Taking into account the literature reports on the involvement of prolactin (PRL) in the regulation of immunity against Toxoplasma gondii, we decided to check whether this parasite has the ability to bind the lactotrophic hormone. We examined T. gondii binding of sheep fluoresceine- and biotine-labeled prolactin isolated from pituitary (shPRL). In this work we announced for the first time that shPRL was bound to live tachyzoites of RH (type I) and ME49 (type II) strains. Furthermore, by use of competitive inhibition analysis, we confirmed that this binding was specific for both tested T. gondii…

Toxoplasma gondii has a very wide intermediate host range and is thought to be able to infect all warm blooded animals. The parasite causes a spectrum of different diseases and clinical symptoms within the intermediate hosts and following infection most animals develop adaptive humoral and cell-mediated immune responses. The development of protective immunity to T. gondii following natural infection in many host species has led researchers to look at vaccination as a strategy to control disease, parasite multiplication and establishment in animal hosts. A range of different veterinary vaccines…

The antifungal agent itraconazole is an effective drug against systemic mycoses inhibiting cytochrome P-450-mediated ergosterol synthesis, essential for fungal survival. In this work, we show the activity of this azole as a potential agent against Toxoplasma gondii, the causative agent of toxoplasmosis. Monolayers of LLC-MK2 epithelial cells infected with tachyzoites of RH strain were incubated with different concentrations of itraconazole for 24 and 48 h. The IC(50) values obtained were 114.0 and 53.6 nM for 24 and 48 h, respectively. Transmission electron microscopy (TEM) analysis of itracon…

Toxoplasmosis is caused by an intracellular protozoan, Toxoplasma gondii, which has a wide geographical distribution. The main infection routes are ingestion of cysts from raw or badly-cooked meat, ingestion of oocysts from substrates contaminated with the feces of infected felines and congenital transmission by tachyzoites. The congenital form results in a severe systemic disease, because if the mother is infected for the first time during gestation, she can present a temporary parasitemia that will infect the fetus. Many of the clinical symptoms are seen in congenitally-infected children, fr…

The phylum Apicomplexa contains parasites responsible for a variety of diseases including malaria, cryptosporidiosis, and toxoplasmosis. One of the common features of these parasites is that they contain a set of apical organelles whose sequential secretion is required for the invasion of host cells. Microneme proteins are the main adhesins involved in the attachment to the host cell surface by apicomplexans. The microneme protein MIC2, produced by Toxoplasma gondii, is conserved in apicomplexans and serves as a model to understand the first steps of invasion by the phylum. New data about the …

The indirect fluorescent antibody technique was used to study the relationship between endogenous uveitis and toxoplasma infection. The seropositive rates of toxoplasma antibody in healthy people, farmers and patients with endogenous uveitis were 10.7%, 15.2% and 30.8% respectively. Compared with the healthy people, the infection rate in uveitic patients was significantly higher (P less than 0.01). The odds ratio of toxoplasma infection in endogenous uveitic patients was 3.72. The results suggest that toxoplasma infection is an infectious cause of endogenous uveitis.

Sera of 532 patients suffering from different ophthalmological diseases were studied for the presence of toxoplasma antibodies. Complement fixation test was positive in 44.9% of the patients. As compared with the 26.8% positivity rate determined earlier in the normal population the seropositivity in eye-patients was significantly higher. The highest incidence, 50%, was found in the age group of 21--30 years. In diseases with a confirmed aetiological role of toxoplasmosis, the incidence of seropositivity was higher than in the total material. This suggests that Toxoplasma gondii may have had a …

Toxoplasma gondii is the causative agent of toxoplasmosis, a zoonotic disease that poses a threat to human health and a considerable loss to livestock farming. At present, clinical therapeutic drugs mainly target T. gondii tachyzoites and fail to eradicate bradyzoites. Developing a safe and effective vaccine against toxoplasmosis is urgent and important. Breast cancer has become a major public health problem and the therapeutic method needs to be further explored. Many similarities exist between the immune responses caused by T. gondii infection and the immunotherapy for cancers. T. gondii den…

The risk of Toxoplasma gondii infection in solid organ transplant recipients is well known but mainly after heart transplantation. We discuss a case of acute respiratory distress caused by Toxoplasma gondii on Day 32 after liver transplantation. The reported case, which is extremely rare, emphasizes how direct examination and Quantitative Polymerase Chain Reaction (QPCR) in broncholaveolar lavage help to diagnose the infection. Given Trimethoprim/Sulfamethoxazole toxicity, systematic prescription of Toxoplasma gondii chemoprophylaxis is not commonly approved in liver transplantation.

Endocytosis is critical for many functions in eukaryotic cells. Uptake of host cell cytosol, an indispensable endocytic process in malaria blood-stage parasites, has been known for a long time. However, it is only recently that the proteins involved in this process have started to emerge. Unexpectedly, some of these proteins revealed a critical role for endocytosis in artemisinin resistance. More recently, endocytosis was discovered in both intracellular and extracellular Toxoplasma gondii parasites. Here, we review these findings, compare the endolysosomal systems of Toxoplasma and Plasmodium…

The protozoan Toxoplasma gondii, described by Nicolle and Manceaux in 1908, is a ubiquitous and cosmopolitan parasite that infects a wide range of mammal and bird species with high prevalence. The biological success of T. gondii is associated with the formation of a specific relationship between the parasite and host cells leading to the establishment of a latent, chronic infection. During primary infection, acquired mostly by the oral route, the quickly multiplying tachyzoites disseminate through the body crossing several structural-functional barriers as blood-brain or blood-retina, then the…

Toxoplasma gondii is a nucleated cell obligate parasite, and can cause severe toxoplasmosis. The genetypes of Toxoplasma gondii isolates of different host infections have significant differences, and the pathogenicity and sensitivity to drugs of different genotypes of Toxoplasma gondii isolates are also significantly different. At present, the analysis of Toxoplasma gondii genotypes is performed by using PCR-RFLP, PCR of highly repetitive sequences, multiple PCR and nested PCR techniques, and the multiple loci of amplification are usually B1, SAG2, HSP70, GRA6 and other genetic markers. There …

In a period of 5 years, 21 children with ocular lesions caused by toxoplasma (chorioretinitis due to toxoplasma) were seen at "Pedro Kourí" Tropical Medicine Institute. Of the 21 children, 5 (23.8%) had lesions in both eyes and 16 (76.1%) in one eye (9 in the left eye and 7 in the right one). 9 children (42.8%) initially had active lesions that responded satisfactorily to medical treatment whereas 12 (57.1%) came to the hospital with healed lesions that did not require any treatment. They were all followed up as outpatients with favorable evolution in all cases.

Considered rare and occurring only in immunocompromised patients or patients with severe infection, Toxoplasma gondii chorioretinitis--as a unique presenting sign of toxoplasmosis appears to occur with increased frequency over the last few years in immunocompetent patients. The typical clinical picture (central chorioretinal lesion with recurrent vasculitic reactions) and positive IgG Toxoplasma gondii serology confirmed our diagnosis. Negative Toxoplasma IgG serology in the mothers of 2 patients excluded the possibility of congenital toxoplasmosis. Clinicians should be aware of the possibilit…
