Mortality rates and population density of tsetse flies correlated with satellite imagery.

Tsetse flies are a major constraint on animal production in about 10 million km2 of Africa through their transmission of animal trypanosomiasis. Up to 25 million people are at risk from human trypanosomiasis, or sleeping sickness. Tsetse research has been concentrated on the factors that control the distribution and abundance of these vectors and the means by which their numbers can be reduced. Eradication successes in some countries are insignificant compared with the continental scale of the problem and the long-term reduction in the area infested by tsetse has been negligible. We report her…

A new method for identifying blood meals of human origin in tsetse flies.

A new sensitive technique using the electrophoresis of superoxide dismutase to distinguish between tsetse blood meals of human and non-human origin is described. In Côte d'Ivoire, 602 blood meals were collected; 170 were from man (28.3%), 377 from animals (62.6%) and 55 were unidentified (9.1%) because no pattern was observed. When calculating the index of epidemiological risk, it is strongly correlated with the incidence of sleeping sickness cases.

Negative Density-dependent Dispersal in Tsetse Flies: A Risk for Control Campaigns?

Tsetse flies are vectors of parasites that cause diseases responsible for significant economic losses and health issues in sub-Saharan Africa, including sleeping sickness in humans and nagana in domestic animals. Efficient vector-control campaigns require good knowledge of the demographic parameters of the targeted populations. In the last decade, population genetics emerged as a convenient way to measure population densities and dispersal in tsetse flies. Here, by revealing a strong negative density-dependent dispersal in two dimensions, we suggest that control campaigns might unleash dispers…

Optimal strategies for controlling riverine tsetse flies using targets: a modelling study.

BACKGROUND: Tsetse flies occur in much of sub-Saharan Africa where they transmit the trypanosomes that cause the diseases of sleeping sickness in humans and nagana in livestock. One of the most economical and effective methods of tsetse control is the use of insecticide-treated screens, called targets, that simulate hosts. Targets have been ~1 m2, but recently it was shown that those tsetse that occupy riverine situations, and which are the main vectors of sleeping sickness, respond well to targets only ~0.06 m2. The cheapness of these tiny targets suggests the need to reconsider what intensit…

Standardizing visual control devices for tsetse flies: East African species Glossina swynnertoni.

BACKGROUND: Here we set out to standardize long-lasting, visually-attractive devices for Glossina swynnertoni, a vector of both human and animal trypanosomiasis in open savannah in Tanzania and Kenya, and in neighbouring conservation areas used by pastoralists. The goal was to determine the most practical device/material that would induce the strongest landing response in G. swynnertoni for use in area-wide population suppression of this fly with insecticide-impregnated devices. METHODS AND FINDINGS: Trials were conducted in wet and dry seasons in the Serengeti and Maasai Mara to measure the

Patterns of Genome-Wide Variation in Glossina fuscipes fuscipes Tsetse Flies from Uganda.

The tsetse fly Glossina fuscipes fuscipes (Gff) is the insect vector of the two forms of Human African Trypanosomiasis (HAT) that exist in Uganda. Understanding Gff population dynamics, and the underlying genetics of epidemiologically relevant phenotypes is key to reducing disease transmission. Using ddRAD sequence technology, complemented with whole-genome sequencing, we developed a panel of ∼73,000 single-nucleotide polymorphisms (SNPs) distributed across the Gff genome that can be used for population genomics and to perform genome-wide-association studies. We used these markers to estimate

Does isometamidium chloride treatment protect tsetse flies from trypanosome infections during SIT campaigns?

African animal trypanosomosis is a major pathological constraint to cattle breeding across 10 million km2 of sub-Saharan West African countries infested by tsetse flies, their cyclic vectors. The release of sterile males (sterile insect technique [SIT]) is a potentially important control technique aimed at eliminating the vectors. Prior to release, tsetse are generally treated with isometamidium chloride, a trypanocide, to prevent them from transmitting parasites. The present study investigated the preventive action of isometamidium chloride (0.5 mg/L) on the subsequent susceptibility of tsets…

Recombinant DNA probes reveal simultaneous infection of tsetse flies with different trypanosome species.

The utility of recombinant DNA probes in the detection of natural trypanosome infection of tsetse flies has been assessed in Lambwe Valley, near the shores of Lake Victoria, Kenya. The tsetse flies were surveyed during two different seasons in 1988. Three different probes used each contained highly repetitive DNA sequences specific for a species or subspecies of trypanosomes of the Nanomonas subgenus. A fourth probe contained repetitive sequences common to trypanosome species of the Trypanozoon subgenus. Mixed mature or immature infections were detected in a variety of combinations in differen…

Conjugal DNA Transfer in Sodalis glossinidius, a Maternally Inherited Symbiont of Tsetse Flies.

Stable associations between insects and bacterial species are widespread in nature. This is the case for many economically important insects, such as tsetse flies. Tsetse flies are the vectors of Trypanosoma brucei, the etiological agent of African trypanosomiasis-a zoonotic disease that incurs a high socioeconomic cost in regions of endemicity. Populations of tsetse flies are often infected with the bacterium Sodalis glossinidius Following infection, S. glossinidius establishes a chronic, stable association characterized by vertical (maternal) and horizontal (paternal) modes of transmission. …

Tsetse flies: their biology and control using area-wide integrated pest management approaches.

Tsetse flies are the cyclical vectors of trypanosomes, the causative agents of 'sleeping sickness' or human African trypanosomosis (HAT) in humans and 'nagana' or African animal trypanosomosis (AAT) in livestock in Sub-saharan Africa. Many consider HAT as one of the major neglected tropical diseases and AAT as the single greatest health constraint to increased livestock production. This review provides some background information on the taxonomy of tsetse flies, their unique way of reproduction (adenotrophic viviparity) making the adult stage the only one easily accessible for control, and how…

Genetic Diversity of African Trypanosomes in Tsetse Flies and Cattle From the Kafue Ecosystem.

We clarified the genetic diversity of Trypanosoma spp. within the Kafue ecosystem, using PCR targeting the internal transcribed spacer 1 and the cathepsin L-like cysteine protease (CatL) sequences. The overall prevalence of Trypanosoma spp. in cattle and tsetse flies was 12.65 and 26.85%, respectively. Cattle positive for Trypanosoma vivax had a significantly lower packed cell volume, suggesting that T. vivax is the dominant Trypanosoma spp. causing anemia in this area. Among the 12 operational taxonomic units (OTUs) of T. vivax CatL sequences detected, one was from a known T. vivax lineage, t…

Molecular identification of different trypanosome species and subspecies in tsetse flies of northern Nigeria.

BACKGROUND: Animal African Trypanosomiasis (AAT) is caused by several species of trypanosomes including Trypanosoma congolense, T. vivax, T. godfreyi, T. simiae and T. brucei. Two of the subspecies of T. brucei also cause Human African Trypanosomiasis. Although some of them can be mechanically transmitted by biting flies; these trypanosomes are all transmitted by tsetse flies which are the cyclical vectors of Trypanosoma congolense, T. godfreyi, T. simiae and T. brucei. We present here the first report assessing the prevalence of trypanosomes in tsetse flies in Nigeria using molecular tools. …

Developing a continental atlas of the distribution and trypanosomal infection of tsetse flies (Glossina species).

BACKGROUND: Tsetse flies (Genus: Glossina) are the sole cyclical vectors of African trypanosomoses. Despite their economic and public health impacts in sub-Saharan Africa, it has been decades since the latest distribution maps at the continental level were produced. The Food and Agriculture Organization of the United Nations is trying to address this shortcoming through the Atlas of tsetse and African animal trypanosomosis. METHODS: For the tsetse component of the Atlas, a geospatial database is being assembled which comprises information on the distribution and trypanosomal infection of Glos…

Is there safety in numbers? The effect of cattle herding on biting risk from tsetse flies.

In sub-Saharan Africa, tsetse (Glossina spp.) transmit species of Trypanosoma which threaten 45-50 million cattle with trypanosomiasis. These livestock are subject to various herding practices which may affect biting rates on individual cattle and hence the probability of infection. In Zimbabwe, studies were made of the effect of herd size and composition on individual biting rates by capturing tsetse as they approached and departed from groups of one to 12 cattle. Flies were captured using a ring of electrocuting nets and bloodmeals were analysed using DNA markers to identify which individual

[Incidence of trypanosoma in the population of tsetse flies at the game ranch of Nazinga (Burkina Faso)].

The frequency of trypanosome infection in Glossina morsitans submorsitans and G. tachinoides, at the game ranch of Nazinga (Burkina Faso), was examined. Considering the absence of human activities and domestic animals, the study area can be considered as "primitive" and the infection rates as "natural". These results will contribute to clarifying some points concerning the epidemiology of the transmission of the parasite, in particular in those areas where the setting up of a natural game reserve is planned.
