of broilers, a total of 100 one-day-old chicks were obtained and randomly divided into two groups including thiram group (50 mg/kg) and untreated control group. Thymus…

Nanofibrillar cellulose/Au@Ag nanoparticle nanocomposite as a SERS substrate for detection of paraquat and thiram in lettuce.

A nanocomposite based on nanofibrillar cellulose (NFC) coated with gold-silver (core-shell) nanoparticles (Au@Ag NPs) was developed as a novel surface-enhanced Raman spectroscopy (SERS) substrate. SERS performance of NFC/Au@Ag NP nanocomposite was tested by 4-mercaptobenzoic acid. The cellulose nanofibril network was a suitable platform that allowed Au@Ag NPs to be evenly distributed and stabilized over the substrate, providing more SERS hotspots for the measurement. Two pesticides, thiram and paraquat, were successfully detected either individually or as a mixture in lettuce by SERS coupled w…

Reasons for the decomposition of the fungicide thiram during preparation of fruit and vegetable samples and consequences for residue analysis.

The concentration of thiram in aqueous solution decreases by 50-75% within 20 min in the presence of cut pieces of apple, cucumber or celeriac with a section surface area of 160 cm2 each. The decomposition rate is predominantly influenced by the section surface area of the cut fruit and vegetable samples. Denaturing reaction conditions (exchange of the solvent water by methanol; boiling of sample material) will significantly slow down the decomposition rate. It was concluded that the thiram decomposition had been caused by enzymes on the section surface of the fruit and vegetable samples. For …

Thiram Induced Hematological Disorders, Gills Toxicity, Oxidative Stress, and Antioxidant Alterations in Common Carp (Cyprinus carpio).

Thiram is a dithiocarbamate fungicide that protects turf, fruits and ornamental plants. The purpose of this experiment was to ascertain the toxico-pathological effects of thiram induced toxicity in Cyprinus carpio. The fish were divided into four groups: Group A was kept as control, compared to groups B, C, and D exposed to 40, 80, and 120 μg/L thiram, respectively. The results indicated a significant (p < 0.05) reduction in body weight, while both the absolute and relative weights of gills increased significantly (p < 0.05) in fish exposed to a higher concentration of thiram. Hematological pa…

Glutathione-S-transferase A3 protein suppresses thiram-induced tibial dyschondroplasia by regulating prostaglandin-related genes expression.

Tibial dyschondroplasia (TD) is an intractable avian cartilage disease in which proximal growth plates of tibia lack blood vessels and conta … tic prostaglandin D synthases (HPGDS) expression were identified and could significantly respond to thiram-induced TD chicken. Interestingly, the expression of rate-limiting enzyme COX-2 and PGE2 were induced after the treatment of rGSTA3 protein. These findings demonstrated that the occurrence of TD is closely related to the inhibition of PGs. Moreover, …

Transcriptome Analysis of Thiram-Treated Zebrafish (Danio rerio) Embryos Reveals Disruption of Reproduction Signaling Pathways.

Thiram, a dithiocarbamate fungicide, is used for the treatment of various fungal infections affecting crops and ornamentals. However, thiram-associated toxicity has been reported in animals, including fish, and the underlying molecular mechanisms are unclear. Herein, we employed zebrafish (ZF) to gain further insights into thiram toxicity-associated molecular mechanisms. We studied developmental abnormalities and performed whole-transcriptome analysis of ZF embryos exposed to thiram for 96 h. Embryos exposed to 4.0 μg/L thiram exhibited several phenotypic abnormalities, including bradycardia, …

Integrated assessment of thiram toxicity in pangasius fish: Erythrocytic abnormalities, gill and intestinal defense mechanisms, and genotoxicity.

Pesticides play a crucial role in agriculture, public health management, and veterinary practice to control pests. However, the persistent use of pesticides poses a significant threat to aquatic ecosystems and non-target organisms, such as fish. The genotoxicity, oxidative stress, and erythrocytic toxicity were assessed in fish exposed to different sublethal concentrations (5, 10, and 15 µg/L) of thiram for 12 days. Blood and other tissues (gills and intestine) were obtained from each fish reared in each group at days 4, 8, and 12 of the experiment. Thin blood film/smear examination indicated …

Thiram-driven XBP1 and CHOP pathways orchestrate ER stress and avian tibial dyschondroplasia via metabolic inflammatory axis.

Thiram, one of the most widely used dithiocarbamate fungicides, has turned out to be a very potent cellular metabolic homeostasis disruptor through the induction of endoplasmic reticulum (ER) stress. The present study examines the mechanistic foundation of thiram-induced tibial dyschondroplasia in birds, emphasizing ER stress and inter-organ crosstalk. According to our findings, thiram caused impairment in the function of hepatocytes, as well as inducing an inflammatory cascade of signals in the tibial growth plate and liver tissues. This dual effect on tissue underlines the systemic disruptio…

Recovery of chicken growth plate by TanshinoneⅡA through wnt/β-catenin pathway in thiram-induced Tibial Dyschondroplasia.

Tibial Dyschondroplasia (TD), a metabolic disease of fast growing poultry birds that effects the growth of bone and cartilage, is characterized by anorexia, mental depression and lameness. Wnt/β-catenin pathway can mediate the occurrence of TD, and previous study showed the therapeutic effect of TanshinoneⅡA to TD Broilers. However there is no report about the effect of TanshinoneⅡA treating TD broiler chicken through wnt/β-catenin pathway. The objective of this study was to explore the potential mechanism of how Tanshinone II A treats TD. Hematoxylin and eosin staining was used to study histo

Quercetin alleviates thiram induced tibial dyschondroplasia in broiler chicken through modulating oxidative stress and cecal microbiota.

INTRODUCTION: Tibial dyschondroplasia (TD) is characterized by unmineralized cartilage plugs in the proximal tibial growth plate and is clinically associated with lameness and impaired growth performance in broilers. This study investigated the protective effects of quercetin, a natural flavonoid compound, against thiram-induced TD in broilers. METHODS: A total of 180 one-day-old broilers were randomly assigned to a control group, a TD group, and a quercetin (QUE) group. TD was induced in the TD and QUE groups by feeding a diet containing 100 mg/kg thiram from days 4 to 7. The QUE group addit…

The autophagy-mediated mechanism via TSC1/mTOR signaling pathway in thiram-induced tibial dyschondroplasia of broilers.

Thiram is a member of the dithiocarbamate family and is widely used in agriculture, especially in low-income countries. Its residues lead to various diseases, among which tibial dyschondroplasia (TD) in broiler chickens is the most common. Recent studies have also demonstrated that thiram residues may harm human health. Our previous study showed that the activity of the mTOR (mammalian target of rapamycin) signaling pathway has changed after thiram exposure. In the current study, we investigated the effect of autophagy via the mTOR signaling pathway after thiram exposure in vitro and in vivo. …

Ligustrazine recovers thiram-induced tibial dyschondroplasia in chickens: Involvement of new molecules modulating integrin beta 3.

Tetramethyl thiuram disulfide (thiram) is a dithiocarbamate, which is extensively used in agriculture as pesticide and fungicide for treating grains intended for seed purposes and also for storing food grains. One of the most evident and detrimental effect produced by thiram is tibial dyschondroplasia (TD) in many avian species, by feeding diets containing thiram, a growth plate cartilage disease. TD is characterized by the lack of blood vessels and impaired vascular invasion of the hypertrophic chondrocyte resulting in the massive cell death. This study investigated the effects of ligustrazin…

Effects of commonly used carbamates (carbaryl and thiram) on the regulatory, secretory and motor functions of bovine cervixes in vitro.

Previously studied classes of pesticides, including organochlorines, organophosphates and pyrethroids disturb the mechanism that causes bovine myometrial contractions. Hence, the aim of this study was to investigate the effects of carbaryl and thiram, which are representative carbamate pesticides commonly used in global agriculture, on the motor and secretory functions of bovine cervixes. Additionally, the impacts of these pesticides on intra- and intercellular signaling in vitro were estimated. In this study, cervical cells or strips were obtained from cows at days 18-20 of the estrous cycle …

Cu-triggered ratiometric fluorescence sensor based on dual-emission carbon dots for visual monitoring of thiram in food.

A copper and nitrogen co-doped carbon dots (Cu, N-CDs)-based fluorescent sensor is developed for thiram detection. In detail, with increasing thiram concentration the fluorescence intensity of Cu, N-CDs at 405 nm decreases , while that at 495 nm remains largely unchanged. This phenomenon results from the Cu-induced complex formation, which increases the aggregation of Cu, N-CDs and triggers fluorescence resonance energy transfer. Based on these findings, a good linear correlation is observed ranging from 0 to 100 µM (R2 = 0.99489) thiram under the optimized conditions, with a calculated detect…

Recombinant glutathione-S-transferase A3 protein regulates the angiogenesis-related genes of erythrocytes in thiram induced tibial lesions.

Tibial dyschondroplasia (TD) is a skeletal deformity disease in broilers that occurs when vascularization in the growth plate (GP) is below normal. Although, blood vessels have been reported to contribute significantly in bone formation. Therefore, in the current study, we have examined the mRNA expression of angiogenesis-related genes in erythrocytes of thiram induced TD chickens by qRT-PCR and performed histopathological analysis to determine regulatory effect of recombinant Glutathione-S-Transferase A3 (rGSTA3) protein in response to the destructive effect of thiram following the injection …

Thiram-induced ER stress promotes mitochondrial calcium signaling and NLRP3 inflammasome activation in a tissue specific manner.

Thiram, a broadly used dithiocarbamate fungicide, exaggerates endoplasmic reticulum (ER) stress and interferes with mitochondrial function, thus disrupting cellular homeostasis. Here, we intend to identify the molecular actions of thiram at the mitochondrial-associated ER membranes (MAMs) that lead to the induction of ER stress and mitochondrial calcium overload in both liver and bone tissues. Taken together, we show that thiram-induced remodelling of MAMs leads to huge ER stress and calcium dysregulation. Histological and immunohistochemical examinations revealed that thiram-induced hyperacti…
