Identification and Functional Analysis of Novel Nonstructural Proteins of Human Bocavirus 1.

UNLABELLED: Human bocavirus 1 (HBoV1) is a single-stranded DNA parvovirus that causes lower respiratory tract infections in young children worldwide. In this study, we identified novel splice acceptor and donor sites, namely, A1' and D1', in the large nonstructural protein (NS1)-encoding region of the HBoV1 precursor mRNA. The novel small NS proteins (NS2, NS3, and NS4) were confirmed to be expressed following transfection of an HBoV1 infectious proviral plasmid and viral infection of polarized human airway epithelium cultured at an air-liquid interface (HAE-ALI). We constructed mutant pIHBoV1

Extracellular vesicles and their miRNA contents counterbalance the pro-inflammatory effect of air pollution during physiological pregnancy: A focus on Syncytin-1 positive vesicles.

The impact of exposure to respirable particulate matter (PM) during pregnancy is a growing concern, as several studies have associated increased risks of adverse pregnancy and birth outcomes, and impaired intrauterine growth with air pollution. The molecular mechanisms responsible for such effects are still under debate. Extracellular vesicles (EVs), which travel in body fluids and transfer microRNAs (miRNAs) between tissues (e.g., pulmonary environment and placenta), might play an important role in PM-induced risk. We sought to determine whether the levels of PM with aerodynamic diameters of

Association of Long-term exposure to air pollution and residential greenness with lipid profile: Mediating role of inflammation.

Lipidemic effect of air pollutants are still inconsistent and their joint effects are neglected. Meanwhile, identified inflammation pathways in animal have not been applied in epidemiological studies, and beneficial effect of residential greenness remained unclear. Therefore, we used data from typically air-polluted Chinese cities to answer these questions. Particulate matter (PM) with a diameter of ≤ 1 µm (PM1), PM with a diameter of ≤ 2.5 µm (PM2.5), PM with a diameter of ≤ 10 µm (PM10), sulphur dioxide (SO2), nitrogen dioxide (NO2), and ozone (O3) were predicted by space-time extremely rand

Association between gaseous air pollutants and biomarkers of systemic inflammation: A systematic review and meta-analysis.

BACKGROUND: Studies have linked gaseous air pollutants to multiple health effects via inflammatory pathways. Several major inflammatory biomarkers, including C-reactive protein (CRP), fibrinogen, interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) have also been considered as predictors of cardiovascular disease. However, there has been no meta-analysis to evaluate the associations between gaseous air pollutants and these typical biomarkers of inflammation to date. OBJECTIVES: To evaluate the overall associations between short-term and long-term exposures to ambient ozone (O3), nitrogen

Competing discrete interfacial effects are critical for amyloidogenesis.

Amyloid accumulation is associated with pathological conditions, including type II diabetes and Alzheimer's disease. Lipids influence amyloidogenesis and are themselves targets for amyloid-mediated cell membrane disruption. Amyloid precursors are surface-active, accumulating at hydrophobic-hydrophilic interfaces (e.g., air-water), where their biophysical and kinetic behaviors differ from those in the bulk solution with significant and underappreciated consequences. Biophysical modeling predicted the probability and rate of beta-sheet amyloid dimer formation to be higher and faster at the air-w

Effect of the polar headgroup of phospholipids on their interaction with actin.

It is generally admitted that actin filaments are anchored to a membrane by membranar actin-binding-proteins. However, we found that actin may also interact directly with membrane phospholipids. The actin-phospholipid complex has been investigated at the air-water interface using a film balance technique. In order to probe the effect of the phospholipid headgroup on the actin-phospholipid interaction, we focus mainly on phospholipids that have the same acyl chain length but different headgroups. For all the phospholipids, the apparent area per molecule (the total surface divided by the number

Lung growth and maturation in experimental oligohydramnios in the rat.

On day 16 of gestation the amniotic sacs of either the right or left uterine horns of timed-pregnant Sprague-Dawley rats were punctured with a 20-gauge needle. The fetuses of the opposite horn served as controls. On day 21 of gestation (1 day prior to natural delivery), the fetuses were delivered by Cesarean section. Amniotic sac puncture resulted in a significant loss of amniotic fluid. Fetal growth retardation occurred. Lung growth was specifically retarded, as indicated by significantly reduced lung weight to body weight ratios and lung volume to body weight ratios following amniotic sac pu

Establishment of Repeated In Vitro Exposure System for Evaluating Pulmonary Toxicity of Representative Criteria Air Pollutants Using Advanced Bronchial Mucosa Models.

There is mounting evidence that shows the association between chronic exposure to air pollutants (particulate matter and gaseous) and onset of various respiratory impairments. However, the corresponding toxicological mechanisms of mixed exposure are poorly understood. Therefore, in this study, we aimed to establish a repeated exposure setting for evaluating the pulmonary toxicological effects of diesel exhaust particles (DEP), nitrogen dioxide (NO2), and sulfur dioxide (SO2) as representative criterial air pollutants. Single, combined (DEP with NO2 and SO2), and repeated exposures were perform

Chronic exposure to aerosolized PM2.5 in rats alters plasma protein levels and ex vivo coronary artery response to sphingosine-1-phosphate.

Growing evidence shows that air pollution exposure plays a critical role in the development and progression of cardiovascular disease. Pollutants such as airborne particles smaller than 2.5 μm (PM2.5) are linked to systemic inflammation, oxidative stress and vascular dysfunction, thereby increasing the risk of hypertension, atherosclerosis, stroke and myocardial infarction. Understanding the pathology and the disease progression is essential for developing targeted interventions to mitigate the global cardiovascular burden associated with polluted air. We aimed to study how different PM2.5 exp

Intravenous Administration of Liposomes Encapsulating Oleanolic Acid Ameliorates Lung Injury Caused by Air Pollutants.

Health problems caused by air pollutants, such as fine particulate matter, are becoming more prevalent worldwide, and there is an urgent need to develop prevention and treatment strategies to combat this increase. A main mechanism by which air pollutants cause health problems is by entering the respiratory system and increasing oxidative stress. Oleanolic acid (OA) is a natural pentacyclic triterpenoid compound found in various plants that exhibits many physiological effects, including antioxidant, anticancer, and anti-inflammatory effects. However, the effect of OA on lung injury caused by ai

The influence of ripening of semi-hard goat cheese in oil on its physicochemical composition and sensory properties.

The aim of this paper was to determine the influence of ripening of semi-hard goat cheese in oil (mixture of Mljet's extra-virgin olive oil and refined sunflower oil; 50:50) on its physicochemical composition and sensory properties, as well as to determine the optimal time of immersion of the cheese in oil. Five batches of cheeses were produced, and the cheeses of the same batch were randomly divided into 3 groups according to the ripening method: (1) ripening in air (control group, treatment 1), (2) ripening in oil after 10 d of ripening in air (treatment 2), (3) ripening in oil after 20 d of

Lipopolysaccharide in cells infected by Chlamydia trachomatis.

In view of the controversy concerning the expression of chlamydial lipopolysaccharide (LPS) in the eukaryotic host cell, the presence of this molecule was examined in three cell types which were experimentally infected with Chlamydia trachomatis serotype E. LPS was detected in the McCoy cell line, human endometrial epithelium and human amniotic epithelium with two monoclonal antibodies. The appearance and distribution of LPS at the host cell surface during the chlamydial developmental cycle and its transmission to neighbouring cells were examined by immunofluorescence microscopy after air dryi

Minimizing aggregates when nebulizing antibody Fab fragments: high thermostability and high concentration are key.

The therapeutic potential of inhaled biologics is limited by protein instability during aerosolization, which can cause aggregation and increase immunogenicity risks. This study aims to identify key molecular and formulation parameters that minimize the aggregation of antibody Fab fragments during vibrating mesh nebulization. A set of 14 engineered Fabs with a broad range of melting temperatures (Tm 60-90°C) were nebulized using two different commercial vibrating mesh devices. We systematically assessed the impact of Fab thermostability, protein concentration (10-80 mg/mL), formulation excipie

Impact of keratin subfractions on the ultrafine particles filtration of sustainable electrospun human hair keratin/PVA nanofibers.

Ultrafine particulate matter, especially particles below 100 nm, poses severe health risks due to their deep penetration into pulmonary regions and systemic circulation, emphasizing the need for sustainable and effective air filtration technologies. Conventional air filtration systems often face limitations in capturing these particles efficiently while maintaining low airflow resistance. A suggested solution lies in the use of bimodal nanofiber architectures, which combine coarse and fine fiber populations to enhance particle capture while preserving breathability. In this study, we proposed

The association between maternal exposure to outdoor air pollutants, inflammatory response, and birth weight in healthy women.

Increased maternal inflammatory response has been noted in women with pregnancies complicated by preterm birth and small-for-gestational age infants. However, the association between gestational exposure to air pollutants, maternal inflammatory response, and fetal growth remains unclear. In this study, we aimed to investigate the association between exposure to air pollutants during pregnancy and the concentration of inflammatory indicators in maternal and fetal circulations, as well as fetal growth. We recruited 108 healthy pregnant women living in northern (n = 55) and southern (n = 53) area
