Endoplasmic reticulum glucosidases and protein quality control factors cooperate to establish biotrophy in Ustilago maydis.

Secreted fungal effectors mediate plant-fungus pathogenic interactions. These proteins are typically N-glycosylated, a common posttranslational modification affecting their location and function. N-glycosylation consists of the addition, and subsequent maturation, of an oligosaccharide core in the endoplasmic reticulum (ER) and Golgi apparatus. In this article, we show that two enzymes catalyzing specific stages of this pathway in maize smut (Ustilago maydis), glucosidase I (Gls1) and glucosidase II β-subunit (Gas2), are essential for its pathogenic interaction with maize (Zea mays). Gls1 is r…

Cell death in Ustilago maydis: comparison with other fungi and the effect of metformin and curcumin on its chronological lifespan.

Ustilago maydis is a Basidiomycota fungus, in which very little is known about its mechanisms of cell survival and death. To date, only the role of metacaspase1, acetate and hydrogen peroxide as inducers of cell death has been investigated. In the present work, we analyzed the lifespan of U. maydis compared with other species like Sporisorium reilianum, Saccharomyces cerevisiae and Yarrowia lipolytica, and we observed that U. maydis has a minor lifespan. We probe the addition of low concentrations metformin and curcumin to the culture media, and we observed that both prolonged the lifespan of …

Analysis of ferrichrome biosynthesis in the phytopathogenic fungus Ustilago maydis: cloning of an ornithine-N5-oxygenase gene.

By using a non-enterobactin-producing enb-7 mutant of Salmonella typhimurium LT2 as a biological indicator, a novel screening method was developed for identifying mutants of Ustilago maydis defective in the biosynthesis of the siderophores ferrichrome and ferrichrome A. Two classes of siderophore mutations, both recessive, were isolated after mutagenesis of haploid cells of the corn smut fungus. Class I mutants no longer produced ferrichrome while retaining the ability to produce ferrichrome A; class II mutants were defective in the production of both ferrichrome and ferrichrome A. Genetic and…

TurboID-Based Proximity Labeling for Exploiting Effector Interacting Proteins in the Fungal Maize Pathogen Ustilago maydis.

TurboID-based proximity labeling (PL) is an advanced method for identifying proteins in the immediate molecular environment of a protein of … ity. Elucidating these proteins is meaningful for crop improvement and design. The corn smut fungus Ustilago maydis causes tumor formation on all aerial parts of maize. The U. maydis-maize pathosystem is a model system for studying biotrophic fungi and their interactions with plants. To identify U. maydis effector targets in maize, a TurboID-based PL appro …

Functional dissection of Gα subunits in Ustilago esculenta: Uegpa3 governs cAMP-PKA-dependent dimorphism and virulence to Zizania latifolia.

Ustilago esculenta is a dimorphic endophytic fungus that specifically infects Zizania latifolia, inducing stem swelling to form the economically important aquatic vegetable "Jiaobai". The pathogenicity of U. esculenta is closely associated with its dimorphic switch. This study systematically characterized the four G protein α subunit genes (Uegpa1-Uegpa4) in U. esculenta. Functional analysis revealed that Uegpa3 plays an indispensable role in pathogenicity by regulating dimorphic transition via the cAMP-PKA signaling pathway. Deletion of Uegpa3 resulted in defective conjugation tube formation,…

A Genomic DNA-Based Real-Time PCR Assay for Quantification of Ustilago maydis Biomass in Plant Samples.

Ustilago maydis is a basidiomycete fungus that causes common smut in maize, serving as a model system for studying smut fungi and biotrophic pathogens. Reliable quantification of fungal biomass in planta facilitates tracking fungal proliferation and disease development, evaluating fungal mutant virulence, and assessing host resistance. Traditional methods such as symptom scoring or microscopy after staining are semiquantitative and prone to subjectivity. In this chapter, a DNA-based quantitative real-time PCR (qPCR) assay for sensitive and reproducible measurement of U. maydis biomass in infec…

Prebiotic and Chemopreventive Potential of Digested Cooked Huitlacoche (Ustilago maydis) in Human HT-29 Colon Cancer Cells.

Huitlacoche (Ustilago maydis) is a popular and widely consumed edible fungus that infects corn, inducing gall formation rich in phenolic compounds. During consumption, huitlacoche-based phenolic compounds are affected by gastrointestinal conditions, thereby altering their bioaccessibility and, in turn, their prebiotic and chemopreventive potential. This study investigated the in vitro chemopreventive effects of cooked huitlacoche on human colon cancer cells (HT-29) and its prebiotic potential. Cooked huitlacoche underwent in vitro gastrointestinal digestion, followed by colonic fermentation, t…

Characterization of the Ustilago maydis sid2 gene, encoding a multidomain peptide synthetase in the ferrichrome biosynthetic gene cluster.

Ustilago maydis, the causal agent of corn smut disease, acquires and transports ferric ion by producing the extracellular, cyclic peptide, hydroxamate siderophores ferrichrome and ferrichrome A. Ferrichrome biosynthesis likely proceeds by hydroxylation and acetylation of L-ornithine, and later steps likely involve covalently bound thioester intermediates on a multimodular, nonribosomal peptide synthetase. sid1 encodes L-ornithine N(5)-oxygenase, which catalyzes hydroxylation of L-ornithine, the first committed step of ferrichrome and ferrichrome A biosynthesis in U. maydis. In this report we c…

A low-cost "plant-scanner" platform for automated detection of Ustilago maydis infection in maize using deep learning.

Ustilago maydis is a biotrophic fungus that causes smut disease in maize, leading to tumor formation on aerial parts of the plant. While U. maydis has been a model for plant-fungal interaction studies, no tool has existed to automatically quantify infection symptoms under laboratory conditions for deep learning analysis. To address this, we developed a rotating camera system that captures videos of plants under customized lighting and shutter settings. These videos were used to train machine learning models to distinguish between healthy and infected plants. Two detection approaches have been …

Wheat Germ Agglutinin-Alexa Fuor488/Propidium Iodine Costaining and Fluorescence Microscopic Imaging of Ustilago maydis Infected Maize Anthers.

Ustilago maydis infects all aerial maize tissues, forming tumors and altering host development. However, most published methods focus on seedling leaf infections in order to understand the interaction between maize and U. maydis. Cytological analysis, particularly fluorescence microscopy, is vital not only for examining fungal infection progression, but also for understanding the host's response. Thus, a detailed protocol is presented here for dissecting U. maydis-infected maize anthers, followed by costaining with wheat germ agglutinin-Alexa Fluor 488 (WGA-AF488) and propidium iodide (PI). WG…

Unusual presentation of fungal infective endocarditis as the first manifestation of HIV in a child with a structurally normal heart.

We report a 9-year-old boy who presented with mitral valve perforation secondary to Ustilago species fungal infective endocarditis. Subsequent evaluation revealed previously undiagnosed HIV infection. Fungal endocarditis due to Ustilago species is exceedingly rare, and presentation as the initial manifestation of HIV has not been previously described in the pediatric population. The atypical presentation posed significant diagnostic challenges, emphasizing the need to consider underlying immunodeficiency, including HIV infection, in children presenting with unusual or opportunistic infections.…

FLP/FRT-Mediated Recombination for Resistance Marker Recycling.

The availability of selection markers for metabolic engineering of Ustilago species is limited. The inducible flippase (FLP) recombinase system allows for the recycling of a selection marker after a successful recombination event. Following a traditional marker-based homologous recombination of DNA for deletion or insertion of a gene using special constructs with two flanking Flippase-Recognition Targets (FRT) at both sides of the selection marker, FLP-mediated recombination allows for marker removal after transformation. This enables its recycling for a further transformation approach. In Ust…

Synchronous Induction and Microscopic Analysis of Hyphal Growth Using Laboratory Strain AB33.

In nature, hyphal growth of Ustilago maydis is strictly coupled to plant infection and initiated after mating of two genetically compatible yeast cells on the plant surface. Compatibility is defined by mating type loci: the biallelic a locus mediates recognition and fusion through a pheromone/receptor system, whereas the multiallelic b locus, encoding transcription factor subunits bEast (bE) and bWest (bW), establishes and maintains the dikaryotic hypha controlled via the assembly of compatible subunits into active heterodimers after plasmogamy. The laboratory strain AB33 enables a controlled …
