Abnormal concentrations of glycosaminoglycans (GAG) have been reported for various types of tumors, suggesting that they may play a role in neoplasia. This study was carried out in order to determine whether osteogenesis imperfecta affected individuals showed a different pattern of glycosaminoglycan excretion in urine than do normals. A heteropolysaccharide formed by hexoses, hexosamines, fucose, sialic acid, uronic acids and sulphates was isolated from submandibular glands of rabbits during postnatal development. The presence of sulphates in the submandibular glands of growing animals and their absence in the submandibular glands of adult animals is rather difficult to explain. This biochemical investigation and preliminary results obtained by light and electron microscopy on the sublingual gland seem to suggest a substitutive role of the submandibular gland during the neonatal period.

Acid GAG were isolated from hepatic tissue from 3 patients with Hurler syndrome and 3 normal controls. Gross elevations in the uronic acid and hexosamine contents were found in Hurler livers compared with the normal ones. The total GAG concentration was significantly increased (about 25fold) in Hurler patients.

In order to determine if either the proteoglycans or collagen in the cartilagenous epiphyses of a Miniature Poodle with spondyloepiphyseal dysplasia were abnormal, the cartilage was dissociatively extracted in 4 M guanidine HCl in the presence of protease inhibitors and subjected to isopycnic cesium chloride dissociative density gradient ultracentrifugation. Dissociative extraction solubilized 97% of the uronic acid and 88% of the protein. The urinary excretion of uronic acid and the electrophoretic composition of urinary glucosaminoglycans were studied in 10 members of a family, of which 3 had progressive familial myoclonus epilepsy. Histochemical studies of mucins were carried out on 17 autopsy specimens of cholangiocarcinomas associated with clonorchiasis. The urinary excretion of uronic acid and the hydroxyproline of a urinary fraction of high molecular weight collagen peptides (% of total Hyp) indicate disease activity in generalized scleroderma. A significant decrease in the content of Hyp and Hyl in the skin of sclerodermal patients is not related to activity, but rather to the duration of the disease. Parallel analyses of skin and urine of the same patients are reported.
